Pileus was flat, round, and reddish brown. The diameters of the pilei and stipe length of the fruiting bodies were 67.1 mm and 16.9 mm, respectively. The scales were white or slightly brown and distributed evenly. The gill density was sparse and showed a rippled texture. The stipe was cream in color and the fluff was medium. ‘Sanjanghyang’ had a short cultivation period and fruiting bodies occurred sporadically. Temperature for fruiting body formation was a medium, between 15 to 19°C. ‘Sanjanghyang’ was different from ‘Sanbaekhyang’ with regard to its pileus diameter (67.1 mm) and autumn and spring fruiting body production period. ‘Sanbaekhyang’ had pileus diameter of 74.7 mm, and fruiting body formation occurred in spring and autumn. Belamcanda chinensis) from China was previously identified as Puccinia iridis. Accordingly, the identity of the rust fungus on the same host species in Korea needs to be reexamned. Morphological characteristics of the Korean materials matched with P. iridis. Molecular phylogenetic analyses based on internal transcribed spacer and large subunit rDNA sequences of two representative materials confirmed the identification by high sequence similarities of 100% and 99% with the reference sequences available in NCBI GenBank database. Phylogenetic tree inferred from neighbor-joining method proved them to be clustered in P. iridis group. Thus, the rust fungus on I. domestica in Korea was determined as P. iridis. Hong, Yong-ho Lee","doi":"10.4489/KJM.20190031","DOIUrl":"https://doi.org/10.4489/KJM.20190031","url":null,"abstract":"Between August and September 2017, zonate leaf spots with severe defoliation were observed on Parthenocissus tricuspidata in Gimcheon, Korea. The initial symptoms included water-soaked, gray to grayish brown, circular or irregular-shaped leaf spots. As the disease progressed, the spots enlarged, coalesced, and became cream-colored with characteristic target-shaped rings, leading to leaf blight and premature defoliation. Based on cultural and morphological characteristics, the fungus associated with the symptoms was identified as Grovesinia moricola. The identity of this fungus was confirmed by analyzing sequences of the internal transcribed spacer region of ribosomal DNA. The pathogenicity of the isolate was demonstrated by artificial inoculation. The strain KMCC04674, used in this study was identified T. fuciformis by internal transcribed spacer (ITS). To define the optimum conditions for the mass production of T. fuciformis, we investigated the effects of different culture conditions and various nutrient sources on the fungal growth. The optimum initial pH and temperature for the fungal growth were 5.0 and 25°C, respectively. These fungal isolates were analyzed based on their morphological characteristics and the molecular phylogenetic analysis of the internal transcribed spacer (ITS) rDNA sequences. On the basis of their morphology and phylogeny, KNU17-1 and KNU17-199 isolates were identified as Albifimbria verrucaria and Deconica coprophila, respectively. To the best of our knowledge, A. verrucaria and D. coprophila have not yet been reported in Korea. Fourteen strains were used to develop the best varieties of L. edodes, and hybridization was carried out by monohybrid cross. The number of hybridized strains was 1,638 among 3,100 combinations. They were cultivated on sawdust medium, and fruiting bodies were formed in 364 strains. Among them, 65 strains were selected as superior candidate strains based on the shape and size of the fruiting bodies. Forty strains formed fruiting bodies without lamellae structure. The shape of the stipe was cylindrical (255 strains), thick to lower part (15 strains), and thick to upper part (94 strains). By the combinations of 2462 n1-10 and 3420 n1-10, 2462 n1, 2462 n2, 2462 n10, and 3420 n3 were selected as excellent monokaryotic strains.
