Due to the unique flooded conditions of rice cultivation, paddy soils exhibit distinct microbial compositions and carbon storage mechanisms compared to upland soils. However, indiscriminate fertilization, without a thorough understanding of the underlying soil carbon pool storage and turnover mechanisms, not only yields suboptimal results but also poses environmental risks. Currently, a comprehensive literature review exploring the impacts of fertilization on crop growth and soil fertility accumulation mechanisms within rice cultivation systems remains lacking. This review summarizes the nutrient characteristics and turnover mechanisms in paddy soils, revealing slower organic matter turnover and stable soil organic carbon due to the unique paddy soil anaerobic environment. Microbial mineralization and organic matter accumulation vary with the planting cycle. We discuss the impacts of fertilization on soil fertility and microbial communities, highlighting the superior and more environmentally friendly effects of organic fertilizers. Importantly, a saturation threshold exists for soil carbon storage; exceeding this limit renders fertilization ineffective. Appropriate fertilizer application positively impacts microbial communities by modifying soil pH and physicochemical properties. Furthermore, this paper elaborates on the mechanisms of microbial-mediated carbon sequestration and its influencing factors. Future research leveraging a clear understanding of paddy soil nutrient mechanisms should integrate synthetic biology, materials engineering, and data-driven intelligent decision-making systems. This approach promises to be an effective pathway toward achieving intelligent, green, and high-yielding rice cultivation.</p>","PeriodicalId":519973,"journal":{"name":"Crop health","volume":"4 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2026-06-09","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC13249996/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"148206884","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Entomopathogenic fungi (EPF), traditionally used as biocontrol agents against insect pests, also exhibit a remarkable ability to live within plants as endophytes, significantly boosting plant resilience to abiotic stresses. This review summarises the mechanisms by which EPF endophytes colonise plants and confer robust tolerance to drought, salinity, and heavy metal toxicity. These beneficial fungi orchestrate host physiological adaptations by activating antioxidant systems, up-regulating stress-responsive genes, modulating phytohormone signalling, and enhancing nutrient assimilation. Through improved photosynthetic efficiency, osmotic adjustment, nutrient uptake, and ion homeostasis, EPF endophytes substantially increase plant biomass, yield stability, and productivity under severe stress. Harnessing these fungi as dual-purpose bioagents offers a sustainable alternative to chemical pesticides, bridging ecological pest and disease management and climate-resilient agriculture. Previously, a link between changes in ubiquitination and the progression of diseases caused by Lso was established. Among the Lso haplotypes, LsoA and LsoB infect solanaceous crops. LsoA induces chlorosis and stunting in tomato, while LsoB infection leads to more severe symptoms, including plant death. To better characterize the molecular basis of the differences in the diseases caused by these haplotypes, we cataloged the ubiquitinome of tomato leaves four weeks after LsoA and LsoB-infection and following infestation with Lso-free psyllids. Our analysis revealed haplotype-specific changes in the plant ubiquitinome. Gene Ontology (GO) terms related to glucose metabolism and proteasome-mediated ubiquitin-dependent protein catabolism were enriched among the differentially ubiquitinated proteins between LsoA-infected and Lso-free infested plants. Additionally, a KEGG pathway analysis determined an overrepresentation of proteins involved in carbon metabolism and fixation via the Calvin cycle. In contrast, the comparison of LsoB-infected and Lso-free infested plants revealed a broader range of enriched GO terms and KEGG pathways, including those associated with carbohydrate and energy metabolism, proteasome activity, and oxidative stress. These results contribute to our understanding of the development of Liberibacter-related diseases and reveal the importance of ubiquitin-related pathways in Lso infection. They often function as important defensive secondary metabolites. Currently, research aimed at elucidating the biosynthesis of maize terpenes is limited. Thus, a rapid and efficient platform for identifying genes involved in the synthesis/regulation of maize volatile terpenes is highly desirable. Through optimization of various conditions, including light, substrate accumulation, nutrient and ionic composition of the culture medium, culture vessel size and conditions, organic solvent extraction, and promoter selection, we established a maize protoplast transfection system for analyzing the biochemical functions of terpene synthases (TPSs) and the functions of regulatory factors, such as transcription factors. Specifically, dim light-cultured maize seedlings are first treated with methyl jasmonate, followed by preparation of protoplasts and transfection of target genes driven by the ubiquitin promoter. The transfected protoplasts were cultured overnight in glass headspace tubes. Maize protoplasts were extracted by n-pentane and concentrated, and after being heated, the volatile terpenes were absorbed by solid-phase microextraction and analyzed on a gas chromatograph. This system was successfully used to detect the products of ZmTPS10, (E)-β-farnesene and (E)-α-bergamotene and to analyze the function of the transcription factor ZmMYC2a in regulating the production of (E)-β-farnesene and (E)-α-bergamotene. Understanding its pathogenic mechanisms is critical for developing novel control strategies. In this study, we investigated the role of spermine and its transporter BcTpo1 in fungal virulence. Spermine, but not spermidine, specifically inhibited conidial germination, infection structure formation, and pathogenicity. Deletion of BcTpo1 resulted in pleiotropic defects, including altered mycelial growth, conidiation, sclerotia formation, and melanin biosynthesis. Under oxidative stress, ΔBcTpo1 exhibited reduced intracellular spermine levels compared to the wild-type strain, correlating with elevated hydrogen peroxide sensitivity and impaired infection cushion formation. Exogenous spermine partially rescued these defects by enhancing antioxidant enzyme (superoxide dismutase and peroxidase) activities. However, the evolution of resistance by pests has reduced the efficacy of Bt crops. Pest resistance to Bt crystalline (Cry) proteins has spurred a shift to crops that produce the Bt vegetative insecticidal protein Vip3Aa together with Cry proteins. With widespread field-evolved practical resistance of the major pest Helicoverpa zea (corn earworm or bollworm) to Cry proteins in the United States, Vip3Aa is the only Bt protein in transgenic corn and cotton that is highly effective against many populations. We generated five strains of H. zea with > 100-fold resistance to Vip3Aa in a susceptible genetic background using F<sub>2</sub> screens of field populations from Louisiana, Mississippi, and Texas followed by crosses to a susceptible strain and additional laboratory selection with Vip3Aa. The results from diet bioassays show that in these Vip3Aa-resistant strains, cross-resistance to Cry1Ab, Cry1Ac, Cry1A.105, Cry1Fa, and Cry2Ab was not consistent and ranged 1100-fold: from 16-fold negative cross-resistance to 65-fold positive cross-resistance. By crossing a Vip3Aa-resistant strain with a Cry-resistant strain followed by selection with both types of Bt protein, we generated a strain highly resistant to Cry1Ac, Cry2Ab, and Vip3Aa. Cry1Ac and Cry2Ab interacted synergistically with Vip3Aa against this triple-resistant strain, such that combinations of the Cry proteins and Vip3Aa caused higher mortality than expected from their individual toxicity. The observed synergy may increase the durability of Bt crops producing combinations of these proteins against H. zea.</p>","PeriodicalId":519973,"journal":{"name":"Crop health","volume":"4 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2026-04-09","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC13066062/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147641131","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

However, how crop rhizosphere microbiomes reorganize under VTM-derived stress-particularly when bacteria and fungi are considered together-remains poorly understood. In the Hongge district (Panzhihua, China), we sampled bulk soils and rhizospheres of lettuce, rapeseed, and pea from croplands within the VTM mining influence zone (mining-impacted fields) and paired croplands outside the zone (reference fields). We measured soil chemistry and profiled bacterial and fungal communities using 16S rRNA and ITS amplicon sequencing, respectively. Mining-impacted soils generally showed a VTM geochemical imprint, neutral-alkaline pH, and reduced plant-available P and K. Bray-Curtis-based ordinations indicated a clear separation between mining-impacted and reference rhizospheres, and taxonomic profiles suggested host-dependent reassembly of both bacterial and fungal communities. Putative functional profiling suggested a shift toward stress-accommodation processes, and fungal guild assignments tended to tilt toward saprotrophic/endophytic categories. Mantel analyses identified pH as one of the strongest correlates of community turnover, whereas structural equation modeling was consistent with nutrient availability (available N/P/K composite) explaining a substantial portion of the VTM effect; the composite total-metal-load axis (Fe/V/Ti/Zn) showed limited explanatory power, especially for fungi. Using solid-phase microextraction (SPME), we characterized the variability in the sex pheromone composition of C. suppressalis females and its implications for male attraction. Although the female reproductive system matures by day one, the titer and release rhythm of the major component, (cis)-11-hexadecenal [(Z)-11-16:Ald], showed substantial individual variation, particularly among females aged 1 to 3 days. Two-day-old females exhibited the most consistent detection frequency and the longest release window, accompanied by a significantly altered ratio of (Z)-11-16:Ald to the minor component (cis)-9-hexadecenal [(Z)-9-16:Ald]. A positive correlation was established between the proportion of (Z)-11-16:Ald in the blend and its absolute titer. Higher temperatures increased the release of (Z)-11-16:Ald from the pheromone gland surface, while mating drastically reduced it. Subsequent field trials demonstrated that the total dosage influenced the attractiveness of different pheromone ratios. At a low dose of 760 μg, the (Z)-11- to (Z)-9-16:Ald ratios had no significant effect, with 10:1, 16:1, and 32:1 each attracting a comparable number of males. At a medium dose of 1520 μg, the 10:1 and 16:1 ratios were most attractive, while at a higher dose of 2280 μg, the 16:1 ratio was clearly superior. Crucially, after five years of mass trapping with a fixed 10:1 blend, local populations shifted their behavioral responses: one site showed enhanced attraction to a 30:1 ratio, while another exhibited increased response to a 7:1 ratio, indicating directional adaptation. Intraspecific variation in female pheromone release and male response is substantial in C. suppressalis. Long-term use of a single pheromone blend can drive behavioral resistance, necessitating rotation of multiple blends or integration with complementary control tactics for sustainable pest management.</p>","PeriodicalId":519973,"journal":{"name":"Crop health","volume":"4 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2026-04-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC13038809/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147597360","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

In Fusarium graminearum, a devastating cereal pathogen, SMs are best known as virulence factors, but their broader contributions to fungal physiology are poorly defined. Here, we present a genome-scale functional dissection of 53 predicted SM-BGCs by constructing a knockout library targeting cluster backbone genes and systematically quantifying 24 phenotypic traits, generating 1,272 phenotypic measurements. This dataset reveals that secondary metabolism is not a dispensable metabolic burden; instead, SM-BGCs are broadly integrated into vegetative growth, asexual development, and abiotic stress adaptation. Transcriptome analyses further uncover pronounced spatiotemporal regulation and tissue-dependent requirements of SM-BGCs during infection of wheat heads versus coleoptiles, thereby revealing an ecological dimension of pathogenesis. Mechanistic investigation identified two previously uncharacterized clusters, PKS-type BGC36 and NRPS-type BGC47, as critical for full virulence. BGC36 positively regulates deoxynivalenol (DON) biosynthesis, whereas disruption of BGC47 compromises cell wall/membrane stress tolerance and is associated with reduced phosphorylation of the kinase Mgv1, impaired DON-toxisome formation, and reduced DON production.
