Trametinib is considered to be a promising potential indirectly targeted KRAS inhibitor in PDAC. However, the clinical outcomes were poor. JQ1 displayed a significant synergistic effect when combined with chemotherapy or potential targeted therapy in pancreatic cancer. The impact of Trametinib and JQ1 combination treatment in PDAC remains to be fully elucidated.</p><p><strong>Methods: </strong>The efficacy of trametinib and JQ1 on cell proliferation and cytotoxicity was assayed in 7 <i>KRAS</i> mutant pancreatic cancer cell lines. The cytotoxic effects of drugs either alone or in combination were evaluated using a luminescent cell viability assay. Immunoblot analysis was carried out to investigate changes in p62 and autophagy.</p><p><strong>Results: </strong>We found that either trametinib or JQ1 alone inhibited the proliferation of some pancreatic cancer cell lines with <i>KRAS</i> alterations, irrespective of the mutational loci of <i>KRAS</i> and the aberrant status of the other driver genes. The synergistic effects of combination treatment of trametinib and JQ1 were observed in both trametinib-resistant and trametinib-sensitive cells. In trametinib-sensitive PDAC cells, the combined treatment definitely inhibited p62 expression compared with trametinib alone, while LC3 expression at high levels changed little. In trametinib-resistant PDAC cells, the combination of MEK/BET inhibitor dramatically decreased p62 expression compared with single agent, while p62 expression increased after anti-autophagic therapy was added.</p><p><strong>Conclusions: </strong>Blocking RAS downstream signaling and epigenetic pathway synergistically increases the antiproliferative activity in <i>KRAS</i> mutant PDAC cells. The Schur multiple zeta functions are a generalization of the multiple zeta functions of Euler-Zagier type. Among many relations, the duality formula and its generalization are important families for both Euler-Zagier type and Schur type multiple zeta values. In this paper, following the method of previous works for multiple zeta values of Euler-Zagier type, we give an interpolation of the sums in the generalized duality formula, called Ohno relation, for Schur multiple zeta values. {"title":"On $mu$-Zariski pairs of links","authors":"M. Oka","doi":"10.2969/jmsj/89138913","DOIUrl":"https://doi.org/10.2969/jmsj/89138913","url":null,"abstract":"The notion of Zariski pairs for projective curves in $mathbb P^2$ is known since the pioneer paper of Zariski cite{Zariski} and for further development, we refer the reference in cite{Bartolo}.In this paper, we introduce a notion of Zariski pair of links in the class of isolated hypersurface singularities. Such a pair is canonically produced from a Zariski (or a weak Zariski ) pair of curves $C={f(x,y,z)=0}$ and $C'={g(x,y,z)=0}$ of degree $d$ by simply adding a monomial $z^{d+m}$ to $f$ and $g$ so that the corresponding affine hypersurfaces have isolated singularities at the origin. They have a same zeta function and a same Milnor number (cite{Almost}). We give new examples of Zariski pairs which have the same $mu^*$ sequence and a same zeta function but two functions belong to different connected components of $mu$-constant strata (Theorem ref{mu-zariski}). Two link 3-folds are not diffeomorphic and they are distinguished by the first homology which implies the Jordan form of their monodromies are different (Theorem ref{main2}). We start from weak Zariski pairs of projective curves to construct new Zariski pairs of surfaces which have non-diffeomorphic link 3-folds.
