The finding was made in the municipality of Bacabal, in a transition zone between the Amazonian Domain and the Cerrado. Microalgae are a promising biomass source for biofuels, and this work evaluates their growth rates, lipid content, and fatty acid profiles. It is highlighted that Pseudokirchneriella subcapitata exhibits the highest growth rate (μmax), while Coelastrum sp. and P. subcapitata show the highest lipid levels. Palmitic acid is the main fatty acid accumulated in these microalgae. {"title":"Fungal Identification for Microalgae Biomass Hydrolysis using FTIR","authors":"Sofía Valentina Ordoñez Morales","doi":"10.59801/sma.v2i1.183","DOIUrl":"https://doi.org/10.59801/sma.v2i1.183","url":null,"abstract":"Fungi offer a promising avenue for utilizing residual microalgae biomass after lipid extraction, harboring valuable carbohydrates that can be converted into useful compounds, such as sugars. Fourier Transform Infrared Spectroscopy (FTIR) stands out as an effective and environmentally friendly tool for selecting microorganisms with biotechnological potential. This study utilizes FTIR to identify suitable fungal strains for the hydrolysis of microalgae biomass (Desmodesmus subspicatus and Chlorella sp) post-lipid extraction. Eleven fungal strains are selected, applying FTIR and multivariate analysis to choose filamentous fungi. The maximum cell density is 28.7 × 106 cells mL-1 for Chlorella sp. and 15.8 × 106 cells mL-1 for D. subspicatus, with total carbohydrates of 23.1% and 16.9%, respectively. Principal Component Analysis (PCA) and Hierarchical Cluster Analysis (HCA) are useful for strain identification. Following the analysis, Penicillium G12 stands out for its ability to utilize D. subspicatus biomass, generating glucose and xylose (sugar yields of 9.4% and 6.6% respectively), and Trichoderma auricularis for Chlorella sp. (sugar yields of 12.9% and 9.6% respectively). FTIR proves successful in detecting fungal strains with biotechnological potential.","PeriodicalId":446097,"journal":{"name":"Revista Boaciencia. The effectiveness of controlled composting (with rotation and irrigation) and natural composting was compared. Over a period of 120 days, tests were conducted on two composting beds using different materials (soy straw, wheat straw, tree prunings, and combinations thereof). Variables such as temperature, humidity, pH, carbon and nitrogen content, as well as reduction in mass and volume, were evaluated. The controlled treatments maintained appropriate humidity, exhibited a milder thermophilic phase, and achieved greater reduction in mass and volume. pH values remained neutral in all cases, and a decrease in the carbon/nitrogen ratio was observed, indicating the formation of mature compost. In summary, controlled composting proved to be more efficient in transforming organic matter during the 120-day study period, regardless of the types of waste used.","PeriodicalId":446097,"journal":{"name":"Revista Boaciencia. La Espectroscopía Infrarroja por Transformada de Fourier (FTIR) ha demostrado ser una herramienta eficiente y de menor impacto ambiental para la selección de microorganismos con potencial biotecnológico. Este estudio tuvo como objetivo aplicar FTIR para la selección de cepas de hongos con potencial para hidrolizar la biomasa de las microalgas Desmodesmus subspicatus y Chlorella sp después de la extracción de lípidos. Se seleccionaron once cepas fúngicas para la hidrólisis de biomasa residual y se aplicó FTIR seguido de análisis multivariado para la selección de hongos filamentosos. La mayor densidad celular fue de 28.7× 106 células mL-1 para Chlorella sp. y 15,8 × 106 células mL-1 para D. subspicatus y los valores de contenido de carbohidratos totales fueron 23,1 y 16,9%, respectivamente. El análisis de componentes principales (PCA) y el análisis de conglomerados jerárquicos (HCA) fueron herramientas útiles para detectar cepas de hongos. Luego del análisis multivariante, se pudo observar que las cepas de hongos que presentaron mayor capacidad de aprovechamiento de la biomasa de microalgas fueron Penicillium G12 debido a los azúcares glucosa y xilosa obtenidos tras la extracción de lípidos de D. subspicatus (con rendimiento de azúcar de 9.4 y 6.6 %, respectivamente) y Trichoderma auricularis para Chlorella sp. Muchos laboratorios e industrias de todo el mundo buscan cepas más productivas y mejores modelos de cultivo. El objetivo de este trabajo fue evaluar los parámetros cinéticos y de crecimiento de seis microalgas, y los contenidos de humedad y lípidos y perfiles de ácidos grasos de la biomasa cosechada al final de cada cultivo. Se aislaron microalgas en la Región Centro-Oeste de Brasil cultivadas en sistemas lagunares abiertos. Se observó que entre las seis microalgas utilizadas en este trabajo, Pseudokirchneriella subcapitata mostró la mayor tasa máxima de crecimiento específico (μmax) (0,12 día-1). La microalga Coelastrum sp. y P. subcapitata presentaron los niveles más altos de lípidos, con aproximadamente 20% de masa seca. El principal ácido graso acumulado en estas condiciones fue el ácido palmítico, con porcentajes del 59 al 69% para todas las microalgas evaluadas.
