The research was conducted at the Tissue Culture Seed Technology Laboratory, Faculty of Agriculture, Padjadjaran University, Jatinangor. The study was conducted from October 2017 to February 2018. The source of planting material is in the form of shoots from the turmeric rhizome. The source of explants or planting material came from the field collected at the Tissue Culture Seed Technology Laboratory, Faculty of Agriculture, Padjadjaran University. Explants were taken from rhizome buds with a size of 0.6-2.0 cm. The experiment used a Completely Randomized Design which was analyzed using the Student’s T-test method. The number of experimental and control groups in this study were seven groups. Variation in treatment with different BAP, thidiazuron, zeatin, and NAA concentrations in each group. The results show that Thidiazuron 1 mgL-1 + NAA 1 mgL-1 gives better results on the percentage of live explants and number of shoots on turmeric plants (Curcuma domestica Val.) Clones 41 at the age of 14 weeks after planting.RESPONS EKSPLAN KUNYIT PADA SITOKININ DAN AUKSIN DALAM MEDIA MURASHIGE DAN SKOOGPenelitian ini bertujuan untuk mendapatkan salah satu konsentrasi dari beberapa jenis sitokinin dan auksin untuk induksi tunas kunyit secara in vitro. Penelitian dilakukan di Laboratorium Kultur Jaringan Teknologi Benih, Fakultas Pertanian, Universitas Padjadjaran, Jatinangor. Waktu pelaksanaan penelitian ini dimulai pada awal bulan Oktober 2017 sampai bulan Februari 2018. Sumber bahan tanam berupa tunas dari rimpang tanaman kunyit. Sumber eksplan atau bahan tanam berasal dari lapangan yang dikoleksi di Laboratorium Kultur Jaringan Teknologi Benih, Fakultas Pertanian, Universitas Padjadjaran. Eksplan diambil dari mata tunas rimpang dengan ukuran 0,6-2,0 cm. Percobaan menggunakan Rancangan Acak Lengkap yang dianalisis menggunakan metode Student’s T-test. Jumlah kelompok eksperimen dan kontrol dalam penelitian ini adalah tujuh kelompok. Variasi perlakuan dengan penambahan konsentrasi BAP, thidiazuron, zeatin, dan NAA yang berbeda pada setiap kelompok. Growth Regulating Substances (GRS) used are BAP, TDZ, Zeatin and NAA. The explant source used was derived from the shoot of turmeric clones from Bogor. The experimental design in this study was Completely Randomized Design (CRD) with seven treatments and four replications. The planting medium used was Murashige Skoog Modification (MS) with the addition of Benzyl Amino Purine (BAP) 9 mg L-1, Thidiazuron (TDZ) 1 mg L-1, Zeatin 0.1 mg L-1 and NAA (0.01 mg L-1, 1 mg L-1). The results show that the combination of BAP 9 mg L-1 and NAA 0.01 mg L-1 produced the highest shoot induction at 12 weeks after planting. Giving zeatin 0.1 mg L-1 and NAA 1 mg L-1 produces the highest shoot length at 12 weeks after planting.PENGUJIAN MULTIPLIKASI EKSPLAN KUNYIT DENGAN PENAMBAHAN AUKSIN DAN SITOKININ PADA MODIFIKASI MEDIAPenelitian ini bertujuan untuk mendapatkan jenis dan konsentrasi sitokinin serta konsentrasi auksin yang optimal untuk multiplikasi tunas kunyit secara in vitro. Percobaan dilaksanakan pada bulan September 2017 sampai Februari 2018 di laboratorium Kultur Jaringan Teknologi Benih Fakultas Pertanian Universitas Padjadjaran. Bahan yang digunakan dalam penelitian ini adalah murashige dan skoog modified multiplication medium, agar-agar, aquades steril, HgCl2, alkohol 70%, clorox, tween 80, fungisida, dan bakterisida. Zat Pengatur Tumbuh (ZPT) yang digunakan yaitu BAP, TDZ, Zeatin, dan NAA. Sumber eksplan yang digunakan yaitu berasal dari tunas kunyit klon asal Bogor. Rancangan percobaan pada penelitian ini yaitu Rancangan Acak Lengkap (RAL) dengan tujuh perlakuan dan empat ulangan. Ettingsh mindi plant parasite (Melia azedarach L.). This research was conducted by maceration method using ethanol solvent, partitioning sequentially with n-hexane, chloroform, and ethyl acetate. Ethyl acetate fraction was separated by gravity column chromatography (GCC) in two stages. Phase I GCC used n-hexane : ethyl acetate (9 : 1). Phase II GCC used chloroform : methanol (9 : 1) eluent to obtain one pure compound. Purity identification used thin layer chromatography. Characterization of pure compounds obtained was carried out using UV-Vis and IR. The results show that the isolated compounds maximum wavelengths are at 351.20, 262.60, and 207.20 nm which corresponded to the conjugated synamoyl, benzoyl and chromophore phenol. IR spectrum data shows the presence of O-H, C-H aliphatic, C = O carbonyl, C = C aromatic, and C-O. From these data, the isolated compounds show flavonoid type flavanols.IDENTIFIKASI SENYAWA METABOLIT SEKUNDER DARI FRAKSI ETIL ASETAT BATANG Dendropthoe falcata.Penelitian ini bertujuan untuk mengidentifikasi senyawa metabolit sekunder dari fraksi etil asetat batang Dendrophtoe falcata (L.f.) Ettingsh parasit tumbuhan mindi (Melia azedarach L.). Penelitian ini dilakukan dengan metode maserasi menggunakan pelarut etanol, partisi secara berurutan dengan n-heksana, kloroform, dan etil asetat. Fraksi etil asetat dipisahkan secara kromatografi kolom gravitasi (KKG) dalam dua tahap. KKG tahap I menggunakan eluen n-heksana : etil asetat (9 : 1). KKG tahap II menggunakan eluen kloroform : metanol (9 : 1) sehingga diperoleh satu senyawa murni. Identifikasi kemurnian menggunakan kromatografi lapis tipis. Karakterisasi senyawa murni yang diperoleh dilakukan menggunakan UV-Vis dan IR. Berdasarkan hasil analisis dengan spektrofotometer UV-Vis, senyawa hasil isolasi menunjukkan panjang gelombang maksimum pada 351,20; 262,60; dan 207,20 nm yang sesuai dengan gugus sinamoil, benzoil, dan kromofor fenol terkonjugasi. Data spektrum IR menunjukkan adanya ikatan O-H, C-H alifatik, C=O karbonil, C=C aromatik, dan C-O. {"title":"THE APPLICATION OF BIORATIONAL PESTICIDES ON PADDY STEM BORER AND BACTERIAL LEAF BLIGHT","authors":"Y. Incertulas) insects and testing the effectiveness of biocontrol agents consisting of a mixture of endophytic bacterial cultures (Bacillus firmus E65 and Burkholderia sp E76) and insect pathogens (Serratia marcescens, SKM) based on kaolin bioformulation to control HDB disease. Tests were carried out on the conditions of farmers’ organic land in Cianjur, West Java by installing pheromones to catch male insects in the experimental plots, and applying kaolin-based biopesticide formulations (2.5 g / L). The results show that the application of pheromone traps by using 100 traps in an area of 10 ha, caused the population of male insects to be caught to decrease by less than 40 moths per month. It also effects stem borers to decrease into zero infestations, while stem borer infestation in plots that are not applied to pheromones show <10% infestation. The effect of bioformulation on non-Sintanur cultivars on other minor pests also decrease compared to controls (cv. Sintanur). Kaolin-based bioformulation shows a decrease in bacterial leaf blight of about 84.7% compared to untreated (without bioformulation) plots.APLIKASI PESTISIDA BIORASIONAL TERHADAP PENGGEREK BATANG DAN HAWAR DAUN BAKTERI PADIPenelitian ini bertujuan untuk mempelajari efektivitas perangkap feromon untuk mengendalikan serangga penggerek batang padi kuning (PBPK) (Scirpophaga. incertulas) dan menguji efektifitas agen biokontrol yang terdiri atas campuran kultur bakteri endofitik (Bacillus firmus E65 and Burkholderia sp E76) dan patogen serangga (Serratia marcescens, SKM) berbasis bioformulasi kaolin untuk mengendalikan penyakit HDB. Pengujian dilakukan pada kondisi lahan organik petani di Cianjur Jawa Barat dengan cara memasang feromon untuk menangkap serangga jantan pada petak percobaan, dan melakukan aplikasi formulasi biopestisida berbasis kaolin (2,5 g/L). Hasil penelitian menunjukkan bahwa aplikasi perangkap feromon dengan mengguna-kan 100 perangkap pada area seluas 10 Ha, menyebabkan populasi serangga jantan yang tertangkap menurun kurang dari 40 ngengat per bulan, dan serangan hama penggerek batang juga menurun menjadi nol infestasi, sementara serangan hama penggerek batang pada petak yang tidak diaplikasi feromon menunjukkan <10% infestasi. Pengaruh bioformulasi pada kultivar non-Sintanur terhadap hama minor lainnya juga menurun dibandingkan dengan kontrol (cv. Sintanur). Bioformulasi berbasis kaolin menunjukkan penurunan penyakit hawar daun bakteri sekitar 84,7% dibandingkan dengan petak tanpa perlakuan (tanpa bioformulasi). The study was carried out in December 2017 until February 2018. The soil analysis was carried out at the Integrated Testing Laboratory, Vegetable Crops Research Institute, Lembang, West Java. The manufacture of micron and nano volcanic ash and phosphate rock particles was carried out at the Nanotechnology and Graphine Laboratory, Mathematics and Science Faculty, Padjadjaran University and at Tekmira Research Center, Bandung. The research design used was a Randomized Block Design (RBD) consisting of 10 treatments of the combination of volcanic ash and phosphate rocks which were repeated three times. The results show that there is a significant effect of volcanic ash and phosphate rocks of normal size to a decrease in P retention of up to 42.87%. The combination of the treatment of normal-sized volcanic ash and nano-sized phosphate rocks has a significant effect on the total leaf area of the Gambung 7 clone plant, but it did not significantly affect the available P and delta pH.PENGARUH UKURAN PARTIKEL ABU VULKANIK DAN BATUAN FOSFAT PADA ANDISOLPenelitian ini bertujuan untuk mengetahui pengaruh pemberian berbagai ukuran partikel abu vulkanik dan batuan fosfat, meliputi ukuran normal (2mm), mikron (200 µm) dan nano (400 ղm) terhadap P tersedia, retensi P, delta pH dan luas daun bibit tanaman teh di Perkebunan Teh PTPN XII Ciater, Jawa Barat. Penelitian lapangan dilaksanakan pada Desember 2017 sampai Februari 2018. Analisis tanah dilakukan di Laboratorium Penguji Terpadu, Balai Penelitian Tanaman Sayuran, Lembang, Jawa Barat. Pembuatan partikel mikron dan nano abu vulkanik dan batuan fosfat dilakukan di Laboratorium Nanoteknologi dan Graphine, Fakultas MIPA, Universitas Padjadjaran dan di Puslitbang Tekmira, Bandung. Rancangan penelitian yang digunakan adalah Rancangan Acak Kelompok (RAK) yang terdiri atas 10 perlakuan kombinasi ukuran abu vulkanik dan batuan fosfat yang diulang sebanyak tiga kali. Hasil penelitian ini menunjukkan bahwa terdapat pengaruh nyata pemberian abu vulkanik dan batuan fosfat yang berukuran normal terhadap penurunan retensi P hingga 42,87%. This research was conducted in traditional markets with heaver objects which are often carried out in the process of moving goods. This study used biomechanics and physiology methods to be able to analyze body posture so that it can prevent spinal damage. The data were then analyzed using Maximum Permissible Limit (MPL), which previously carried out data collection on the size of the body posture. The analysis of the results was done to determine the value of the compression force (FC). FC values are the basis for comparison with Action Limit (AL). The results show that FC values = 10,018,078N, FC> MPL. Importantly, the position of the heavers’ posture is at a dangerous level. Proposed improvements using a tool and after being developed again will result in changes in FC values to 3370.94, FC <AL means the position of the body in a safe condition. The results of this study are expected to provide an assessment that helps prevent spinal damage.MANUAL MATERIAL HANDLING PADA PROSES PENGANGKATAN KARUNG MENGGUNAKAN PENDEKATAN BIOMEKANIKA DAN FISIOLOGI Penelitian ini bertujuan untuk meminimumkan keluhan Musculoskeletal disorder (MSDs) pada pekerja kuli panggul di pasar tradisional dengan merancang fasilitas dan memperbaiki teknik memindahkan barang yang sesuai dengan aspek ergonomis. Penelitian ini dilakukan di pasar tradisional dengan objek kuli panggul yang sering melakukan proses pemindahan barang. Penelitian ini menggunakan metode pendekatan biomekanika dan fisiologi untuk dapat menganalisis postur tubuh sehingga dapat mencegah cedera tulang belakang. Data kemudian dianalisa menggunakan Maximum Permissible Limit (MPL) yang sebelumnya dilakukan pengambilan data ukuran postur tubuh pekerja tersebut. Analisis hasil dari perhitungan dilakukan untuk mengetahui nilai dari force compresion (FC). Nilai FC menjadi dasar untuk dibandingkan dengan Action Limit (AL). Hasil dari analisa didapat nilai FC = 10.018,078N, FC>MPL. Artinya, posisi postur tubuh pekerja dalam level berbahaya. Usulan perbaikan dengan menggunakan alat bantu dan setelah dianalisis kembali maka akan menghasilkan perubahan nilai FC menjadi 3370,94; FC< AL artinya posisi tubuh dalam keadaan aman. The study was conducted from February 2018 to April 2018 at the Ciparanje Experimental Field, Faculty of Agriculture, Padjadjaran University. The study used a Randomized Block Design (RBD) with nine treatments and three replications, consisting of controls; mixed hydrogels and urea incubation and urea only incubation . The parameters observed were pH using the Potentiometric method, C-Organic using the Walkey and Black and N-Total using the Kjeldahl method. The data then were analyzed by variance. Significant testing was conducted to determine the effect of treatment using the Fisher Test at the level of 5%. The results show that there is an effect of hydrogels and the incubation time of hydrogels on pH, organic C and N-total soil. The control treatment shows the highest pH of 6.88. The urea treatment and incubation hydrogel of 20 days shows the highest organic C at 2.80% and the highest N-total is found in the treatment of urea and 40 day incubation hydrogel which is 0.34%.PERAN HIDROGEL DAN WAKTU INKUBASINYA TERHADAP pH, C-ORGANIK, DAN N-TOTAL TANAHPenelitian bertujuan untuk mengetahui peran hidrogel dan waktu inkubasinya terhadap pH, C-organik dan N-total tanah dengan indikator tanaman jagung manis (Zea mays Saccharata Sturt) pada inceptisols Jatinangor. Penelitian dilakukan mulai bulan Februari 2018 hingga April 2018 di Lahan Percobaan Ciparanje Fakultas Pertanian Universitas Padjadjaran. Penelitian menggunakan Rancangan Acak Kelompok (RAK) dengan sembilan perlakuan dan tiga kali ulangan, yang terdiri atas kontrol; hidrogel dan urea inkubasi serta urea inkubasi. Parameter yang diamati adalah pH dengan menggunakan metode Potensiometri, C-Organik dengan menggunakan metode Walkey and Black dan N-Total dengan menggunakan metode Kjeldahl. Data hasil pengamatan pada penelitian akan dianalisis dengan sidik ragam. Pengujian signifikan untuk mengetahui pengaruh perlakuan digunakan Uji Fisher pada taraf 5%. Hasil penelitian menunjukkan terdapat pengaruh hidrogel dan waktu inkubasi hidrogel terhadap pH, C-organik dan N-total tanah. Perlakuan kontrol menunjukkan pH tertinggi yaitu 6,88. Perlakuan urea dan hidrogel inkubasi 20 hari menunjukkan C-organik tertinggi yaitu 2,80% dan N-total tertinggi yaitu perlakuan urea dan hidrogel inkubasi 40 hari yaitu 0,34%. The material used in this study was Andisol and steel slag. The Andisol was obtained from PTPN VIII-Ciater tea plantations (1250 m asl) on the eastern slope of Tangkuban Perahu Mountain, Subang, West Java. The steps taken were soil sampling using disturbed soil extraction methods using Minipit technique, soil sample filtering, weighing, mixing soil samples with steel slag from PT Krakatau Steel Serang, refining done using the Top-Down method, incubating and then analyzing properties soil chemistry. The chemical properties of the soil analyzed were P, P-available and H-dd retention. P retention was analyzed by the Blackmore method, P-available was analyzed using the Bray I, Bray II, and Olsen methods. H-dd was analyzed by the Titration method. The data obtained were then carried out statistical tests with variance analysis (ANOVA) with RAL environmental design and Duncan test. The results showed that the micro-application of steel slag particles had an effect on available P and P retention but had no affect on H-dd.PENGARUH PARTIKEL MIKRO TERAK BAJA TERHADAP RETENSI FOSFOR DAN HIDROGEN PADA ANDISOLPenelitian ini bertujuan untuk mengetahui pengaruh partikel mikro terak baja terhadap retensi Fosfor (p), p-tersedia dan Hidrogen dapat dipertukarkan (H-dd) pada Andisol. Bahan yang digunakan dalam penelitian ini adalah andisol dan terak baja. Andisol didapat dari perkebunan teh PTPN VIII-Ciater (1250 m dpl) pada lereng bagian timur Gunung Tangkuban Perahu, Subang, Jawa Barat. Tahapan yang dilakukan adalah pengambilan sampel tanah menggunakan metode pengambilan tanah terganggu dengan teknik minipit, penyaringan sample tanah, penimbangan, pencampuran sampel tanah dengan terak baja yang berasal dari PT Krakatau Steel Serang, penghalusan yang dilakukan dengan metode Top-Down, penginkubasian dan kemudian penganalisaan sifat kimia tanah. Sifat kimia tanah yang dianalisi adalah retensi P, P-tersedia dan H-dd. Retensi P dianalisis dengan metode Blackmore, P-tersedia dianalisis menggunakan metode Bray I, Bray II, dan Olsen. H-dd dianalisi dengan metode titrasi. Data yang didapat selanjutnya dilakukan uji statistik dengan analisis sidik ragam (Anova) dengan rancangan lingkungan RAL dan uji Duncan. The experimental design used was a completely randomized design with a treatment factor of 16 genotypes of sweet corn with 2 replications. The data were analyzed using the F test, if there was a significant difference in Scott Knott’s test at the level of 5%. All of the seed genotypes had different qualities, so there was a significant difference in 100 grain weight parameters, germination and simultaneous growth based on ANOVA test. Seeds that had good initial physical quality are able to maintain quality until the end of storage period. After the storage period there is the best genotype, namely with code 613 which is able to maintain its physical-physiological quality until the end of storage. Based on the results of the study, it can be concluded that the genotype of sweet corn seed npad Genotype 613 (P5 x P1 (x) (3)) has good physical and physiological qualities and can be used as a candidate for hybrid elders. This is seen from the quality of seeds that meet the requirements of quality seed characters from 100 grain weight parameters and germination.PENGARUH MUTU GENOTIP BENIH JAGUNG MANIS UNPAD TERHADAP MUTU FISIOLOGISPenelitian ini bertujuan untuk mengetahui pengaruh mutu genotip benih jagung manis Unpad terhadap mutu fisiologis setelah empat bulan penyimpanan. Rancangan percobaan yang digunakan yaitu Rancangan Acak Lengkap dengan faktor perlakuan 16 genotipe jagung manis dengan 2 ulangan. Data dianalisis menggunakan uji F, apabila terdapat perbedaan yang signifikan dilakukan uji lanjut Scott Knott taraf 5 %.Semua genotip benih memiliki mutu yang berbeda, sehingga terdapat perbedaan yang signifikan pada parameter bobot 100 butir, daya berkecambah dan keserempakan tumbuh berdasarkan uji anova. Benih yang memiliki mutu fisik awal yang baik mampu mempertahankan mutu sampai periode akhir penyimpanan. Setelah periode simpan terdapat genotip terbaik yaitu dengan kode 613 yang mampu mempertahankan mutu fisik-fisiologisnya sampai akhir penyimpanan. Berdasarkan hasil penelitian dapat diambil kesimpulan bahwa genotip benih jagung manis Unpad Genotip 613 (P5 x P1 (x) (3)) memiliki mutu fisik dan fisiologis yang baik dan dapat dijadikan sebagai calon tetua hibrida. The subjects in this study were leaves of parasitic plants on Mindi plants. While the objects in this study were secondary metabolites from the ethyl acetate fraction of the leaves of parasite plants attached to the mindi plant. The research method was carried out by extracting macerated leaves of D. falcata with ethanol solvents. The ethanol extract obtained was partitioned using n-hexane, chloroform, and ethyl acetate. Ethyl acetate fraction was separated by gravity column chromatography (CCG). The relatively non-polar fraction that has shown a single stain is tested for purity using 3 different types of solvents and identified using UV-Vis, IR, and GC-MS. The results showed that the compounds that could be isolated and identified from the relatively non-polar ethyl acetate fraction of D. falcata leaves were stigmast-5-en-3β-ol with an abundance of 6.49%, having a molecular weight of m / z 414 with base peak 55. Subjek dalam penelitian ini adalah daun tumbuhan benalu pada tanaman mindi. Sedangkan objek dalam penelitian ini adalah senyawa metabolit sekunder dari fraksi etil asetat daun tumbuhan benalu yang menempel pada tanaman mindi. Metode penelitian dilakukan dengan ekstraksi maserasi daun D. falcata dengan pelarut etanol. Ekstrak etanol yang diperoleh dipartisi menggunakan n-heksana, kloroform, dan etil asetat. Fraksi etil asetat dipisahkan secara kromatografi kolom gravitasi (KKG). Fraksi relatif non polar yang sudah menunjukkan noda tunggal diuji kemurniannya menggunakan 3 jenis pelarut yang berbeda dan diidentifikasi menggunakan UV-Vis, IR, dan GC-MS. Hasil penelitian menunjukkan senyawa yang dapat diisolasi dan diidentifikasi dari fraksi etil asetat relatif nonpolar daun D. falcata adalah stigmast-5-en-3β-ol dengan kelimpahan 6,49 %, memiliki berat molekul m/z 414 dengan base peak 55. Senyawa sterol merupakan senyawa yag lazim ditemukan dalam tumbuhan benalu.
