The aim of this study was to identify the psycho-physiological peculiarities of Biology students and to establish the connection between the students’ academic performance, their cognitive abilities and their psycho-physiological characteristics, which is essential for improving the academic process and bringing it to the high level of professional education necessary for graduates of higher educational establishments to compete successfully in the job market. It was established that the majority (46%) of Biology students are ambiverts, 34% of them are extroverts, and 18% – introverts. There is a statistically reliable connection between the academic grades and the personality type: the more successful students are ambiverts and introverts. Extroverts demonstrate lower academic results. According to different types of asymmetry manifestation among Biology students, the right-handed accounted for 35% of the tested, those with prevailing right hand – 51%, those with prevailing left hand – 1.2% and the left-handed – 12%. There is no dependence of academic performance on the asymmetry manifestation. Most Biology students have a sanguine-choleric temperament and slightly expressed or absent asymmetry, which testifies to the power of nervous processes and is a precondition for successful studying. The majority of students, irrespective of the academic performance level, have a low level of stress resistance and a medium level of personality anxiety. Students who score A and B points for academic performance, have a medium level of situational anxiety. Those who score C points have a high level of anxiety. There is no connection between the academic points and personality anxiety level, but there is significant connection between academic performance and situational anxiety. There has been established a significant dependence of academic performance on the level of situational anxiety: introverts and extroverts have a high level, ambiverts have a medium level of situational anxiety. Introverts and ambiverts with a low and medium anxiety level demonstrate higher academic performance. Extroverts with a high anxiety level find it more difficult to study. The majority of Biology students have a high level of visual-imaginative, object-active and creative thinking types. 83% of students have a high and a medium level of verbal-logical thinking type. Abstract-symbolic and intuitive thinking are developed in lesser degree. There has been established a reliable negative correlative connection between object-active thinking type and introversion, and a positive connection between intuitive thinking and introversion, irrespective of academic results. Внутрішньом’язове уведення щурам дослідних груп 50% тетрахлоретану у дозі 0,25 мл на 100 г маси тіла тварини спричиняє антигенне навантаження на організм, викликає підвищення проникності клітинних оболонок гепатоцитів та мітохондріальних мембран. Про це свідчить зростання активності амінотрансфераз у сироватці крові протягом усього періоду досліджень. На другу добу досліджень відмічали підвищення АсАТ у 2,5 раза, АлАТ – удвічі, а коефіцієнта АсАТ/АлАТ – на 30% порівняно з контролем. На 14-ту добу досліджень активність даних показників залишалася вищою порівняно з показниками контрольної групи тварин: АсАТ і АлАТ відповідно на 93% та 81%, показник коефіцієнта був вищим і становив 2,89 ± 0,05 проти контролю 2,72 ± 0,04 (на 9% вище). Для нормалізації проникності клітинних оболонок гепатоцитів та мітохондріальних мембран за оксидаційного стресу доцільно застосовувати ліпосомальний препарат, який у своєму складі містить бутафосфан, інтерферон, розторопшу плямисту та вітаміни. У разі застосування ліпосомального препарату щурам, за оксидаційного стресу, у крові настає нормалізація активності ензимів переамінування (нормалізація активності АсАТ і АлАТ сироватки крові). На 14-ту добу досліду показники ензиматичної активності амінотрансфераз у крові дослідних щурів коливалися в межах фізіологічних величин, що вказує на нормалізацію проникності клітинних оболонок гепатоцитів та мітохондріальних мембран за оксидаційного стресу та за дії ліпосомального препарату.","PeriodicalId":345307,"journal":{"name":"Visnyk of Dnipropetrovsk University. Shevtsova","doi":"10.15421/021605","DOIUrl":"https://doi.org/10.15421/021605","url":null,"abstract":"Coronary heart disease is one of the major causes of illness and of deaths in Europe, therefore the definition of the signaling pathways involved in cardioprotection represents a challenging goal in order to discover novel pharmacological approaches. One of the mechanisms included in the implementation of signaling methods is proteolysis, so the aim of our study was to study the balance between activity of the proteolytic enzymes and their inhibitors in the experimental myocardial ischemia of rats and the impact of corvitin (C) and doxycycline (D) on these parameters. Ischemia was induced in Wistar rats by the intraperitoneal injection of epinephrine at a dose of 0.2 mg/100 g of weight during 10 days. All animals were divided into 4 groups: 1 – control (8 rats), 2 – rats with epinephrine-induced myocardial ischemia (EIM, n = 10), 3 – rats, which were entered C after completion of epinephrine injections (n = 10), 4 – rats, which accepted D during 6 days after finishing of epinephrine injections (n = 10). The electrical activity of the heart was recorded on an electrocardiogram (ECG). Activities of matrix metalloproteinases 2 and 9 (MMP2, MMP9) were studied by enzyme-zymography, the activity of trypsin-like enzymes (TLE), levels of alpha-1 proteinase inhibitor (IP1) and alpha-2-macroglobulin (α2-МG) in the blood and the heart were determined by spectrophotometric methods. The experimental results suggest that epinephrine injections lead to changes in the ECG typical for ischemic myocardium. The data obtained showed that the protease-іnhіbіtory balance in the blood and in heart was disturbed. Activity of MMP9, proMMP9/2 and level of IP1 increased in blood plasma of rats with EIM, but activities of ММP2, TLE were unchanged in this group. In the heart muscle the activity of MMP2 and level of IP1 increased. Administration of C resulted in the recovery of MMPs level, but disturbed the balance in the system TLE/IP1-α2-МG. Administration of D blocked the activity of MMP9, significantly reduced the activity of MMP2, which testifies the inhibitory effect of this drug on the activity of matrix metalloproteinases. Based on these results, we conclude that definition of protease-inhibitory balance in the blood plasma of patients with coronary artery disease is promising as an additional diagnostic complex and for monitoring of the effectiveness of therapy.","PeriodicalId":345307,"journal":{"name":"Visnyk of Dnipropetrovsk University. Spleen extract biologically active substances were used as immunomodulators and anti-stressors during pre-slaughter period. Biologically active substances influence on putrescin, spermine and spermidine content in broiler chicken blood before slaughter and on some non-specific resistance indices in people was determined after consumption of broiler chicken meat. Two groups of broiler chickens at one month age were formed for the study. The spleen extract obtained with ultrasound application (I research group) served as biologically active substances was added to the feed of broiler chickens in pre-slaughter period (five days before slaughter). Blood polyamines such as putrescin, spermine and spermidine were determined by the method of High-performance liquid chromatography (HPLC) on the liquid chromatograph Agilent 1200 (USA). The second experiment was conducted on 10 people. We recruited 10 healthy male medical students (20 years old, on average) after the National Medical license examination. Spleen extract polyamines as immunomodulators and anti-stressors have the most effective influence on total quantity of polyamines in broiler chicken blood. As a result of research, it is found that aerosol introduction of spleen extract into broiler chicken feed reliably increases total quantity of polyamines by 39% and, in particular, spermidine concentration by 34%, and spermine by 40% compared with broiler chickens of the control group. Some non-specific body resistance indices in men blood upon consumption of broiler chicken meat varied within the physiological norm. The neutrophils stimulation index increased in men blood (+0,82) after consumption of meat of broiler chickens to which spleen extract as immunomodulator and anti-stressor was introduced before slaughter (experimental group). The neutrophils stimulation index decreased in men blood (–2,21) after consumption of broiler chickens meat at pre-slaughter stress (control group). The neutrophils stimulation index in men blood of experimental and control groups differed by 34,8%, but did not go beyond the physiological norm. At the final stage of poultry feeding it is necessary to consider pre-slaughter stress and to apply biologically active substances of natural origin, such as spleen extract. Bulankina","doi":"10.15421/021529","DOIUrl":"https://doi.org/10.15421/021529","url":null,"abstract":"Проведено исследование экспрессии генов пяти металлотионеинов (МТ 1-5), убиквитина и белка р53 в культуре фибробластов кожи и л е гких белых крыс разного возраста (2 недели, 1, 3 и 24 месяца), а также определено количество их продуктов (металло­тио­неинов 1–5 типов, убиквитина, белка р53). Все эти белки являются защитными, но выполняют свои функции с использованием разных механизмов. Металлотионеины связывают, транспортируют и выводят из организма ионы двухвалентных металлов, убиквитин контролирует расщепление в протеасомах дефектных и короткоживущих белков, белок р53 контролирует механизм апоптоза, обеспечивая таким образом стабильность генома. В связи с этим задачей настоящей работы было сравнение экспрессии указанных генов и количества их белковых продуктов в фибробластах соединительной ткани кожи и л е гких для выявления роли генетических факторов в ходе их онтогенеза. Показано сходство возрастной динамики экспрессии генов МТ и убиквитина из клеток обоих источников – наличие максимума в 3 месяца. Возрастные изменения экспрессии генов всех пяти МТ характеризуются как в коже, так и в легких подъемом в первой половине онтогенеза. Для кожи характерен четкий максимум экспрессии генов МТ в 3 месяца, для л е гких этот максимум менее четок. К 24-м у месяц у экспрессия всех генов МТ в фибробластах как кожи, так и л е гких, резко падает в 2 – 3 раза по сравнению с максимумом. Экспрессия гена убиквитина, контролирующего расщепление короткоживущих или поврежд е нных белков, увеличивается в фибробластах легких к 3 месяцам в 1,5 раза, а далее снижается ниже уровня у двух недельных животных. В коже максимум экспрессии данного гена также приходится на 3 месяца, но снижение не такое существенное. Количество продукта убиквитинового гена в л е гких возрастает до трех месячного возраста и далее практически не меняется. В коже наблюдается падение в период от двух недель до одно го месяца, подъ е м к трем месяцам и резкий спад к старости (примерно в 3 раза по сравнению с трех месячным возрастом). Экспрессия гена р53 имеет иной характер: и в коже, и в л е гких повышение экспрессии происходит вплоть до 24 месяцев. Maslak","doi":"10.15421/021525","DOIUrl":"https://doi.org/10.15421/021525","url":null,"abstract":"141 Tumor growth progression of blood cells occurs due to changes in their genetic apparatus, which affects not only the cells morphological characteristics, but also their functional activity which to a greater extent depends on the membrane surface structures, a significant part of which is of glycoprotein nature. Complex type N-glycans are components of surface glycoproteins in the most of leukocytes. Thus, the study of changes in carbohydrate determinants of glycoproteins on the surface of leucocytes in tumorigenesis can help to reveal the mechanisms of this process. The aim of our study was to investigate the monocytes and granulocytes cytoplasmic membrane N-glycosylation in patients with chronic leukemia. The object of the study were blood cells of patients with chronic lymphocytic leukemia (n = 12) and polycythemia vera (n = 15) aged 58–66 years. Healthy hematologic volunteers (n = 15) aged 55 to 65 years were in the control group. N-glycan exposure on monocytes and granulocytes was investigated by flow cytometer Beckman Сoulter EPICS with Canavalia ensiformis lectin – Con A conjugated with fluorescent labels. The number of dead cells was monitored by means of binding them with propidium iodide. The result has been analyzed with FC Express. According to our data, levels of ConA-positive monocytes and granulocytes were 9,9 ± 1,0% and 32,7 ± 3,2%, respectively, in peripheral blood of healthy persons. The level of ConA-positive monocytes decreased to 31,0 ± 2,3% and the number of ConA-binding granulocytes increased to 66,7 ± 3,8% in patients with chronic lymphocytic leukemia compared with the norm. The number of ConA-positive monocytes decreased 3.3 times, and the level of granulocytes interacting with Canavalia ensiformis lectin slightly increased relative to control in polycythemia vera patients. There is significant increase in Con A-positive epitopes on granulocytes in patients with chronic lymphocytic leukemia and polycythemia vera compared with the control group, and absence of any difference in Con A-positive epitopes on monocytes. 100 times increase of the number of ConA-binding glycotopes is observed on the surface of granulocytes in peripheral blood of patients with polycythemia vera, and 3,3 times increase in patients with chronic lymphocytic leukemia, which may serve as an additional marker for the diagnosing these diseases.","PeriodicalId":345307,"journal":{"name":"Visnyk of Dnipropetrovsk University. Nevertheless, the corresponding subdivision in genetic structure is not revealed in previous investigations: populations of Ukrainians under study were genetically homogeneous on SNP markers of Y chromosome. In the current investigation we studied the Y-chromosomal genetic structure of Transcarpathia and Bukovina populations. Several factors exist to expect the genetic specificity of these populations. Both ones are placed in the Carpathian foothills, at the south-western border of the Ukrainian area. During the last millennium these territories were the parts of different states and were open for ethnically diverse migrations. It was revealed that the major Y chromosomal haplogroups in Transcarpathia population were R1a1a1*(М198), I2a (Р37.2), R1a1a1g (М458), E1b1b1a1 (M78). The major haplogroups in Bukovina population were I2a (Р37.2), R1a1a1*(М198), R1a1a1g (М458), R1b1b2 (М269), E1b1b1a1 (M78), I1 (М253). The Bukovina population differs from the typical Ukrainian population by higher frequency of I2a (Р37.2) and lower frequency of R1a1a1*(М198). Moreover, this is the only population among ones studied in Ukraine where the most frequent haplogroup is I2a (Р37.2) but not R1a1a1*(М198). Such a deviation can be caused by possible mixing with neighbouring southern groups, and Carpathian mountains were not a border for exchange in this case. Interestingly, the haplogroup N1c (M178) is not revealed in Transcarpathia at all, obviously due to the mountain barrier. It was revealed by principal component analysis that Ukrainians from Transcarpathia and Bukovina despite some specific peculiarities are more similar to other Ukrainian populations than to the surrounding ethnic groups such as Poles, Slovaks, Hungarians, Romanians, Moldavians and Gagauzes. Ukrainians of Transcarpathia and Bukovina form the entire genetic continuum with the whole Ukraine on maps of gene distances, confirming the homogeneity of Ukrainian parental gene pool and it’s differentiation from other groups. After performing the analysis of Y-haplogroup spatial distribution, it is supposed that the northern ridges ofCarpathian mountainsare the East-European barrier decreasing the gene flow. In high concentrations adrenaline increases the speed of protein catabolism. Working through the circulatory system, adrenaline affects almost all the functions of organs, causing the body mobiliz ation to counter stressful situations. Due to ELISA the astrocytes-specific protein (S-100b) and neural cell adhesion molecule (N-CAM) were studied . S - 100b is produced mainly by astrocytes іn the brain and depend ing on the concentration it cause s trophic or toxic effect on the neurons and glial cells. Strong stress and ischemia induce re - distribution of calcium-binding protein S-100b and elevation of its level. Quantitative changes of S-100b under the influence of various factors on the body which lead to the metabolic disorder in the brain are considered today as a sign of brain damage (cortical, ischemic one , etc.). Fluctuations in the concentration of S-100b in the brain are not always accompanied by marked deterioration of the physical condition of animals, but they can also lead to a number of violations of integrative functions of the brain depending on over - production of this protein. Most N-CAM are transmembrane proteins that cross the plasma membrane once ; intracellular domains have different size and it is thought they are involved in binding to cytoskeleton or cell signaling. Violation of N-CAM functions leads to disruption of nerve sprouts. Data o btained in our study showed no serio u s re - distribution of S-100b and N-CAM level in the different areas of rat brain (cerebral cortex, hippocampus and thalamus) under effect of adrenalin administered to the animals (under skin) in dosage of 0.45–0.60 mg per rat, 1 time per day during 10 days, probably because of the type of injection and/or short time of adrenalin action. Increased dosage of adrenaline 1 hour before decapitation lead s to the decrease of level of total protein in membrane fraction extracted from brain tissue without changing the level of S-100b and N-CAM.","PeriodicalId":345307,"journal":{"name":"Visnyk of Dnipropetrovsk University. Nedzvetsky","doi":"10.15421/021521","DOIUrl":"https://doi.org/10.15421/021521","url":null,"abstract":"Реактивація гліальних клітин, що індукується метаболічним розладом утилізації глюкози та розвитком окисного стресу в сітківці при цукровому діабеті, є центральним фактором патогенезу діабетичної ретинопатії. Наночастинки фулерену С 60 та окремі його водорозчинні похідні характеризуються потужними антиоксидантними властивостями та нейропротекторною дією у випадку значної кількості патологій та несприятливих впливів. Уперше досліджено вплив низьких доз гідратованого С 60 фулерену (C 60 HyFn) на експресію та поліпептидний склад гліального фібрилярного кислого протеїну (ГФКП) у сітківці щурів при діабеті, індукованому стрептозотоцином (СТЗ). Із застосуванням методу імуноблотингу показано зростання експресії ГФКП у сітківці щурів із діабетом порівняно з контролем у 1,93 раза, що свідчить про суттєву реактивацію гліальних клітин ретини при гіперглікемії. Підвищення ГФКП-імунореактивності, асоційоване з розвитком реактивного астрогліозу у сітківці діабетичних щурів, підтверджено імуногістохімічно на фіксованих зрізах сітківки. Споживання діабетичними щурами розчину C 60 HyFn (90 нМ) з питною водою протягом 12 тижнів викликало зниження вмісту ГФКП у 1,51 раза порівняно з цим показником у групі діабетичних тварин. C 60 HyFn сприяв достовірному (у 1,58 раза) зниженню вмісту глікозильованого гемоглобіну в сироватці крові щурів з СТЗ-діабетом. Наночастинки C 60 HyFn не змінювали рівень інсуліну та глюкози у крові діабетичних щурів. Отримані результати свідчать про те, що протекторна дія гідратованого фулерену при діабетичній ретинопатії зрілих тварин реалізується через пригнічення ним надмірної активації ГФКП-позитивних клітин сітківки.","PeriodicalId":345307,"journal":{"name":"Visnyk of Dnipropetrovsk University. Persky","doi":"10.15421/021523","DOIUrl":"https://doi.org/10.15421/021523","url":null,"abstract":"Mechanisms of influence of volumetric autotransplantation of fibroblasts and of the mixture of fibroblasts and keratinocytes on the development of the local 3rd degree X-ray burn and the radiation skin ulcer in guinea pigs were investigated. We used d eep administration into the irradiation zone on its perimeter of 6 doses, which contained (150 – 160)×10 3 fibroblasts and (130 – 140)×10 3 keratinocytes in 100 µl. It is shown that this autotransplantation carried out 1 hour after the irradiation, and then every 24 hours, reduces the area of burn on the 35th day , compared to the control by 63%. Radiation ulcer appears on the 10th day after irradiation and is completely healed on the 25th day. With the same regimen of administration of only fibroblasts containing (200 – 210)×10 3 cells in 100 µl, these parameters of treatment were equal to 31% on 4th and 35th day, respectively. It is shown that as a result of radiation in the area of burn the level of gene expression of collagen types I and III, elastin, fibronectin, vinculin, decorin, hyaluronan synthases 1, 2, 3 , matrix metalloproteinases 1, 2, 3, 7, 9 and hyaluronidase is reduce d . Besides, i n the burn area the level of gene expression of transforming growth factor α, fibroblast growth factors 1, 2, 8 and anti - inflammatory cytokines – interleukin 10 and transforming growth factor-β1 – is reduced, while the level of gene expression of proinflammatory cytokine ( interleykin 1β ) increase s . Both types of autotransplantation cause the growth of the expression level of all the structural genes and regulatory proteins of biopolymers and decrease in the expression level of interleukin 1β, which leads to activation of tissue regeneration and healing of the burn wound. Reasons for the higher efficiency of autotransplantation using the mixture of fibroblasts and keratinocytes compared to autotransplantation by fibroblasts only are both the larger total number of liv e cells regularly replacing dead cells in the burn area , and mutual stimulation of auto - fibroblasts and auto - keratinocytes to proliferate and to synthesize biologically active substances , i.e. cytokines and growth factors.","PeriodicalId":345307,"journal":{"name":"Visnyk of Dnipropetrovsk University.
