The tested substance was the copolymer PNIPAM-co-MAA. The particle size measurements were performed by dynamic light scattering. It was found that the copolymer tested reacts specifically to temperature increase by shrinking more than two times. Important for stabilization of the structure are the chemical groups -COOH present in methacrylic acid that undergoes dissociation. Gradual increase in temperature results in a decrease in the dissociation constant, in binding of protons and thus causes shrinkage of the entire particle. Identification of metabolic alterations like cholesterol metabolism or free radicals may provide insight into the formation of gallstones and provide a basis for prognostic markers. The aim of the study was to identify the pathogenesis and prognostic value of metabolic disorders in gallstone formation in menopausal women. Methods: Menopause-aged women with (group I, 58 patients) and without gallstone disease (group II, 25 patients) were investigated. In each group blood lipid metabolism and blood redox parameters (free Mn2+-ions) and antioxidant system activity (ceruloplasmine/Fe3+-transferrin) were studied using micro enzymatic method and Electron Pramagnetic Resonance (EPR) spectroscopy. Surgically removed gall bladder stones were studied by routine laboratory methods (cholesterol and bilirubin content) and EPR spectroscopy. In model experiments in vitro purified bilirubin was irradiated with visible light in different conditions (in presence and without oxygen). Results: Intensive signal of oxidized bilirubin free radical was detected in the EPR spectra of gallstones. Reduced activity of blood antioxidant ceruloplasmin/F3+-transferrin system and increased content of prooxidants Mn2+-ions were detected in menopausal women blood with gallstone disease. In vitro experiments demonstrated, that prolonged exposure of bilirubin to visible light in the presence of oxygen induces formation of the bilirubin free radicals (EPR signal g= 2.003 AH = 1.0 mT); irradiation of bilirubin in vacuum was not associated with generation of free radicals. Correlation analysis revealed statistically significant correlation between EPR signal intensity of bilirubin free radical in gallstones, activity of ceruloplazmin/F3+-transferrin antioxidant system and content of free Mn 2+ ions in patients' blood (r=-0.5725, p=0.0324; r=0.7805, p=0.0010, respectively). The tight correlation between marker of oxidative stress, Mn 2+ ions EPR signal intensity and HDL-, LDL-cholesterol content in blood (r = 0.910629, p = 0.0324; r = 0.029477, p = 0.0010, respectively) was also revealed. Conclusion: The results of the present study demonstrated the pathophysiological significance of alterations of blood redox-homeostasis during menopause in the gallstone formation. The risk of bilirubin oxidation, which plays a crucial role in the gallstones formation, increases with intensification of oxidative stress. Such systems have anomalous temporal and spacial response (non-local interactions and long memory) compared to systems without disorder. The systems having no memory can be modeled by linear differential equations with constant coefficients (exponential relaxation); the differential equations governing the systems with memory are known as Fractional Order Differential Equations (FODE). The relaxation of the spin system is best described phenomenologically by so-called Bloch's equations, which detail the rate of change of the magnetization M of the spin system. The Ordinary Order Bloch's Equations (OOBE) are a set of macroscopic differential equations of the first order describing the magnetization behavior under influence of static, varying magnetic fields and relaxation. It is assumed that spins relax along the z axis and in the x-y plane at different rates, designated as R1 and R2 (R1=1/T1,R2=1/T2) respectively, but following first order kinetics. To consider heterogeneity, complex structure, and memory effects in the relaxation process the Ordinary Order Bloch's Equations were generalized to Fractional Order Bloch's Equations (FOBE) through extension of the time derivative to fractional (non-integer) order. To investigate systematically the influence of “fractionality” (power order of derivative) on the dynamics of the spin system a general approach was proposed. The OOBE and FOBE were successively solved using analytical (Laplace transform), semi-analytical (ADM - Adomian Decomposition Method) and numerical methods (Grunwald- Letnikov method for FOBE). Solutions of both OOBE and FOBE systems of equations were obtained for various sets of experimental parameters used in spin !! NMR and EPR spectroscopies. Statistics disorders are a common gynaecological disorder occurring in women. The condition afflicts around 15% of women to the extent of impairing the quality of living. According to scientific reports as many as 50% of women experiencing problems related to genital statistics disorders. The aim of this work was to investigate tissue taken from women with genital statistics disorders using the Electron Paramagnetic Resonance method. The studies on the tissue of women is one of the first studies in this area. Kostrzewska","doi":"10.2478/v10214-012-0001-1","DOIUrl":"https://doi.org/10.2478/v10214-012-0001-1","url":null,"abstract":"Abstract To evaluate the involvement K+ channels in the relaxation induced by exogenous nitric oxide, after preincubation with L-arginine analogue L-NA in the human nonpregnant myometrium. The activity of myometrial strips, obtained from 60 premenopausal hysterectomised women, mounted in an organ bath was recorded under isometric conditions using force transducers with digital output. Concentration-response curves to DEA/NO after inhibition of endogenous NO were constructed in the absence and presence of soluble granulate cyclase and K+ channels’ blockers. The responses were quantified by calculating the area under the curve, the amplitude and frequency of the contractions. The inhibition of NOS results in slight but significant attenuation of the myometrium strips response to DEA/NO. Pre-treatment with both sGC inhibitors after preincubation with L-NA did not counteract the DEA-NOinduced relaxation of the spontaneous contractions of the myometrial strips. Application of blockers of different types of K+ channels to the myometrial strips significantly attenuated relaxing effect of cumulative DEA/NO administration in all cases. Dziliński","doi":"10.2478/v10214-012-0002-0","DOIUrl":"https://doi.org/10.2478/v10214-012-0002-0","url":null,"abstract":"Abstract Mesoporous silica SBA-15 functionalized by (1,4,8,11-tetraazacyclotetradecane) cyclam groups containing nickel ions (Ni-cyclam) was synthesized by two different approaches, and investigated by resonance Raman spectroscopy. Vibrational features of organometallic moleculess are analyzed for (Ni-cyclam) groups grafted in the silica pores. An assignment of bands in resonance Raman spectra was done to monitor the structure and properties of the mesoporous silica material with regard to the methods of synthesis used in this study. It was shown, that Raman scattering can be useful for probing of functionalization's efficiency of mesoporous silica. On the base of the resonance investigation: Raman and EPR spectroscopy, distribution of the functional groups inside pores can be determined. In the present article the Raman spectroscopy is treat as a complementary research to EPR investigation. It was shown that a clustering of the active groups alter significantly the resonance Raman spectra through broadening and shifts of the corresponding bands in comparison with separated molecules. Results obtained from the analysis of the resonance Raman spectra indicate significant differences between the samples prepared by the two procedures. On the basis of these measurements and three models of viscosity for glass-forming liquids: Avramov’s model, free-volume model and power-law model, the activation energy of viscous flow for solutions and ovalbumin molecules, at different temperatures, was calculated. The obtained results show that activation energy monotonically decreases with increasing temperature both for solutions and ovalbumin molecules. The influence of the energy of translational heat motion, protein-protein and protein-solvent interactions, flexibility and hydrodynamic radius of ovalbumin on the rate of decrease in activation energy with temperature has been discussed. One of the parameters in the Avramov’s equation is the glass transition temperature Tg. It turns out that the Tg of ovalbumin solutions increases with increasing concentration. To obtain the glass transition temperature of the dry ovalbumin, a modified Gordon-Taylor equation is used. Those systems may be considered as simple models for transfer of nanoparticles through the biological membrane. Three different surfactants were used: negatively charged sodium dodecyl sulphite, SDS, neutral Triton X-100 and positively charged tetredodecyltrimethyl ammonium bromide, TTABr. We have found that hydrophobic functionalized Ag nanoparticles are encapsulated in neutral Triton X-100 micelles with a diameter of 10 nm without significant change in the size of the micelles. The efficiency of encapsulation of Ag by SDS micelles is lower compared to Triron X-100 and no incorporation of Ag nanoparticles into TTABr occurs.
