{"title":"ANALISIS FILOGENI KURA-KURA BATOK (Cuora Amboinensis) WILAYAH INDONESIA TIMUR (AMBON, LUWU, DAN GORONTALO) BERBASIS SEKUEN GEN CYTOCHROME B","authors":"Candra Hermawan","doi":"10.36526/biosense.v6i01.2602","DOIUrl":"https://doi.org/10.36526/biosense.v6i01.2602","url":null,"abstract":"Penelitian ini dilaksanakan dengan tujuan untuk mengeksplorasi hubungan kekerabatan Kura-Kura Batok (Cuora amboinensis) wilayah Ambon, Luwu, dan Gorontalo dilihat dari analisis konstruksi pohon filogeni, jarak genetik, dan similaritas berdasarkan variasi sekuens gen Cytochrome b. Penelitian ini masuk ke dalam penelitian deskriptif eksploratif yang dilakukan di dalam laboratorium. Tahapan dalam penelitian ini meliputi isolasi DNA jaringan, purifikasi DNA, Poly Chain Reaction (PCR), elektroforesis horizontal, sekuensing gen, dan analisis filogeni menggunakan software MEGA 6. Sampel dalam penelitian ini merupakan 8 spesies yang berasal dari Ambon, Gorontalo, dan Luwu. Hasil analisis filogeni menunjukkan nilai jarak genetik sebesar 0,003-0,349 dan nilai similaritas genetik sebesar 65,13%-99,70%. Cuora amboinensis dari daerah yang sama memiliki hubungan kekerabatan lebih tinggi dibanding dari daerah yang berbeda. Terdapat anomali pada sampel Gorontalo 10 yang berkerabat dekat dengan wilayah Ambon padahal kedua sampel tersebut terletak pada pulau yang berbeda. Tujuan dari penelitian ini adalah untuk mengetahui apa saja jenis, cara pengolahan dan pemanfaatan tumbuhan obat di Desa Tangsi Lama. Penelitian ini dilaksanakan pada bulan Maret – April 2022. Penelitian ini menggunakan metode survei eksploratif melalui wawancara dan observasi langsung pada saat survei dilakukan. Teknik wawancara semi struktural sesuai dengan daftar pertanyaan yang telah dibuat antara lain: identitas nara sumber, nama lokal tumbuhan, bagian tumbuhan yang digunakan, kegunaannya, cara pemanfaatannya, dalam penelitian ini 20 jenis tumbuhan didapatkan. tumbuhan liar yang dimanfaatkan sebagai tanaman obat bagi masyarakat setempat. Pantai yang datar biasanya dapat ditumbuhi oleh mangrove. Sifat kompleks merupakan sifat yang dimiliki oleh hutan mangrove. Ada berbagai macam biota laut yang menempati hutan mangrove Bangladesh, contohnya adalah ikan sirip dan zooplankton. Berdasarkan analisis studi literature diketahui bahwa hutan mangrove Bangladesh belum dikelola dengan baik. Terdapat tumpahan minyak yang cukup banyak di pesisir laut. Tujuan dari penulisan jurnal ini adalah untuk menganalisis secara kritis terkait pemanfaatan ekosistem hutan mangrove sebagai habitat untuk biota laut. Penulisan artikel ini didasarkan pada jenis peneliitian kualitatif dengan menggunakan pendekatan kajian pustaka. Kajian pustakan merupakan kegiatan mengkaji berbagai macam bentuk sumber-sumber yang relevan. Kesimpulan memaparkan bahwa hutan mangrove memiliki berbagai macam manfaat bagi biota laut. However, the difference of phytochemical profiles in purple sweet potatoes among geographical regions has not been fully explored. The objective of this research to find out antibacterial activity of ethanol extract leaves purple sweet potato against bacteria Salmonella sp. and Pseudomonas aeruginosa. This study used Broth microdilution with concentration of 2 mg/mL, 4μg/mL, 8 mg/mL, 16 mg/mL, 32 mg/mL, 64 mg/mL, 128 mg/mL, 256 mg/mL, 512 ug/mL,1024 mg/mL. Chloramphenicol used as a positive control at the same concentration, minimum inhibitory concentration (MIC) determination made by the 50 seen from the transmittance value the test bacteria and minimum bactericidal concentration (MBC) by the determination of the lowest concentration in which no visible bacterial growth. Based on the test microbroth dilution leaves purple sweet potatoes had antibacterial activity against Salmonella sp. and Pseudomonas aeruginosa with MIC 64 mg/mL in Salmonella sp. and 128 ug/mL Pseudomonas aeruginosa. Tumbuhan berbiji (Spermatophyta) merupakan golongan tumbuhan yang mempunyai keanekaragaman jenis tinggi yang dapat ditemukan dan diamati secara langsung dari lingkungan sekitar salah satunya pada pekarangan rumah. Spermatophyta merupakan kelompok tumbuhan yang memiliki ciri utama yaitu terdapat organ berupa biji. Penelitian ini bertujuan untuk mengetahui jenis-jenis Spermatophyta yang terdapat pada pekarangan rumah warga di Kelurahan Lapajung, Kecamatan Lalabata, Kabupaten Soppeng. Penelitian ini termasuk penelitian deksriptif dan eksploratif, yang dilakukan untuk mengidentifikasi, mengklasifikasi, dan mendeksripsikan data yang diperoleh secara sistematis, faktual, dan akurat tentang fakta dan ciri-ciri Spermatophyta yang terdapat pada pekarangan rumah di Kelurahan Lapajung. Thalassia hemprichii is the most dominating species with a frequency to be found about 96,3% while the frequency of Enhalus acoroides is 3,7%. Seagrass has ± 14 hektar distribution area with the pattern of distribution in group. The coverage of seagrass in Karang tirta's coastal included into poor cathegory. Meanwhile, the seagrass bed is one of the important ecosystem in the coastal area that are habitat for some other species and carbon sinc. The application of organic fertilizer not only reduces the use of inorganic fertilizers but also reduces the accumulation of waste and increases the quality of crop yields at the right dose and type. This study utilizes waste substrate from straw mushroom growing media as a mixture plant medium for Tobacco. The fertilizer used is made from liquid coffee waste, cow urine, goat urine, and Kipahit plants. The purpose of this study was to determine the pH, NPK content, C-organic liquid organic fertilizer, determine the effect of the application of liquid organic fertilizer on the growth of Tobacco, and the content of dissolved protein, carbohydrates, and total chlorophyll in Tobacco. The results of laboratory analysis showed the pH value of liquid organic fertilizer meets the Minimum Technical requirements of Organic Fertilizer, Biological Fertilizer and Soil Improver, but the levels of NPK and C-organic do not meet the requirements. Application of liquid organic fertilizer showed a very significant effect on tobacco plant height. Ukuran mikroplastik yang kecil semakin meningkatkan potensi bahaya mikroplastik apabila masuk ke dalam tubuh organisme akuatik. Penelitian ini bertujuan untuk menganalisis kontaminasi mikroplastik di perairan Kabupaten Jember, dengan medeteksi kelimpahan mikroplastik di air dan gastropoda. Hasil penelitian ini diperlukan untuk memberikan informasi terkait kondisi eksisting kontaminasi mikroplastik di Kabupaten Jember sebagai upaya mitigasi pencemaran plastik di Kabupaten Jember. Penelitian ini dilaksanakan pada bulan Oktober - Desember 2021. Lokasi penelitian ini adalah Segmen Sungai Bedadung di Kecamatan Kaliwates Kabupaten Jember. Metode Penelitian meliputi : 1). Persiapan, berupa penentuan lokasi penelitian; 2). Pengambilan Sampel, berupa pengambilan sampel air dan gastropoda; 3). Identifikasi Mikroplastik, meliputi destruksi sampel dan identifikasi dibawah mikroskop stereo; 5). Analisis Data, berupa penghitungan kelimpahan mikroplastik pada masing-masing sampel. Hasil penelitian menunjukkan bahwa mikroplastik ditemukan pada keseluruhan sampel, dengan rerata kelimpahan, berturur-turut air dan gastropoda sebesar 1,87 partikel/liter, 114,3 partikel/individu. Hasil ini membuktikan terjadi perpindahan polutan mikroplastik dari media air ke dalam tubuh organisme. Hasil ini juga mengindikasikan bioavaliabilitas mikroplastik di dalam tubuh organisme sehingga berpotensi terjadinya biomagnifikasi mikroplastik di perairan dan meningkatkan efek bahaya bagi top predator di perairan Sungai Bedadung Kabupaten Jember. Coffee plants are a high foreign exchange earner in the Indonesian economy. Data from the Central Statistics Agency (BPS) show that Indonesia's coffee production in 2021 was the highest in the last decade, reaching 774.6 tonnes. This number increased by 2.75% from the previous year of 753.9 thousand tonnes. Coffee seed germination takes a long time due to seed dormancy. Coffee growth can be optimal with the provision of growth regulators. Good planting media can produce quality coffee seeds, because in good media there are nutrients needed by plants. The research was conducted at the Jember State Polytechnic Soil Wire House Laboratory in February-May 2022. Data were analyzed using Factorial RAK with a 5% BNJ follow-up test.
