However, little information is available on the KNOX gene family in wheat ( Triticum aestivum L.). In this study, 36 KNOX genes with KNOX1 or KNOX2 domain distributed on 18 chromosomes were identified from wheat genome using bioinformatics methods. The phylogenetic evolution, gene structure, protein domains, cis-acting elements and gene expression patterns were analyzed in the present study. Based on the phylogenetic tree, the 36 TaKNOX genes were divided into two major subclasses, Class I and Class II, and the two subclasses were further divided into five evolutionary branches. Most TaKNOX genes contain four typical conserved protein domains: KNOX1, KNOX2, ELK and HOX. Some cis-acting elements are associated with hormonal, plant development and stress in TaKNOX promoters. The analysis result of transcriptome data from wheat different tissue showed that Class I KNOX genes had obvious tissue specificity, while Class II KNOX genes expressed widely in different wheat tissues. And the low temperature of 8°C was significantly reduced the germination potential. With the decrease of germination temperature, the growth of roots and buds were inhibited significantly. There were significant differences both in temperatures and varieties. The germination potential, root length and bud length could be used as the main index to evaluate seed germination of rapeseed under low temperature. In addition, to screen varieties suitable for spring sowing as green manure, the spring sowing test was carried out. The study found that the fresh grass yield of “Huyouzao No.1” was 1712.53 kg/m2, which was much higher than other 7 varieties. Its relative root length was second only to “Huyou17” and its relative bud length next to “Huyoufei No.1” at 8°C, respectively. The results showed that the average number of allelic loci of 12 pairs of primers used in 21 walnut species samples was 7.916 7. The mean number of alleles, Shannon Wiener index (I) and polymorphic information content (PIC) per locus were 4.3516, 1.643 6 and 0.705 5. Twelve pairs of polymorphism information content (PIC) were higher than 0.25, indicating that the 12 pairs of primers had good polymorphism in the samples. As one of its main members, mitogen-activated protein kinase kinase (MAPKK) is located in the middle of the cascade reaction and plays a key role in signal collection and divergence. There were shown that potato ( Solanum tuberosum L.) StMAPKK1 (PGSC0003DMT400000744) gene responded to drought stress. Therefore, StMAPKK1 gene was firstly selected to screen its interacting protein. In this study, the bait vector pGBKT7-StMAPKK1 was constructed by homologous recombination and used to screen potato cDNA library by yeast ( Saccharomyces cerevisiae ) two-hybrid system. Five StMAPKK1-interacting proteins, hydrolase (hydrolyzing O-glycosyl compound), RING-H2 subgroup RHE protein, cyanate hydratase, ARF GTPase activator, and a C2 domain-containing protein, were obtained through this screening and were identificated by bioinformatics analysis, and the interaction was verified by small-scale hybridization verification. Fan, Lianghuan Qu, Li-Ming Deng, Mei Li","doi":"10.5376/MPB.2021.12.0017","DOIUrl":"https://doi.org/10.5376/MPB.2021.12.0017","url":null,"abstract":"The Polima system of cytoplasmic male sterility (Pol CMS) was widely used for hybrid seed production, but with high probability of occurrence of maternal false hybrid due to the fertility of Polima CMS (Pol CMS) line greatly affected by the ambient temperature, therefore, the detection of maternal false hybrids is the key to purity identification. In present study, the SSR markers were designed based on the flanking sequences of Pol CMS restorer gene Rfp , and application of SSR markers in hybrid seed purity test of rapeseed. The main results were as follows: the sequences of Rfp were subjected to basic local alignment search tool queries against the Brassica rapa genome to determine chromosome positions, and 32 SSR markers were developed based on the upstream and downstream sequences of Rfp . Among the 32 SSR markers, one codominant marker was polymorphic between Pol CMS sterile and restorer line, the PCR product length was 162 bp in Pol CMS sterile lines, and 184 bp in restorer lines. Based on the PCR sequence, two insert fragments (15 bp and 7 bp) were identified in Pol CMS restorer lines. The SSR marker co-segregated with plant fertility of individuals in the “fengyou737” F 2 population. The purity of “fengyou737”, “fengyou320” and “fengyou958” were identified by means of PCR-SSR markers and field-planting, and the results of two ways were highly consistent, indicated that the codominant SSR marker tightly linked to Rfp , and the marker can detect paternal and maternal false hybrids.
