Kirliligin genotoksik etkileri, kas, solungac ve karaciger dokularinda antioksidan savunma sinyali (GS, Cu-Zn SOD, CAT), isi sok proteinleri (HSP26, HSP60, HSP70 ve HSP83) ve QRT-PCR testi ile DNA onarimi (SMUG1) metotlari kullanilarak incelenmistir. Ayrica, RAPD analizi kullanilarak balik dokularindaki DNA hasarlari belirlenmistir. Bant degisimleri, ozellikle tum baliklarin solungac dokularinda acikca tespit edilmistir. Ayrica, C. gibellio, S. erythrophthalmus ve S. lucioperca'nin kas dokusunda CuZn-SOD'un nispi ekspresyonunda onemli bir artis gosterdigi tespit edilmistir. CAT ekspresyonu tum balik dokularinda onemli olcude degisiklik sergilemistir. Bununla birlikte, CAT 128,9, 102,1 ve 138,4 fg/ug total RNA'nin ekspresyon seviyesindeki en yuksek artis, S. lucioperca'nin kas, karaciger ve solungac dokularinda belirlenmistir. HSP60 ve HSP70 cevre kirliliginden en az etkilenirken, S. lucioperca'nin karaciger dokusunda en buyuk DNA onarim aktivitesi acik sekilde gozlenmistir. S. obliquus suslari, Ankara (Turkiye) ilindeki cesitli tatli su kuyulari ve havuzlarindan alinan orneklerden izole edilmistir. Izole edilen uc S. obliquus susunun uretiminde, dort kultur besi ortami ile dort dogal maden suyu kullanilmistir. Kulturlerin hucre yogunlugu, kuru agirlik miktari, spesifik buyume oranlari, ikilenme sureleri ve klorofil-a miktarlari hesaplanmistir. Sonuclar, farkli kultur besi ortamlari ve dogal mineralli sularin, son hucre yogunlugunu, biyokutleyi ve suslarin buyume oranlarini onemli olcude etkiledigini gostermektedir. Bu uc sus icerisinde, ScnGP susunda digerlerine oranla hucre yogunlugu, biyokutle agirligi ve spesifik buyume hizi acisindan anlamli bir fark bulunmustur. Hucre yogunlugu (7,4x104±1,3x103 hucre/mL), biyokutle miktari (0,212±0,032 g/mL), spesifik buyume orani (0,024 h-1) ve klorofil-a (1,71±0,22 μg/mL) miktari Bold Wynne besi ortaminda yetisen ScnGP susunda diger uygulamalara gore onemli olcude (P<0,05) daha yuksek tespit edilmistir. Dogal mineralli sulardan elde edilen sonuclarin kultur besi ortamlarindan elde edilen sonuclar kadar iyi oldugu gozlenmistir. Ex Steud, Typha latifolia L., Nasturdium officinale L., Lemna minor L., Lythrum salicaria ve Ceratophyllum demersum L.) bor, cinko ve arsenic miktarlari ile su ve sedimentteki bor, cinko, arsenik, gumus, kursun, bakir, kadmiyum ve krom konsantrasyonlari tarafimizdan arastirilmistir. Bu calismada arastirilan sekiz agir metal arasindan, arsenik sedimentte yuksek konsantrasyonda bulunurken (288,66 mg/kg), su numunelerinde en yuksek konsantrasyonda bor (903 µg/L) tespit edilmistir. Ayrica, bitkideki cinko, bor ve arsenik konsantrasyonlarinin sirasiyla 70-280, 37,6-1682,5 ve 0,2-34 mg/kg kuru agirlik arasinda degistigi saptanmistir. For this purpose, surface water and sediment samples were collected monthly, from five stations between June 2013 and May 2014. A bacteriological assessment of water and sediment samples with a 22 °C to 37 °C temperature range, including total coliform bacteria (TC), fecal coliform bacteria (FC), fecal streptococcus (FS), and Escherichia coli were done. Analysis was performed according to standard methods. Water quality and pollution level in Yaglidere Stream were assessed in accordance with both Turkish legislation and international criteria. The FC, FS and E. coli count were detected higher than the reference value at multiple stations in almost every month of the year indicates. Arastirma, 3 tekerrurlu deneme gruplari uzerinden 330 dakika boyunca yurutulmustur. Deneme suresi boyunca, duzenli araliklarla su sicakligi, pH, cozunmus oksijen ve amonyum degerleri olculmus, TAN (Toplam Amonyum Azotu) ve adsorpsiyon verimi ilgili literature bagli olarak hesaplanmistir. Calisma degerleri artan temas suresi ile tum gruplarda TAN konsantrasyonunun azaldigini gostermistir. Zeolitin farkli partikul buyuklugu ve adsorbant miktari icin en iyi verim (% 44,1) 15 g/l ve 0,075 mm zeolit grubunda belirlenmistir (P˂0,05). Protein duzeyleri arttikca yem tuketimi, yemden yararlanma orani ve protein etkinlik orani dogrusal olarak dusmustur. Tum vucut kuru madde, kul ve lipit konsantrasyonlari diyetsel protein duzeyinin artisi ile dogrusal olarak dusmus, vucut protein duzeyi ise kuadratik olarak etkilenmistir. Diyetsel protein gereksinimlerini tahmin etmek icin, ikinci derece regresyon ve iki kirikli linear model ( IKLM ) kullanilmistir. 4. hafta ve deneme sonu SGR degerleri kullanildiginda IKLM daha dusuk kalinti kareler toplami vermistir. 4. hafta ve final SGR oranlarina gore, IKLM minimum protein gereksinimlerini sirasiyla, % 27,69 ve % 28,93 olarak tahmin etmistir. Kantitatif Real-time PCR teknigi ile tum bireylerde B. dendrobatidis enfeksiyonlarinin prevalansi ve yogunlugu ile arastirildi ve Dogu Karadeniz Bolgesi’ndeki 10 amfibi turunden toplanan 62 bireyin 13’ unde B. dendrobatidis enfeksiyonu varligi ilk kez rapor edildi. Enfeksiyon yogunlugu 403,520-534,340 genomik esdegerler arasinda degistigi belirlendi.
