In contrast to most hypoglycemic drugs, GLP-1 receptor agonists not only do not cause weight gain, but can even lead to weight reduction. This is why they are recently also increasingly used in the treatment of obesity in patients without type 2 diabetes. Current research indicate that liraglutide and semaglutide are effective and safe methods of treatment for overweight and obesity in patients without diabetes. GLP-1 receptor agonists’ mechanism of action is related to the incretin system. They lowers glucose concentrations by increasing insulin secretion and suppressing glucagon release. Moreover, they cause a retardation of gastric emptying and affect the central nervous system, resulting in a decrease in appetite and stimulation of satiety. In addition, a rising number of research studies show that GLP-1 receptor agonists may also have beneficial effects in the treatment of other conditions, such as nervous and cardiovascular diseases, metabolic diseases or endocrine disorders. Due to the new findings, even more research should be carried out to further investigate and describe the multidirectional effects of these drugs.

The developed method has been optimized and validated. Optimum separation of Mn2+, by measuring at 214 nm, was obtained on a 30 cm × 75 μm capillary using a 50 mM phosphate running buffer (pH 3.0), capillary temperature 25°C, voltage 10 kV, and hydrodynamic injection. Good results were obtained for different aspects including stability of the solutions, linearity, and precision. Detection and quantification limits for Mn2+ were reached at the level of 0.97 and 2.94 μg/mL respectively. The sample preparation procedure is very simple and not time consuming. This method has been successfully used to the measuring concentration of Mn2+ in order to determine the content of this ion in parsley leaves and roots. The speed of obtaining the result of the analysis was only 3 minutes.

We report a rare case of a 37-year-old male patient, undergoing testosterone treatment in the course of Klinefelter syndrome. He presented with the general symptoms of fever, weakness and diarrhea lasting for 2 weeks. He also had incorrect results of laboratory studies. The fourth generation ELISA HIV test indicated negative outcomes. The HIV real time polymerase chain reaction test was performed and showed very high plasma viral load, over 107 copies/ml. We discuss the connection between androgen replacement therapy used in Klinefelter syndrome and the diagnosis of PHI. This case report illustrates the importance of obtaining a detailed medical history, especially of chronic diseases and medications, and applying appropriate diagnostic tests. ORMP is also involved in approving and controlling clinical trials and in safety of pharmacotherapy (pharmacovigilance) by collecting and analysing reports of adverse drug reactions (ADR). The aim of the study was to review the activity of ORMP in the field of 3 basic competences of this authority: clinical trials, registration of medicinal products and pharmacovigilance in the last 5 years. The evaluation was carried out by the method of document analysis on the basis of data contained in annual bulletins of medicinal products for the years 2017-2021 published by ORMP. In 2017-2021, an increasing trend of clinical trials submitted to ORMP was observed, and they were mainly in phase II and III. Most of the medicinal products introduced to the Polish pharmaceutical market in 2017-2021 were drugs prescribed by a doctor, registered in a decentralized procedure, used in diseases of the nervous and cardiovascular systems and in the treatment of cancer. A relatively low activity of ORMP in monitoring the safety of pharmacotherapy was demonstrated, which was a consequence of low activity of medical staff in the spontaneous ADR reporting. The conducted analysis confirmed the key role of ORMP in surveillance of clinical trials and introducing new medicinal products to the pharmaceutical market in Poland but also indicated the need for increased activity in the area of pharmacovigilance.

Regulation of their function allows to mute the inflammatory response, which is a desired effect in the treatment of inflammatory diseases. The source of mediators is arachidonic acid, converted to pro-inflammatory mediators by cyclooxygenases (COX) or lipooxygenases (LOX). The 5-LOX pathway is full of target points for regulation of biosynthesis of cysteinyl leukotrienes (CysLT) – one of the major classes of inflammatory mediators. These compounds exert their activity through specific leukotriene receptors – first (CysLT1R) and second (CysLT2R). Drugs that inhibit CysLT synthesis as well as leukotriene receptors antagonists (LTRA) form group of drugs known as anti-leukotriene drugs. Currently, only a few representatives of this group are available in the pharmacies around the world: the 5-LOX inhibitor – zileuton – and three CysLT1R antagonists – montelukast, pranlukast, zafirlukast. LTRAs due to their wide range of anti-inflammatory effects, are a group of drugs with a high potential in the treatment of inflammatory diseases. The study of new applications of known LTRAs and the search for new members of the LTRA group are the main directions of development in this field of pharmacy. Its main and most threatening complication, ischemic stroke, is responsible for a great number of deaths and disability worldwide. This makes anticoagulation a priority in AF management. Non-vitamin K antagonist oral anticoagulants (NOAC) are currently the most effective and safe option for patients with nonvalvular atrial fibrillation. Rivaroxaban, apixaban, edoxaban and dabigatran are non-inferior to warfarin in terms of stroke and systemic embolism prevention. However, warfarin use is associated with higher rates of major bleedings than any NOAC. Different drugs and mechanism of action among NOACs may result in differences in terms of efficacy and safety between them. This review aims to assess these differences by summarizing data available in online databases and European Society of Cardiology guidelines. There are major studies comparing each of NOACs to warfarin, however there is only limited data comparing apixaban, rivaroxaban and dabigatran directly. Both direct and indirect comparisons suggest that apixaban, rivaroxaban and dabigatran use is associated with similar reduction of stroke and systemic embolism risk. In terms of safety, anticoagulation with apixaban results in lower major bleeding risk relative to dabigatran and rivaroxaban. \u0000

The mechanism of their action is based on inhibition of the reabsorption of glucose inside the proximal tubule of nephron, which results in lowering a blood glucose level. In addition, flozins reduce blood pressure, cause weight loss, and reduce the risk of death from cardiovascular causes. Until recently, the potential of flozins was used only to treat patients with type 2 diabetes. The clinical trials published several years ago on three representatives of this group of drugs: dapagliflozin, empagliflozin, and canagliflozin, showed that their use in patients with type 2 diabetes and cardiovascular disease reduces the risk of hospitalization and death from cardiovascular causes. These trials prompted scientists to extend research on the effectiveness of flozins in lowering the risk of complications of heart failure and cardiovascular diseases. Subsequent studies showed that regardless of whether a patient with cardiovascular disease has diabetes, the effectiveness of empagliflozin and dapagliflozin is comparable in reducing hospitalizations and cardiovascular deaths. These discoveries resulted in flozins appearing in the European Society of Cardiology guidelines as one of the first-line drugs in heart failure with reduced ejection fraction. In Poland, empagliflozin, and dapagliflozin obtained a new reimbursement indication on May 1, 2022 – chronic heart failure with reduced ejection fraction in patients meeting specific criteria. It’s a significant step in treating heart failure, which is still a substantial cause of death and hospitalizations in Poland, especially among the aging population. The aim of this review is to show the path that flozins have gone from being an antidiabetic drug to being the first-line drug in the treatment of heart failure and to draw the attention of readers to the new reimbursement indication of flozins in Poland. \u0000 \u0000 \u0000
