{"title":"Identification and Bioinformatics Analysis of β-Defensin Gene Family in Huangsha Soft-Shelled Turtle","authors":"廷雅 杨","doi":"10.12677/ojfr.2022.91001","DOIUrl":"https://doi.org/10.12677/ojfr.2022.91001","url":null,"abstract":"β-defensin is a small cationic antimicrobial peptide rich in cysteine, which plays an important role in innate immunity of the living organism. In this study, the β-defensin genes of Huangsha softshelled turtle (Hs-BDs) family were identified based on transcriptome database of Huangsha softshelled turtle and PCR validation, and the physicochemical properties of protein, protein structure and evolutionary relationship were also analyzed. The results showed that a total of 18 members of Hs-BDs family with complete coding regions were identified. The number of amino acid residues of these Hs-BDs proteins ranged from 59 to 94, the molecular weight ranged from 6759.13 to 10,731.32 Da, and the isoelectric point ranged from 8.82 to 10.45. The 18 members of Hs-BDs family were divided into four subgroups (Class I~IV) according to the phylogenetic tree analysis. Some Hs-BDs were randomly selected for protein structure analysis and the results showed that the secondary structures of Hs-BDs preprotein are mainly composed of α-helix, β-sheet and random coil. Six conserved cysteine residues were linked to form three intramolecular disulfide bonds in the form of Cys1-Cys5, Cys2-Cys4 and Cys3-Cys6, respectively. {"title":"Comparison of Different Extraction Methods for the Genome of Enterocytozoon hepatopenaei, EHP","authors":"俊利 魏","doi":"10.12677/ojfr.2023.101003","DOIUrl":"https://doi.org/10.12677/ojfr.2023.101003","url":null,"abstract":"In order to explore the most suitable extraction method of the Enterocytozoon hepatopenaei (EHP) genome, five methods were used to extract genomes of equal amounts of EHP, including traditional and improved phenol/chloroform extraction, enzymatic hydrolysis of chitin, marine animals DNA kit and plant genomic DNA kit. Results showed that the effect on the genomic concentration was in descending order of marine animals DNA kit (18.825 ng/µL), enzymatic hydrolysis of chitin (17.17 ng/µL), improved phenol/chloroform extraction (15.06 ng/µL), traditional phenol/chloroform extraction (10.8 ng/µL), plant genomic DNA kit (10.8 ng/µL); the effect on the genome integrity was in descending order of marine animals DNA kit, improved phenol/chloroform extraction, plant genomic DNA kit, traditional phenol/chloroform extraction, enzymatic hydrolysis of chitin. {"title":"The Effects of Temperature and Salinity on the Hatching of Fertilized Eggs of Diodon holocanthus","authors":"Cheng Yang, Yanfeng Yue, Shiming Peng, Lianjun Xia, Quanxin Gao","doi":"10.12677/ojfr.2019.63013","DOIUrl":"https://doi.org/10.12677/ojfr.2019.63013","url":null,"abstract":"The effects of temperature and salinity on the hatching of the fertilized eggs of Diodon holocanthus were studied. The temperature (A) was set at 21 ̊C, 24 ̊C, 27 ̊C, 30 ̊C and 33 ̊C. The salinity (B) was set as 5 gradients of 18, 22, 26, 30 and 34, and three repetitions were set for each group. Each has parallel 100 fertilized eggs. The results showed that the incubation time decreased gradually with the increase of temperature. When the temperature was between 24 ̊C and 33 ̊C, the incubation time gradually decreased with the increase of salinity, and the incubation time at 18 ̊C was significantly higher than that of other experimental groups. When the salinity was 26 34, the hatching rate gradually decreased with the increase of temperature, and the hatching rate at 24 ̊C was significantly higher than that in other experimental groups (P < 0.05). When the temperature was 24 ̊C 27 ̊C, the incubation rate was significantly higher than that of other experimental groups when the salinity was 30 ̊C. When the temperature is 30 ̊C 33 ̊C, the hatching rate decreases with the increase of salinity. The survival rate first increased and then decreased with the increase of temperature. When the temperature is 24 ̊C 27 ̊C, the survival rate first increases and then decreases with the increase of salinity, and the survival rate of the 30 groups is higher than that of other groups. At the temperature of 30 ̊C, there was no significant difference between the experimental groups, but at the temperature of 33 ̊C, the survival rate decreased significantly with the increase of salinity. In terms of single factor, temperature and salinity have extremely significant effects on hatching time, hatching rate and survival rate of the fertilized eggs of Diodon holocanthus (P < 0.01), and the interaction between salinity and temperature has extremely significant effects on hatching and survival rate of the fertilized eggs of Diodon holocanthus (P < 0.01).
