Acetylcholine induced concentration-dependent contractions of non-precontracted IMA rings with denuded endothelium (pEC50 = 5.80 +/- 0.04) and was without effect on arterial segments with intact endothelium. The muscarinic receptor antagonists atropine, pirenzepine, methoctramine and p-fluoro-hexahydro-sila-diphenidol (pFHHSiD) antagonized the response to acetylcholine. The constrained pA2 values were 10.14, 7.74, 7.34 and 10.5, respectively. It is concluded that acetylcholine induces concentration-dependent contractions of porcine internal mammary artery rings on basal tone and that this contractile effect is probably due to direct cholinergic stimulation of smooth muscle cells, maybe including activation of muscarinic M1 receptors.</p>","PeriodicalId":23904,"journal":{"name":"Zentralblatt fur Veterinarmedizin. {"title":"Effect of duodenal glucose infusion on blood glucose concentration in endotoxin-treated calves.","authors":"K Bieniek","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The effect of endotoxemia on the intestinal absorption of glucose was evaluated in nine experiments performed on seven 3- to 5-week-old calves fitted with a duodenal cannula. An intraduodenal glucose load trial (infusion of 2 g glucose/kg b.w. as a 10% aqueous solution through the cannula over 60 min) was conducted in a group of 5 calves three times during the 4-day period: 48 h before and at 2 and 24 h after i.v. injection of E. coli 0111:B4 endotoxin (LPS) at a dose of 0.1 microgram/kg b.w. Control calves were treated similarly but instead of glucose they were infused intraduodenally with deionised water at a volume of 20 ml/kg b.w. In trial with glucose load performed 48 h before LPS administration, blood glucose concentration increased during the absorptive phase from 4.32 +/- 0.32 mmol/l to 11.45 +/- 0.87 mmol/l at 60 min and then decreased to a minimum value of 3.16 +/- 0.51 mmol/l at 240 min. During the initial phase of endotoxemia, blood glucose concentration did not change from baseline values in both groups of calves. Glucose concentration in control calves started to decrease at 165 min reaching a minimum value of 1.39 +/- 0.17 mmol/l at 210 min and then increased to 2.44 +/- 0.11 mmol/l at 480 min after LPS administration. The intraduodenal infusion of glucose at 2 h after LPS administration resulted in an increase in blood glucose concentration during the absorptive phase only in one calf. Blood glucose concentration in this calf increased between 30 and 90 min reaching a maximum value of 7.19 mmol/l at 60 min, and then decreased to a minimal value of 0.94 mmol/l at 180 min after glucose load. In the remaining four calves in this group, blood glucose concentration ranged from 3.89 +/- 0.37 mmol/l to 4.48 +/- 0.45 mmol/l up to 120 min, and then steadily decreased to a minimal value of 2.41 +/- 0.41 mmol/l at 300 min. In trial with glucose load performed 24 h after LPS administration, the rate of entry of glucose into the circulation during the absorptive phase was similar to that observed in the trial performed 48 h before LPS administration. In conclusion, these results indicate that endotoxemia impairs the intestinal absorption of glucose in calves. The magnitude of the absorption disturbance may vary in individual calves, and the inhibitory effect of LPS on the intestinal glucose absorption lasts less than 24 h.</p>","PeriodicalId":23904,"journal":{"name":"Zentralblatt fur Veterinarmedizin. {"title":"Influence of diet on the occurrence of intraepithelial microabscesses and foreign bodies in the ruminal mucosa of reindeer calves (Rangifer tarandus tarandus).","authors":"T D Josefsen, T H Aagnes, S D Mathiesen","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The forestomach mucosa was examined for pathological lesions in 12 healthy free-ranging reindeer calves at different seasons, and in 32 reindeer calves fed lichen (n = 3), baled grass silage (n = 24) or pelleted feed (n = 5). No gross lesions were seen. Samples for histology were taken from reticulum, omasum and four sites in rumen. In histological sections a total of 182 intraepithelial microabscesses (IEMAs) and 16 foreign body lesions (FBLs) were observed in the ruminal mucosa, while 1 IEMA was found in omasum and no lesions in reticulum. The number of animals having 0, 1-10, 11-20 and > 20 IEMAs per four ruminal sections (one from each sample site) was 19, 19, 5 and 1, respectively. None of the free-ranging or lichen-fed animals had more than 1 IEMA per four ruminal sections, while 58% (14/24) of the animals fed silage and 40% (2/5) of the animals fed pelleted feed had 2 or more IEMAs per 4 ruminal sections. FBLs occurred more frequently in animals with high numbers of IEMAs, and it is suggested that both IEMAs and FBLs are caused by plant particles penetrating the ruminal epithelium. {"title":"[Endocrine function of the testicle in the Mexican crossbred ram at different times of the year and its opioidergic control during anoestrus].","authors":"V Fuentes, V Sánchez, H González, P Fuentes, A García, R Rosiles","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>This work was carried out with the objective of studying the endocrine function of the testes in the Mexican Crossbred Ram (Suffolk x Hampshire) during the different seasons of the year in a latitude of 19 degrees 13' N. Also studied was the opioidergic control of this gland during anoestrus. For this purpose 8 rams were used, from which 3 ml of jugular blood were obtained three times a week to study the seasonal fluctuations in plasma testosterone. The experiment was carried out from May 1993 until June 1994. The androgen was determined by radioimmunoassay. To study the opioidergic control of androgen secretion during seasonal anoestrous (May-June) four of the rams were selected at random and treated with 0.5 mg naloxone at 12 h intervals during 3 continuous weeks. It was observed that blood testosterone was lowest during long days, and concentration was highest during the breeding season. Chronic naloxone medication during anoestrus resulted in a significant increase in blood testosterone (P < 0.001), an effect that was evident after the first week of treatment. When naloxone administration was discontinued, blood testosterone levels returned to basal. With this experiment it was demonstrated that an opioid antagonist such as naloxone administered in low doses, is capable of altering plasma testosterone concentration in the anoestrus ram, giving further support to endogenous opioids as important modulators of reproduction in seasonal breeders such as sheep.</p>","PeriodicalId":23904,"journal":{"name":"Zentralblatt fur Veterinarmedizin. A total of 87 cats was included in the study, 30 of them that were clinically healthy served as the control group. The urine protein pattern of this group had, besides the band representing the market albumin, and additional broad band within the size of the marker transferrin. In some cases, weak bands were present within the range of the Tamm-Horsfall-protein and immunoglobulin G. Micromolecular protein bands were not demonstrable. The remaining 57 animals had a histologically proven nephropathy. Thirty-eight cats had elevated urea and/or creatinine values in the plasma (group 1), and 19 animals had values within the reference range (group 2). The urine protein pattern as evidenced by SDS-urine electrophoresis was altered in all cats with histologically proven nephropathy, and it is thus concluded that with this technique a nephropathy can be diagnosed very early and prior to changes of plasma urea and creatinine (group 2). Moreover, in most of the cases, the nephrological changes can be classified as glomerular or tubulo-interstitial (group 1 and group 2). Sensitivity of the cell lines to radiation and rHuIFN varied according to the histologic origin of the cells. In radiation experiments, one fraction of 400 cGy produced survival rates of between 76 and 25%. Sensitivity to IFN was higher in cell lines derived from round cell tumours compared to those derived from solid tumors. At doses of 100 and 1000 U/ml IFN alpha and IFN gamma, survival rates of between 70 and 80% were observed. In combined treatment experiments, increase in IFN or radiation dose produced higher cell kills. Increasing the number of fractions had the most pronounced effect. In the stud farm Lindenhof (Templin, Germany), however, these first 2 years are used for a special fitness training for the young animals: 2-3 times a week, a group of the yearlings is forced to run a distance of about 1700 m on a track at an average speed of up to 10 m/s. Until now, little was known about changes of blood parameters which may occur during such special exercise. This study therefore investigated the activity of selected serum enzymes (aspartate-amino-transferase (AST), alanine-amino-transferase (ALT), gamma-glutamyl-transferase (gamma-GT), lactic dehydrogenase (LDH), alkaline phosphatase (AP), creatine kinase (CK)) and the variations of hematocrit, hemoglobin, lactate, protein, and urea concentration before and after exercise. Except for activity of AP and CK and concentration of urea, all parameters showed a distinct increase after exercise. In particular, the rise in lactic-acid concentration with values up to 23.08 mmol/l was remarkable, however, none of the parameters reached a pathological level. It is therefore concluded that exercise for young trotters over a medium distance-even at high speed-does not cause any injury of myocardium, skeletal muscles or liver cells.</p>","PeriodicalId":23904,"journal":{"name":"Zentralblatt fur Veterinarmedizin. Local tissue reaction was examined on macroscopic and microscopic level. Other hens were injected with HSA in physiological sodium-chloride solution and served as controls. The immunization of hens without using FCA resulted in a short-term rise in immunoglobulin G (IgG) antibody titres, while the hens injected with HSA and FCA developed a long-term response (plateau-form of antibody titres). Local tissue reaction after injection of HSA in sodium-chloride solution was a mild and transient form of interstitial myositis. The immunization with HSA combined with FCA regularly resulted in a persisting granulomatous myositis. The piglets were divided into three groups. The antibody group received egg powder with specific antibodies to E. coli K88, K99, 987P, and rotavirus, while one control group was fed with egg powder without specific antibodies and a second control group received no egg powder at all. The piglets were fed ad libitum. The egg powder was offered in a 5% feed ration. Compared with the control groups, the piglets of the antibody group showed significant differences (P < 0.05, chi 2-test) in the parameters rate of diarrhea (17.2%) (antibody group) to 60.7% (control egg group) or 56.7% (control group without egg powder), severity of symptoms (5.2-39.3% or 26.7%) and frequency of additional treatments (8.6-55.7% or 51.7%). Because of the additional treatments, no difference could be found in body-weight gain. {"title":"[Estimation of the heritability of abomasal displacement in German black-pied cattle in Hesse].","authors":"T Geishauser, M Diederichs, R Beuing","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The degree of heritability of displacement of the abomasum was estimated in 30 Hessian herds breeding German Black Pied cattle in which displacement of the abomasum occurred more frequently than normal. Estimation was made by investigating the similarity between mothers and daughters. A total of 2626 pairs were used for this investigation, based on the absence or occurrence of abomasal displacement in the mothers and daughters. The fourpoint correlation coefficient was determined for each herd using a quadrant square. The arithmetic means were then calculated from the correlation coefficients. The number of herds examined was limited by assuming the correlation coefficient to be stable when it varied by less than 0.02 units during the investigation of more than 15 herds. Heritability was then calculated using the correlation coefficient. The correlation coefficient was stable after examining 15 herds; for the examination of the last six herds, it was 0.12 (0.117-0.124) units.
