{"title":"[A new ELISA micromethod for the determination of tetanus antibodies].","authors":"F Ambrosch, G Wiedermann, H Müller","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Due to its simplicity, accuracy and economy, the ELISA technique is a suitable method also for the determination of tetanus antibodies. By the method described, a titration curve is established from each serum to be investigated as well as from a standard serum. The horizontal distance between standard and unknown serum expressed in log2 titre steps is called titre difference. From this titre difference and the concentration of the standard serum the concentration of the unknown serum can be calculated. By means of an appropriate computer this evaluation can be done automatically. The variation coefficient with regard to concentration was 18.3%. A comparison with the toxin neutralisation test showed a correlation coefficient of 0.87. {"title":"[Degradation of glutamic acid and proline in Clostridium sordellii under cadaveric bacteriological inspection].","authors":"W Huckenbeck, T Daldrup","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Glutamic acid (GLU) is decarboxylated to gamma-aminobutyric acid (GABA) by Clostridium sordellii isolated from a decaying human brain. The dependence of this reaction on temperature, pH and substrate concentration has been established. The pH-optimum is in the range of 5.0 to 5.2. Near optimal pH the temperature optimum is greater than 28 degrees C. At higher pH-values (6 to 7) activity is relatively independent of temperature. The similarity to results in decaying human brains (10, 11, 12) shows a good correlation between postmortem bacterial flora and GLU-degradation. {"title":"[Effect of iron on Pasteurella multocida].","authors":"K D Flossmann, G Müller, P Heilmann, H Rosner","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Iron is an important factor for growth, virulence and immunogenicity of the species Pasteurella multocida. This has been demonstrated in numerous experiments with bacterial cultures in vitro and immunized and not immunized animals in vivo (mice, piglets, calves). Iron substrates or iron chelators affect in different manner the virulence of P. multocida in vivo, depending on chemical character of the given compounds, their dose, route and time of application, and also depending on the host. P. multocida has an up to time unknown iron transport system, which can acquire the essential iron from physiological substances, such as heme, ferritine, transferrine, lactoferrine etc. This conclusion results from in vitro experiments with growing cultures, with insertion of radioactive iron (Fe-59) from different sources, and with iron solubilization in neutral pH ranges. In the same way, the iron of iron dextran and low molecular iron compounds is available for P. multocida. Iron of unphysiological complexes, potassium ferrocyanide, and ferrocene is unavailable. On the other side such iron chelating agents as nitrilotriacetate, tirone, ferrocene, citrate, EDTA, and apotransferrine do not or only a little affect growth, and such chelators as alpha, alpha'-dipyridyle, phenanthroline and the microbial siderophores deferrioxamin B and enterobactin are inhibitory substances for multiplication of P. multocida. This substances also inhibit the insertion of Fe-59 into the bacterial cell. The conclusion is drawn that neither enterobactin nor deferrioxamine B as typical representatives of phenolate or hydroxamate siderophores take part in Fe-transport of P. multocida.</p>","PeriodicalId":23821,"journal":{"name":"Zentralblatt fur Bakteriologie, Mikrobiologie, und Hygiene. It could be demonstrated for the first time that alpha-aminobutyric acid is definitely formed from threonine by clostridia. The pH-optimum of this reaction is greater than 8, the temperature optimum is greater than 28 degrees C. {"title":"Immunohistochemical investigations of leptospira-infected golden hamsters in potency tests.","authors":"B Hein, H Freudenstein","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Golden hamsters taken from routine potency testing of leptospirosis vaccines were investigated for leptospirae after a challenge infection. Kidneys, livers, and spleens were examined immunohistochemically by the unlabelled antibody enzyme (PAP) method, and samples of these organs were additionally cultured for the evidence of living leptospirae. In non-vaccinated animals the recultivation method indicated leptospirae during the bacteraemic phase, beginning immediately after infection. Both methods resulted in positive reactions after organ manifestations. Intravascular evidence of leptospirae in the kidneys could be seen first on day 3 after infection. On day 7, there was intracellular and intratubular evidence as well, combined with tubulo-interstitial alterations. In vaccinated animals the bacteraemia after infection was reduced. {"title":"Survival of leptospires in commercial blood culture systems.","authors":"M Palmer, S A Waitkins, W Zochowski","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Laboratory diagnosis of leptospirosis depends on the serological findings and, if possible, the isolation of the organisms. During the period 1981-1982 there were 40 blood cultures submitted from serologically positive patients and a successful isolation rate of only 12% was attained. Correct media, method of collection of suspect blood and laboratory technique all contribute to the successful isolation of the organism. Because of the low isolation rate an investigation was undertaken of various blood culturing systems used in Great Britain. Results obtained showed that certain blood culture media were able to maintain the organism better and therefore increase the isolation rates of leptospira.
