Toprak altinda uzun-oval sekilli sogani ile genellikle 3-4 yapraklidir. Kallus kulturu kullanarak endemik turlerin in vitro uretimi, bu turlerin korunmasinda umut verici bir calisma haline gelmistir. Bu calismanin amaci, in vitro C. chalcedonicum uretimi icin verimli kallus protokolunun olusturulabilmesidir. Explantlarin sterilizasyonunda, 20 dk %0.25 (w/v) civa klorur (HgCl2) kullanilmistir. Civa klorure ilaveten, yuzey sterilizasyonunda 30 dk boyunca Tween 80, %6,5 NaCl ile birlikte kullanilmistir. Bu calismada, 19 farkli besiyeri kullanilmis olup primer kallus olusumu 2,4-D (2 mg L-1), 2IP (0,5 mg L-1), %3 sukroz ve %0,05 aktif karbon iceren Murashige & Skoog bazal besiyerinde elde edilmistir. Calismamiz, aktif karbon kullaniminin primer kallus olusumunda etkili oldugunu gostermistir. Bu calisma, C. chalcedonicum’un primer kallus olusumu icin ilk rapordur. Kriyo-koruma islemi biyoteknoloji, klinik calismalar ve hayvan veya bitki hucreleriyle ilgili bircok calismada cok onemli bir rol oynadigindan dolayi, kriyo-korumada kullanilan ajanlarin arastirilmasi son derece muhimdir. Kriyo- koruma ajanlari, hucresel metabolizma ve biyofizyoloji uzerindeki etkileri nedeniyle uzun sureli kriyo-korumanin ardindan hucresel canliligin korunmasini saglarlar. Mevcut calismada; hucre canlilik testi, koloni olusturma testi ve gercek zamanli polimeraz zincir reaksiyonu tekniklerinden yararlanilarak, borun kriyo-koruma uzerindeki etkisi, insan akciger kanser hucre hatti, A549 ve fare fibroblast hucre hatti, L929 kullanilarak arastirilmistir. Hucre dondurma ortamini 15 µg/ml bor ile desteklemenin hucre canliligi uzerine olumlu etki ettigi gozlemlenmistir. Ayrica, tekrar eden dondurma - cozme donguleri ve uzun sureli kriyo-koruma sonucunda, gen anlatim profilleri degisen A549 ve L929 hucre hatlarinin, bor takviyesi sonrasinda, programli hucre olumuyle alakali genlerinin anlatiminda azalma, hucre cogalmasi ile ilgili genlerinde de artis gozlemlenmistir. Enzimlerin yeniden kullanilabilirligi arastirildi. Enzimlerin immobilizasyon verimlilikleri yaklasik olarak % 50-55 arasindaydi. Liyofilize Bacillus proteazi ile yapilan calismalarda glutaraldehitle aktiflestirilmis PA6 nanolifler ve glutaraldehitle aktiflestirilmeyen PA6 nanoliflerde glutamik asit aminoasidi varliginda immobilizasyonun daha basarili oldugu saptanmistir. Glutaraldehit ile aktiflestirilmemis ve aktiflestirilmis yuzeylerde immobilize edilen liyofilize proteaz enziminin 4 kez kullanimi olmasina ragmen, en iyi islevsel stabilite 2 kez kullanim ile elde edilmistir. Saf PA6/glutamik asit nanoliflerinde iki tekrarli kullanim sonucu enzimin immobilizasyon verimi % 38 olarak bulunmustur. Glutaraldehitle aktiflestirilmis PA6 nanoliflerde de PA6/glutamik asit nanolif yuzeyleri iki tekrarli kullanim sonucu enzimin immobilizasyon verimi % 65 olarak bulunmustur. Nanoliflerin glutaraldehitle aktiflestirmesi sonucu enzim immobilizasyon verimi iki kat arttirilmistir. Ticari proteaz ile yapilan calismalarda ise glutaraldehitle aktiflestirilmemis nanolif yuzeylerde enzimin 6 kez kullanimi olmasina ragmen en islevsel stabilite 3 tekrarli kullanimda elde edilmistir. En basarili immobilizasyon verimi PA6 nanoliflerde % 58 olarak bulunmustur. Spike (S) glikoprotein SARS CoV-2 virionunun en distaki bilesenidir ve anjiyotensin donusturucu enzim 2 (ACE2) reseptoru araciligi ile virusun hucreye girisinde onemlidir. ACE2, vazokonstriktor anjiyotensin 2'yi vazodilator anjiyotensin 1-7'ye donusturerek insanda kan basincinin duzenlenmesinde onemli roller ustlenir. Bu calismada, Asya izolatlarindaki mutasyonlarin S glikoprotein yapisinda neden olabilecegi degisiklikler analiz edilmis ve ilac ve asi hedefleme calismalarina katkida bulunmak uzere modellenmistir. Genom, proteom ve mutasyon analizleri biyoinformatik araclari (MAFFT, MegaX, PSIPRED, MolProbity, PyMoL) kullanilarak yapildi. Protein modellemesi ProMod3 kullanilarak yapildi. S glikoproteinde 26 mutasyon tespit edilmistir. Bu mutasyonlarin S glikoproteininin genel topolojik ve konformasyonel yapisinda ortaya cikardigi degisiklikler, SARS CoV-2’nin virulans ozelliklerini etkileyebilir. Mutasyonlarin reseptor baglanma bolgesini (RBB) kapali formasyondan acik formasyon benzeri bir yapiya donusturdugu belirlenmistir. RBB'de mutasyondan sonra meydana gelen konformasyonel degisimin reseptor afinitesinde bir artisa neden olabilecegi dusunulmektedir. In the present study, it was shown that Candida yeasts in an aqueous solution accumulate single Cu(II) and Ni(II) cations. The effect of heavy metal ions on the specific growth rate of biomasses and the uptake of metal ions during the growth phase was investigated in a batch system. Bioaccumulation efficiency decreased with increasing metal ion concentrations at constant sucrose concentrations. Both the specific growth rate and the biomass concentration were more inhibited in the bioaccumulation media containing Ni(II) ions singly as compared with the bioaccumulation media containing Cu(II) ions singly. The maximum specific growth rate and the saturation constant of yeasts were examined with a double-reciprocal form of Monod equation. Metal uptake performance decreased from 81.68% to 46.28% with increasing Ni(II) concentration from 25 mg/L to 250 mg/L for Candida lipolytica. Candida biomasses may be an alternative way of removal of heavy metals from wastewaters and may constitute a sample to produce new biomass. Bu calismanin amaci da 1) kitosani nano-gumus (Ag) kaplamalar icin baglayici olarak kullanarak yeni nesil antibakteriyel apreler gelistirmek, 2) karides ve kerevitlerden uretilen kitosanin tekstil uretimi icin uygulanabilirligini belirlemek ve 3) cevre dostu tekstil uretimine katkida bulunmaktir. Calismada, karides ve kerevit atiklarindan uretilen kitosan, Ag nanopartikullerinin kumas yuzeylerine baglanmasinda yapiskan olarak kullanilmistir. Nano-Ag partikullerinin kitosan araciligiyla kumas yuzeylerine baglanma ozellikleri Fourier donusumlu kizilotesi spektroskopi (FTIR), Taramali elektron mikroskobu (SEM) ve Enerji dagilimli x isini (EDX) analizleri ile incelenmistir. Kumaslarin Escherichia coli ATCC 8739' ye karsi antibakteriyel aktiviteleri JIS L 1902-2015 standardina gore test edilmistir. Calismanin sonuclari kerevit ve karides kitosanlarinin renksiz bir film olusturdugunu ve Ag nanoparcaciklarini pamuklu kumas uzerine homojen bir sekilde kapladigini gostermistir. Kerevit kitosani ve karides kitosani ile kapli pamuklu kumaslarin antibakteriyel aktivite degerleri sirasiyla, 3,10 ve 5,74 olarak hesaplanirken, kerevit kitosani+nano-Ag ve karides kitosani+nano-Ag ile kaplanmis pamuklu kumaslarin antibakteriyel aktivite degerleri sirasiyla 5,37 ve 5,10 olarak bulundu. E. coli ATCC 8739' ye karsi iyi bir antibakteriyel aktivite sergileyen (% 99,99 azalma) kitosan+nano-Ag kaplamalar, tibbi tekstiller, bebek kiyafetleri ve ic camasirlari gibi giysilerin imalatinda kullanilabilir. Binder olarak kitosanin kullanilmasi, tekstil baskisinda, pigment boyamada, terbiye maddelerinde, kirletici desarjlarinda ve endustriyel kaynakli emisyonlarda kimyasallarin kullanimini azaltabilir. The aim of the present study was to determine the interaction of two commercial lavender ( Lavandula angustifolia Miller) essential oils in combination with ketoconazole against clinical and standard strains of the human pathogens Candida albicans ATCC 10231, C. krusei NRRL Y-7179 and C. glabrata ATCC 66032. The chemical compositions of the investigated essential oils were confirmed both by gas chromatography/flame ionization detector (GC/FID) and gas chromatography/mass spectrometry (GC/MS) methods. Anticandidal activities of the essential oils were screened in vitro by the microdilution method. The resulting interaction of combining lavender essential oils and ketoconazole was tested using the checkerboard method. The results showed that the interaction between essential oils and ketoconazole revealed additive and indifferent effects against the tested strains. Virusun neden oldugu sitopatik etkilere karsi %50 koruma saglamasi icin gerekli konsantrasyon EC 50 olarak tanimlanmis, CC 50 (%50 Sitotoksik Konsantrasyon)’nin EC 50 ’ye oranindan da secicilik indeksi (SI) belirlenmistir. Arastirma sonucunda, B. glandulosissima ’nin hem metanol (EC 50 = 12.45 µg/mL; SI = 24.84) hem de su ekstraktinin (EC 50 = 19.12 µg/mL; SI = 24.59) RSV’ye karsi; pozitif kontrol olarak kullanilan ribavirin (EC 50 = 3.25 µg/mL, SI = 34.89)’e gore onemli sayilabilecek oranda anti-RSV aktiviteye sahip oldugu tespit edilmistir. Sonuc olarak, B. glandulosissima ekstraktlarinin, RSV’ye karsi klinikte kullanilan ilaclara karsi bir alternatif olarak gelistirilebilmesi icin; daha ileri calismalara layik oldugunu soyleyebiliriz. Bu amacla; ekmeklik bugdayda ( Triticum aestivum L. cv. Adana 99) genetik varyasyonlar yaratmak icin embriyonik kalluslara 0-5 mM Sodyum Azid (NaN 3 ), 30 dakika boyunca in vitro ortamda uygulandi. Islem gormus ve gormemis kalluslar, somatik embriyo tesvik ortamina konularak somatik embriyo tesviki icin 3 ve 4 mM NaN 3 uygulamasi optimum mutasyon dozlari olarak tespit edildi.
