Many epiphytic lichens demonstrate high specificity to a habitat type and hygrophilous species are, as a rule, confined to close-to-natural forest complexes. Tolerance to desiccation stress and the rate of photosynthesis activation upon thallus hydration vary between species. Analyzes of chlorophyll fluorescence and photosynthesis efficiency have been widely applied to determine the viability of lichens. The aim of this study was to determine the activation photosynthesis rate upon hydration in epiphytic lichens exposed to short-term desiccation stress and to find potential links between their activation pattern and ecological properties. The results proved that even highly sensitive hygrophilous lichens, i.e., Cetrelia cetrarioides , Lobaria pulmonaria , Menegazzia terebrata , do not exhibit any delay in the restart of the photosynthesis process, compared to mesophytic or xerophytic ones. All examined lichens achieved nearly 100% of their maximum photosynthetic efficiency just one hour after they had been supplied with a relatively small quantity of water. Moreover, the increase in photosynthesis efficiency, measured at 20-minute intervals upon hydration, started from a relatively high level. In addition, the differences in the content of photosynthetic pigments and water holding capacity between species did not affect the general pattern of activation, which is comparable across various lichens. It can be concluded that healthy hygrophilous lichens do not require long hydration time to regain a high level of photosynthesis efficiency after a short rainless period. This fact supports the idea of applying chlorophyll fluorescence analysis in the field to assess vitality of lichens and the condition of their natural habitat.","PeriodicalId":224123,"journal":{"name":"Acta Biologica Cracoviensia s. In the present study, the impact of short-term exposure to strontium on the accumulation of the metal as well as the content of isoflavones in soybean sprouts was investigated. The seeds were germinated in hydroponics with 0, 1, 1.5, 2.5, 5.0, or 10.0 mM of Sr for 72 hours. The content of strontium was assessed using flame atomic absorption spectrometry and the amount of isoflavones was determined with high performance liquid chromatography. dose-dependent accumulation of Sr and a linear correlation between the Sr concentration in the growth medium and the content of the element in the plant samples were observed. The largest changes in the isoflavone content, compared to the control, were noted in soy sprouts germinated in the presence of 5 and 10 mM of strontium. daidzin, genistin, malonyldaidzin, and malonylgenistin were the dominant isoflavones and their content increased by approx. 28, 44, 34, and 47%, respectively, compared to the control. low amounts of aglycones were found; moreover, their content decreased by ca. 19–30%. Our research can be important for obtaining a natural product enhanced with strontium and isoflavones, which contribute to prevention of osteoporosis associated with endogenous oestrogen deficits.","PeriodicalId":224123,"journal":{"name":"Acta Biologica Cracoviensia s. It has been reported that calcium concentration in pistil papilla cells was increased after self-pollination in transformed self-incompatible Arabidopsis thaliana. In this study, we found that CML27 changed its expression level for both mRNA and protein when compared to transcriptome and proteome. At the same time, CML27 was expressed in the anther and pistil at a high level and reached up to 5-fold up-regulated expression in the pistil at 1 h post-pollination when compared to 0 min. In order to find out potential proteins that may interact with BoCML27, BoCML27 was expressed in and isolated from E. coli. After its co-incubation with Brassica oleracea pistil proteins, the products were separated on SDS-PAGE gels. We found a specific band at the position between 130–180 kDa. Through LC-MS-MS (Q-TOF) analysis, eight proteins were identified from the band. The proteins include 26S proteasome non-ATPase regulatory (26S), Phospholipase D, alpha 2 (PLDα2) involved in Ca2+ binding and Coatomer subunit alpha-2-like (Coatomer) involved in vesicle mediated transport. All of these identified proteins provide new insights for the self-incompatibility response in B. oleracea, specific for increasing Ca2+ concentration in pistil papilla cells.","PeriodicalId":224123,"journal":{"name":"Acta Biologica Cracoviensia s. The effect of sodium hydrogen sulfide as H2S agent was investigated in basil plants under cadmium stress. A completely randomized design with three different concentrations (0, 50 and 100 μM) of CdCl2 and two levels of NaHS (0 and 100 μM) was used in this study. Cadmium exposure reduced growth parameters and relative water content. Cd also caused a significant increase in ion leakage and higher oxidative stress in terms of lipid peroxidation and H2O2 production. Although exogenous NaHS used in non-stressed control plants negatively affected growth and physiological parameters, it improved the root/shoot length ratio and fresh weight in basil plants under Cd 50 μM exposure. Moreover, NaHS alleviated deleterious effects of cadmium on ion leakage, relative water content and photosynthetic pigments of leaves. The activity of antioxidant enzymes like catalase, peroxidase and ascorbate peroxidase were also enhanced by NaHS in plants under moderate cadmium stress. Our results show that NaHS 50 μM ameliorates growth retardation induced by cadmium 50 μM stress in basil plants, probably through regulating physiological parameters such as photosynthetic pigments content, relative water content and the activity of antioxidant enzymes.","PeriodicalId":224123,"journal":{"name":"Acta Biologica Cracoviensia s. This species has also been traditionally used as an edible plant in Eastern Europe because of its nutritional value. During the study, qualitative and quantitative sex-related differences of phenolic constituents in methanolic leaf extracts of R. thyrsiflorus were evaluated. The presence of the same substances (nine phenolic acids before, and six phenolic acids after acid hydrolysis, nine flavonoids, and a catechin) was estimated in both female and male specimens, using the HPLC-DAD method. A statistically significant higher content of eleven constituents in female plant extracts (acids: chlorogenic, p -coumaric, cryptochlorogenic, gallic, protocatechuic, neochlorogenic, vanillic; flavonoids: quercitrin, rhamnetin, rutoside; and a catechin) was shown. This is the first report concerning the relation between the sex and the content of biologically active phenolic secondary metabolites in leaf extracts of R. thyrsiflorus . Female plants of R. thyrsiflorus could be useful for pharmaceutical purposes as a preferential source of bioactive phenolic acids, flavonoids and especially catechin.","PeriodicalId":224123,"journal":{"name":"Acta Biologica Cracoviensia s.
