The role of CO and PEEP on 19 isolated rabbit lung preparations perfused with hypoxic mixture (6% CO2, 10% O2, and 84% N2), which resulted in a constant oxygen venous pressure (64 +/- 5.6 mmHg) has been studied. The first group of 11 preparations were used to study the influence of CO modifications with room air ventilation on the Qs/Qt when the CO rises in 48%; in the second group simultaneous modifications in CO and PEEP (0.5 and 10 cm H2O) were performed. A positive correlation (p < 0.01) in Qs/Qt (0.048 +/- 0.04 to 0.12933 +/- 0.09) was found when the CO increased in the first experimental group, the fluid filtration rate (FFR) also increased and the pulmonary vascular resistance (PVR) remained stable. In the second group an increase of 5 and 10 cm H2O of PEEP at constant CO reduced the Qs/Qt (0.0361 +/- 0.02 to 0.0184 +/- 0.006) while it increased the arterio-venous oxygen difference, PVR and FFR. During high CO conditions increase of 5 and 10 cm H2O of PEEP reduced the Qs/Qt (0.099 +/- 0.03 to 0.027 +/- 0.02) and FFR. No significant correlations were found between the arachidonic acid uptake activity and fasting plasma glucose, total cholesterol, HDL-cholesterol or triglyceride levels. The arachidonic acid uptake activity was significantly reduced by the presence of indomethacin in platelets obtained from both under and over-forty year old subjects, and by the presence of nordihydroguaiaretic acid in platelets obtained from over-forty year old subjects. In conclusion, these data show that arachidonate uptake activity by platelets increased with age. The separation of saturated lecithin, the main component of surfactant, is the basis for using the precipitation procedure but there is still a controversy as to whether cold acetone precipitable lecithin can be equated with saturated lecithin. Following up a previous paper in which the effect of cold acetone precipitation on phospholipids of amniotic fluid was studied, the present work reports that non-precipitated L/S and phosphatidylglycerol/sphingomyelin (PG/S) ratios correlate well with the precipitated L/S ratio (r = 0.93, r = 0.84, n = 92). Furthermore, the actions of VIP and pituitary adenylate cyclase activating peptide (PACAP) on the exocrine pancreatic secretion were compared at a similar molar dose. After the infusion of VIP a linear dose-response relationship for pancreatic flow rate and bicarbonate output, up to the dose of 4 micrograms/kg, was observed. VIP acts as a partial agonist of secretin, the rabbit pancreas being less sensitive to VIP compared with other mammals. Moreover, VIP did not significantly stimulate the pancreatic protein output. PACAP stimulated the hydroelectrolyte fraction of the exocrine pancreatic secretion in a similar manner to that of VIP. Unlike what was observed with VIP, PACAP, on the same molar basis, significantly stimulated the protein and amylase outputs. The VEP is defined by four components: P1, N1, P2 and N2, which appear during the 100 ms following the stimulus. This model is repeated for the majority of recording points although N1 and P2 do not appear to be homogeneous over the entire cortex. The variability of the VEPs recorded at the same point in different cats is lower than the one observed by means of stimulation with flashes. Beginning at 20 days of age, for a period of 130 days, control (C) and malnourished (M) animals were fed three different diets: standard diet (C, M), standard diet enriched with soybean oil (CS, MS) and standard diet enriched with olive oil (CO, MO). Body weight, systolic and diastolic blood pressure, total cholesterol, HDL-cholesterol and triglycerides levels of pups were measured at periods of 15, 30, 90 and 150 days of age. Malnutrition produced a decreased body weight and experimental diets were ineffective in body weight recovery. The controls, however, displayed overweight. Blood pressure values, taken at different ages and under different diets were within normal limits. Plasma triglycerides increased up to 30 days of age with early undernutrition returning, thereafter, to normal limits. The results indicated changes, mainly size-independent, in kidney carbohydrate metabolism during transfer of rainbow trout to seawater. An enhanced glycogenolysis and a concomitant increase in gluconeogenic enzyme activity were clearly observed in kidneys of both sizes of animals during transfer to seawater. They were divided into three equal groups: 1st placed under ordinary vivarium conditions (vivarium control animals); 2nd subjected to hypokinesia (unsupplemented hypokinetic animals, HK); and 3rd submitted again to HK and daily supplemented with water (5 ml/100 g b.w.) and NaCl at 0.9 g % (3 ml/100 g b.w.) orally administered (supplemented hypokinetic animals). The hypokinetic effect was carried out by keeping the rats in small individual wood cages which restricted all their movements without hindering feed and water intake. Determination was made of weight and volume of their entire bone, head and distal epiphysis, as well as density, ash and mineral content. Thickness of the cortical layer and width of the bone marrow canal were measured on frontal and lateral x-ray projections. Histological transverse sections of the femoral diaphysis were prepared from the femoral bone fragments. The concentrations of calcium, phosphorus and creatinine in serum were also measured. {"title":"Peroxisome enzyme modification and oxidative stress in rat by hypolipidemic and antiinflammatory drugs.","authors":"A Rivero,&nbsp;J I Monreal,&nbsp;M J Gil","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Changes in the activities of two peroxisomal enzymes (catalase and thiolase), some parameters related to oxidative situations, such as conjugated dienes, zinc, iron, copper and superoxide dismutase after the administration of two known peroxisome proliferators (clofibrate and acetylsalicylic acid), and two drugs pharmacologically related to the former (probucol and diflunisal) have been studied in male Wistar rats. Administration of the drugs was made by p.o. for 15 days. After the treatment the rats were killed, their livers and brains were taken out, and their blood was collected. Peroxisomes were purified by differential centrifugation followed by ultracentrifugation. Total RNA was also extracted and the acyl CoA oxidase mRNA expression was studied. Clofibrate was inactive on both enzymes studied in liver and diflunisal in brain. However, the acyl CoA oxidase mRNA expression increased by clofibrate treatment. Results are justified by the liposolubility and protein-binding properties of the drugs. Otherwise, the present results show the existence of an increased lipid peroxidation, lower value of superoxide dismutase, and variable results for zinc, copper and iron trace elements. Survival of young and aged fibroblasts was determined in the presence and absence of different concentrations of two vitamins. Vit C at doses of 5, 12.5, 25 and 50 mumol/L and water-soluble Vit E (Trolox) at 1, 5, 10 and 50 mg/L, were added 30 minutes before oxidative stress, consisting of exposure to 5 mM hydrogen peroxide for 30 minutes. A non-radioactive cell proliferation cytotoxicity assay (MTT) was used to determine the protective effect of the vitamins studied. Vit C produced a clear cytoprotective effect on aged cells over the entire range of doses applied.
