Polyclonal E2 (C902), and progesterone (C914) antibodies were raised in rabbits using 6-keto-17 beta-estradiol 6-carboxymethyloxime:BSA, and 11 alpha-hydroxyprogesterone 11 alpha-hemisuccinate:BSA. Horseradish peroxidase was used as label, conjugated to 17 beta-estradiol 3-hemisuccinate, and to progesterone 3-carboxymethyloxime. Standard dose response curves covered a range between 0 and 1 ng/well (100 microliters). The low detection limits of the technique were 1.99 pg/well for E2, and 13.21 pg/well for progesterone. Intra- and interassay coefficient of variation percentage (% CV) were < 6.3 and < 7.8 for E2 and progesterone, respectively (n = 10). The recovery rate of known E2 or progesterone concentrations added to a pool of culture maturation medium averaged 96.39%, and 98.65%, respectively. Compared with RIA, EIA values were in close agreement for E2 (n = 15, R = 0.96, P < 0.001), and progesterone (n = 15, R = 0.99, P < 0.001). Medium samples were obtained after oocyte maturation in vitro for 16 h. Use of IGF-I significantly elevated steroids production in the oocyte surrounded cumulus cells. The present study was designed to determine the effect of energy restriction on fatty acid profile of adipose tissue triglycerides from two different anatomic locations, in genetically obese rats. The impact of the addition of a high amount of olive oil to a restricted diet was also considered. Lean and obese male Zucker rats, 11 weeks old, were used. The former (n = 7) were fed a control diet ad libitum during 4 weeks, and the latter (n = 21) were randomized into 3 groups and fed either a control diet ad libitum or diets restricted to 75% of lean rat intake which provided different amounts of olive oil as fat source. Obesity induced strong modifications in fatty acid profiles from subcutaneous and mesenteric adipose tissues that were not corrected when a standard-fat 25% energy-restricted diet was used for feeding. In contrast, when a high amount of olive oil was included into the restricted diet, a new fatty acid profile in adipose tissue from obese rats, that mirrors the one of lean rats, was observed. Plasma viral RNA levels were inversely correlated (r = -0.72894) with total CD4+ cell counts and directly (r = 0.86964) with serum titre beta 2-microglobulin in chronically infected patients. They improve its accuracy and applicability, especially when slowly absorbed substrates are used or when volume flux measurements are desired. Calculations for the absorbed substrate and net fluid volume change determinations are reported. Male rats were treated on days 5-9 or 15-19 with 1 or 3 IU/rat.day, respectively, of recFSH, 10 IU/rat.day of hCG or vehicle and sacrificed the day following the last injection. Proliferation was studied by the immunohistochemical demonstration of DNA-incorporated 5-bromodeoxyuridine (BrdU) in both paraffin- and Epon-embedded tissues. No differences existed for the labeling indices in paraffin- or Epon-embedded tissues, but the identification of some cell types were easier in these latter one. At 10 days of age, no significant differences were found for the proliferative activity of the different cell types, except for the labeling index of foetal Leydig cells that was increased in hCG-treated animals. At 20 days of age, treatment with recFSH increased the testicular weight and tubule diameter, as well as the size of adult type Leydig cells and the labeling index of macrophages. Treatment with hCG increased the testicular weight and the number and size of adult-type Leydig cells, as well as the labeling indices of mesenchymal, adult-type Leydig cells and macrophages. The purpose of this study was to determine, in vivo, the effects of acute and/or subchronic cadmium chloride administration (6 mg/kg or 4 mg/kg/day during 14 days) on prolactin, luteinizing hormone (LH), follicle stimulating hormone (FSH), growth hormone (GH) and thyroid stimulating hormone (TSH) secretion in adult male rats. Six h after a single injection of CdCl2, a diminution of plasma levels of prolactin (4.52 +/- 0.53 vs. 16.2 +/- 2.7 ng/mL, p < 0.01), GH (3.39 +/- 0.41 vs. 6.71 +/- 1.30, ng/mL, p < 0.01), TSH (2.76 +/- 0.64 vs. 7.65 +/- 1.15 ng/mL, p < 0.01) and LH (4.1 +/- 1.3 vs. 5.18 +/- 0.28, p < 0.05) was observed, where as plasma FSH levels did not change. On the other hand, subchronic cadmium chloride administration for 14 days, increased plasma levels of GH (13.39 +/- 2.74 vs. 6.71 +/- 1.3 ng/mL, p < 0.05), TSH (27.8 +/- 4.42 vs. 6.65 +/- 1.15 ng/mL, p < 0.001), LH (11.1 +/- 1.3 vs. 5.18 +/- 0.28 ng/mL, p < 0.001) and FSH (53.16 +/- 3.66 vs. 12.51 +/- 1.45, p < 0.001), whereas plasma prolactin levels decreased (6.86 +/- 1.38 vs. 16.2 +/- 2.7 ng/mL, p < 0.01). In animals subchronically treated with CdCl2, body weight gain was lower than in control rats (p < 0.001). Histamine evoked a transient increase in [K/]o. The removal of calcium from the superfusing solution and addition of 10(-4) M EGTA caused a significant reduction in the histamine-evoked potassium outflow. Replacement of chloride (Cl-) in the physiological salt solution by nitrate (NO3-) caused a significant reduction in the histamine-evoked potassium release. However, when Cl- was replaced by bromide (Br-) the response to histamine was unaffected. Pre-treatment of pancreatic segments with furosemide (10(-4 M) or ouabain (10(-3) M) caused a marked reduction in the histamine-induced potassium release. The presence of specific inhibitors of cytochrome P450 activity significantly diminished the antioxidant capacity of estradiol. Twelve swimmers were selected to determine their performance in 25 meter crawl races. Their body temperature was also measured. This group was subsequently divided into 3 subgroups: Sub-group 1, made up of 2 subjects who acted as the control subgroup, trained only in the afternoon; Sub-group 2, composed of 5 subjects, trained only in the morning; and Subgroup 3, also made up of 5 subjects, trained in the afternoon but with an additional session of 10 min of swimming in the morning. All subjects followed their corresponding training pattern for three weeks after which they were subjected to a second study or testing day. Thereafter, they all trained one more week according to the same habitual schedule in the evening; after that, they were tested again.
