This study was performed to investigate putative alternative hosts of the pathogen, aiming to find support for disease management. Bacteria similar to Xcv were isolated from the weeds Alternanthera tenella, Amaranthus sp., Glycine sp. and Senna obtusifolia showing canker-like symptoms and collected in vineyards of Juazeiro and Petrolina at the Submedio Sao Francisco. No bacteria were isolated from Commelina benghalensis and Azadirachta indica showing similar symptoms. The pathogenicity of the isolates was confirmed in plants of A. tenella, Amaranthus sp., Glycine sp., S. obtusifolia and grapevine cv. Red Globe, under greenhouse conditions. The weeds Chamaesyce hirta, Dactyloctenium aegyptium, Eragrostis pilosa and Pilea sp., artificially inoculated with strains Xcv1 and UnB1216, also developed typical symptoms of bacterial canker.","PeriodicalId":211183,"journal":{"name":"Fitopatologia Brasileira","volume":"44 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2007-04-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"131955283","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Monosporic isolates from each species of Stenocarpella from infected corn stalks, collected in the experimental area of the University of Passo Fundo RS, were used throughout this work. Temperature range between 23 and 28 oC allowed the fastest mycelial growth for two isolates, both under continuous light and under 12 h photoperiod. No mycelial growth was observed at 5 oC and 45 oC for either isolate. Mycelium of both isolates grew faster under a photoperiod of 12 h. Conidia of both fungi did not germinate even after 24 h of incubation at 5 and 45 oC, independently of photoperiod. The highest proportion of conidial germination of the isolate of S. maydis was observed between 28 and 33 oC. On the other hand, conidia of S. macrospora germinated under lower temperature range than S. maydis. Conidia of both species germinated faster in the presence of light. For both isolates, sporulation on corn stalk segments was highest at temperatures ranging from 30 to 35 oC under continuous light.","PeriodicalId":211183,"journal":{"name":"Fitopatologia Brasileira","volume":"37 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2007-04-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"114771564","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Roots of Mucuna aterrima, Crotalaria juncea, Tagetes erecta and Lycopersicon esculentum were macerated, separately, in 1000 ml of tap water in a blender at low speed for 30 s, then sieved. Each of the resulting suspensions or just tap water (control treatment) was poured on one of the different soil samples inside a plastic bag and stored at 28 °C for 24 h. Seventy-eight bacterial isolates were obtained from the soil samples by serial dilution and selected for a subsequent assay for the biocontrol of Meloidogyne javanica. Tomato seeds were soaked in suspension of each bacterial isolate, then sowed in substrate inside plastic tubes in the greenhouse. The seedlings were each inoculated with 400 eggs of the nematode. The isolate UFV-6 of Escherichia coli reduced the number of galls to the highest extent (80%), and UFV-8 of Citrobacter freundii was the most effective in reducing number of eggs (83%). The identification of the isolates was done by fatty acid analyses and biochemical tests.","PeriodicalId":211183,"journal":{"name":"Fitopatologia Brasileira","volume":"87 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2007-02-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"126376904","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

However, only SCMV and Sorghum mosaic virus (SrMV) naturally infect sugarcane. In Brazil, SCMV seems to be the only causal agent of mosaic in sugarcane. Most sugarcane cultivars are believed to be resistant to mosaic, although new cases have been reported in the field. Symptomatic samples, from several clones and cultivars, were collected in sugarcane crops. Among these cultivars, RB72-454, one of the most grown in Brazil, and considered resistant, showed plants with mosaic symptoms. Samples were tested by DAS-ELISA with antisera to SCMV, Johnsongrass mosaic virus (JGMV) and Maize dwarf mosaic virus (MDMV), showing negative results. However, sorghum \"Rio\" and \"TX2786\" showed mosaic symptoms when mechanically inoculated with the viral isolates, indicating infection by SCMV. Total RNA was extracted and submitted to amplification with specific primers to SCMV and SrMV in RT-PCR. Specific bands with ca. 880 bp were amplified with the SCMV-specific primer pair, corroborating results from mechanical inoculation. PCR products were cloned and sequenced. One of the SCMV isolates comprises a new and more severe strain capable of infecting RB72-454 and other tolerant cultivars in the field.","PeriodicalId":211183,"journal":{"name":"Fitopatologia Brasileira","volume":"20 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2007-02-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"131322127","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Isolates exhibiting compatible reaction to cv.Primavera were incompatible to cv.Maravilha and vice versa, with the exception of six isolates that were compatible to both cultivars. The virulence of isolates from cv. Maravilha on 32 test genotypes of rice was significantly higher (t = 9.09, p < 0.0001) than the isolates from cv.Primavera. A phenogram constructed from virulence data showed two main groups, one constituted mainly of isolates from cv.Primavera (97.6%) and the other of isolates from cv.Maravilha (91.17%). Rep-PCR analysis of isolates using two primers designed from sequences of Pot2 showed that isolates could be clustered broadly into two groups. The average similarity within a cluster of isolates from cv.Primavera was significantly greater than the average similarity among the isolates of cv.Maravilha (t = 5.37, p < 0.0001). There was close correspondence between clusters based on PCR and virulence data (r = 0.48, p < 0.011). The results showed that isolates of P. grisea were cultivar specific and had low phenotypic and genetic diversity.","PeriodicalId":211183,"journal":{"name":"Fitopatologia Brasileira","volume":"17 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2007-02-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"128627604","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Infested substrates with 0.5, 1, 2, 4 and 8 x 105 CFU g-1 were introduced in plates where seeds of melon type Piel de Sapo cv. PS 1430 were sowed in seeding trays after two weeks of incubation. The seedlings were transplanted to pots with a similar substrate, infested additionally with 2.5 x 104 CFU of Chaetomium per gram and 20 CFU of M. cannonballus per gram of substrate. Three controls were considered: one infested with Chaetomium alone, another with M. cannonballus and one without any treatment. The experimental design was completely randomized with five replicates. General disease index and control percentage were evaluated forty-five days after transplant. The General disease index varied according to the increased Chaetomium concentration. The results demonstrated that 4 and 8 x 105 CFU of Chaetomium g-1 were equivalent to 0.9 and 0.6, respectively.
