Ratio values were highest in the group with the highest lymph lipid content (the oil-fed group, 2.20 +/- 0.08) and fell significantly in the other groups (fed group 1.46 +/- 0.09, unfed group 1.15 +/- 0.05). The areas under the blood and lymph concentration curves after the intravenous (i.v.) and intraduodenal (i.d.) administration of diazepam were used to determine absolute (F) and lymphatic (FL) bioavailability. The F value in the blood can be raised by increasing the amount of lipids, whereas in the lymph, under the same conditions, it falls. During the i.v. and i.d. administration of diazepam, FL always rises with an increase in the amount of lipids in the lymph. The role played by the lymphatic system in total diazepam absorption was determined from the experimental results of its i.d. administration. The enzyme was determined by the method of Kato et al. (1974). We found that the given plasma enzyme activity was significantly higher in females than in males, throughout the whole life-span. In addition, we found that minimum activity was reached on about the 14th and 21st day of postnatal life and again on the 40th day, while maximum activity was recorded at the ages of 5, 30 and 35 days and in adult rats. In adult animals (males and females), exposure to altitude hypoxia was followed by a statistically significant increase in plasma DBH activity, which was much more pronounced in females than in males. In males, 240 min after terminating hypoxia plasma DBH activity had returned to normal, but in females it was still significantly raised; after 48 h, plasma DBH activity in females was identical to the activity before exposure to hypoxia. In rats aged 5 and 35 days, hypoxia evoked a fluctuating response. A decrease in activity immediately after terminating hypoxia was followed at 60 min by a return to normal, but at 240 min there was again a significant decrease. The animals came from two different locations and altitudes, Srinagar, Kashmir at 1250 m and Sakti in Ladakh, 4500 m. We used spreading depression to eliminate functionally the cortex and the hippocampus. Suction ablation of the cortical somatosensory projection area was also used to test its involvement in the SerW SSAD induction. The hippocampal spreading depression but not the cortical one abolished the SerW SSAD induced by the stimulation of the thalamic nuclei. The animals with the suction ablation of the somatosensory projection area also produced SerW SSADs when the stimulation electrodes were placed in the thalamic ventrobasal complex (in intact animals this stimulation induces spike-and-wave SSADs but not SerW-SSAD). The ischaemia was induced by 4-hour occlusion of both common carotid arteries and was followed by reperfusion of different duration (10, 30 or 60 min). The extent of lipid peroxidation was determined by measurement of conjugated dienes (CD) and TBA reactive products. Maximal values of CD and TBA reactive products were found after 10- and 30-minute reperfusion. The isolated perfused heart according to Langendorff is a preparation which is easily obtained from all warm-blooded animals. The heart is usually perfused at constant pressure, but frequently at constant flow, mostly with oxygenated saline solutions. The ventricles do not fill with the perfusate and therefore do not perform pressure-volume work. If, however, an intraventricular balloon is inserted, the ventricle can contract isovolumetrically. 2. Measuring parameters. Mechanical parameters of the working myocardium (contractile force, volume, ventricular diameter), mean coronary flow, bioelectrical parameters (EKG, monophasic injury potentials), and cardiac rhythm can be measured and recorded. 3. Function tests. The intact function of the working myocardium and the coronary vessels should be examined using various simple tests regarding the myocardium, the smooth musculature, and the endothelium of coronary vessels. 4. Possible applications. In the Langendorff-heart of normal or pretreated animals, inotropic, chronotropic, antiarrhythmic or vasoactive substances can be investigated in the steady state or by means of specific stress tests. In addition, the preparation is particularly suitable for biochemical studies of myocardial metabolism. 6. Advantages. Due to the technical ease with which a variety of parameters can be measured, a single heart can provide maximum information about the condition of the myocardium and the coronary vessels. Both sexes reacted to the stressors (mainly immobilization) by a marked increase in B at the beginning of the light period. The maximum reaction, to both immobilization and ACTH, was found in males in the spring; females reacted more evenly throughout the year, with a maximum in the winter. The reactivity of both sexes was the lowest in the summer. These salt intake alterations induced moderate kinetic changes of the Na(+)-K+ pump which did not result in significant changes of ouabain-sensitive (OS) Rb+ uptake or Na+ net extrusion at in vivo Na+i and K+o concentrations because a decreased affinity for Na+i in salt-loaded animals was compensated by an increased maximal transport rate. High furosemide-sensitive (FS) Rb+ uptake in red cells of salt-deprived rats was caused by an increase of both the maximal transport rate and the affinity for Rb+o. Cation leaks were also higher in salt-deprived than in salt-loaded rats. In three age groups of rats fed a 1% NaCl diet FS Rb+ uptake (but not FS Na+ net uptake) rose with age due to an increasing maximal transport rate whereas the affinity of the cotransport system for Rbo+ did not change. The age-dependent changes in the kinetics of the Na(+)-K+ pump resulted in a slight decrease of OS Rb+ uptake with age that was not paralleled by corresponding Na+ net extrusion. No major age-related changes of cation leaks were found. Ischemia decreased the inositol phospholipid (PI) concentration in Dp and Vp and this was recovered during reperfusion. Within 6 h following ischemia, ethanolamine (PE) and serine (PS) phospholipid concentrations decreased in Dp and PS also in Vp. During the long reperfusion intervals the concentrations of the two major phospholipids, PE and choline phospholipid (PC) declined in Fg, Dp and Vp. No changes were observed in sphingomyelin (SM). The concentration of unesterified cholesterol (UC) was lower throughout the reperfusion period in Dp and Vp, while the decrease in Fg was delayed. The molar ratio UC/TPL was reduced starting from 24 h of reperfusion. Since hypoxia, inflammation and vascular remodelling are all central to the pathology of chronic bronchitis and emphysema, we have studied these cells in the lungs of subjects with this condition.
