A watermark is encrypted using double random phase fractional Fourier domain encoding technique and then encoded into the digital hologram. The hologram is watermarked in a plane at some known distance from the object so that even if a new hologram is generated from the original hologram the watermark can always be traced by propagating the new hologram back to the object and then onto the watermark plane. The watermark is retrieved successfully using the correct encryption parameters. We consider both numerical (full complex field) and optoelectronic (phase-only) reconstruction methods. In the paraxial approximation, the on-axis intensity as a function of the grating duty cycle shows a perfect fractal behavior. We discuss the properties of lens scaling and phase binarization in the paraxial, the non-paraxial and the rigorous case. Furthermore, we investigate the optimal focusing conditions for binary phase gratings. Here we present a superresolution technique that requires only a single exposure. Inspired by the compressive sensing approach that takes advantage of the information redundancy in typical images we show that true superresolution is feasible from a single exposure. The single shot superresolution system is based on the well known double random phase encoding setup which was broadly investigated for its encryption purposes. A proper optical design involving one diffractive lens outside a Michelson interferometer enables the recording of full-field off-axis holograms with high resolution and optical sectioning. We demonstrate our approach with some experimental results that show optical sectioning with a maximum resolution of 3.5 µm. A high resolution approach has been implemented in the off axis Deep UV DHM setup using oblique illumination to overcome the limitation introduced by the optical system and the theoretical resolution limit of 250 nm (according to Abbe criterion) has been verified, experimentally. Most notably, x-ray diffraction data was instrumental in the elucidation of the structure of DNA. To explore the inherent complexity present in biological systems, existing technologies are being pushed to their limits. Once again, scientists are looking to engineers to create innovative solutions to enable their exploration and discovery. Many of the new methods currently being developed focus on increasing the sensitivity of the detection technique by inventing new devices as well as increasing the specificity of the device by engineering synthetic targeting moieties and improved attachment methods. This presentation will focus on new optical technologies with an emphasis on bio/chem-detection applications. Specifically, the experimental and theoretical optical properties of several new optical devices, which were designed for the express purpose of biological and chemical detection, will be discussed.
