The second-order scheme has a single reference arm with a temporal lens and dispersive elements, and an object arm containing an object surrounded by dispersive media. The third-order imaging setup contains two reference arms instead of one. Both setups have a temporally completely incoherent light source providing maximum resolution. Perceived depth in an adjustable random pattern stereogram was matched to the depth in holographic objects. The objects in holograms were a microscopic biological cell and a macroscopic coil. Stereoscopic presentation increased perceived depth substantially in comparison to non-stereoscopic presentation. When stereoscopic cues were weak or absent e.g. because of blur, motion increased perceived depth considerably. However, when stereoscopic cues were strong, the effect of motion was small. Our goal is to fabricate large-surface-area plasmonic nanostructures that provide local enhancement of electromagnetic fields for applications in fluorescence and Raman scattering spectroscopy. Periodic two-dimensional arrays of nanogrooves and nanopillars are obtained by holographic photolithography that makes use of surface relief gratings on azo-polymer films. Individual cells are immobilized by use of optical tweezers and are monitored live while the concentration of Ca2+ ions in the buffer is changed simultaneously. Ca2+ ions affect cells' membrane morphology, which results in the change of effective radius as well as drag coefficient of the cell, both of which affect the Brownian motion of the cell in solution. This motion is measurable indirectly by tracking the forward scattering light and its dependence on size and drag coefficient of the cell.
