Several methods for determining PQ-redox have been established. Although some of these methods are quantitative, such as those based on liquid chromatography, they are typically sensitive to sample preparation. Here, we critically evaluate the use of fast chlorophyll a fluorescence induction kinetics (the so-called OJIP transient) for semi-quantitative PQ-redox estimation in green algae (Chlorella vulgaris) and cyanobacteria (Synechocystis sp. PCC 6803). The method, based on the evaluation of relative fluorescence yield at the J-step of the OJIP transient (VJ, VJ’), has already been reported; however, thus far, it has been used mostly for studying dark-acclimated leaves, which limits its range of application. Here, we show that the OJIP transient can be used for semi-quantitative estimation of PQ-redox in algal and cyanobacterial cell cultures, in addition to plants. We further show that it can reflect PQ-redox in both dark-acclimated and light-acclimated samples. Our systematic comparison of Multi-Color PAM, AquaPen, and FL 6000 fluorometers demonstrates that accurate measurement of VJ and VJ’ parameters in suspension cultures requires low culture density and a high-intensity saturation pulse. We further show that with increasing light intensity to which the cells are exposed, the state of photosystem II (PSII) changes due to light-induced reduction of quinone A (QA−) and conformational changes, which in turn influence both the sensitivity and dynamic range of the VJ’ parameter towards PQ-redox estimation. A comparison of fluorescence transients in Chlorella and Synechocystis revealed relatively narrow range of PQ-redox states across diverse conditions in Synechocystis, maintained by terminal oxidases present at the thylakoid membrane. While we discuss certain limitations, our systematic assessment suggests that the OJIP method has great potential to become a routine tool for semi-quantitative PQ-redox estimation under a wide range of experimental conditions in green algae and cyanobacteria.
","PeriodicalId":20130,"journal":{"name":"Photosynthesis Research","volume":"164 1","pages":"6"},"PeriodicalIF":3.8,"publicationDate":"2026-01-19","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC12816086/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"145998684","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":3,"RegionCategory":"生物学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

This scenario occurs in the peridinin-chlorophyll protein (PCP) from dinoflagellates, which contains clusters of peridinins surrounding chlorophyll a. Here, we present MPS-TR-EPR data for PCP from Heterocapsa pygmaea and a high-salt PCP variant from Amphidinium carterae. Spectral analysis reveals the orientation of the optical transition dipole moments (TDMs), reflecting the chromophore contributions to the exciton states. The results indicate a predominant role of Per614/624 in the lowest singlet exciton state, consistent with theoretical models, while, at the same time, also suggesting refinements to better align the models with the experimentally determined TDM orientations. The vertical migration of epipelic pennate diatoms is a key adaptive trait enabling optimal light acquisition. However, it remains unclear whether these organisms can detect and respond to long-wavelength light, especially because deeper photic layers are enriched in far-red due to attenuation of shorter wavelengths. Here, we investigated the phototactic responses of a natural microphytobenthic biofilm, primarily composed of epipelic pennate diatoms, to long-wavelength light at two low irradiance levels (5 and 20 µmol photons m⁻² s⁻¹), using a custom-built multispectral LED illuminator. Red light (660 and 680 nm) induced a strong upward vertical migration and a high effective quantum yield of photosynthesis. Far-red light (720 and 740 nm) also triggered a significant upward migratory response, although more moderate than red light. In contrast, near-infrared wavelengths (770 and 810 nm) elicited no significant migratory activity, indistinguishable from the dark controls. Phototactic migration was observed even at 5 µmol photons m⁻² s⁻¹, suggesting a high sensitivity to light at intensities potentially below the photosynthetic compensation point. Our results provide evidence that benthic pennate diatoms can behaviorally respond to long-wavelength, low-intensity light. This response, likely mediated by phytochrome-like photoreceptors, suggests the existence of a low-light, long-wavelength sensing mechanism that enables diatoms to detect fine-scale spectral gradients as cues for surface detection and vertical positioning within the sediment matrix.
","PeriodicalId":20130,"journal":{"name":"Photosynthesis Research","volume":"164 1","pages":"4"},"PeriodicalIF":3.8,"publicationDate":"2026-01-08","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC12783169/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"145934561","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":3,"RegionCategory":"生物学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Light between 50 and 150 µmol·m⁻²·s⁻¹ significantly enhanced biomass accumulation, chlorophyll synthesis, PSII efficiency, and the biosynthesis of phenolics, flavonoids, and anthocyanins. Gene expression analysis showed that key photoreceptor and flavonoid biosynthetic genes (TsCRY1-like, TsCHS-like, TsFLS, TsWD40, TsDFR-like) were highly induced at 100 µmol·m⁻²·s⁻¹, while TsCHS-like and TsANS-like were downregulated at 150 µmol·m⁻²·s⁻¹, indicating feedback regulation under excessive irradiance. OJIP fluorescence confirmed PSII saturation near 100 µmol·m⁻²·s⁻¹. Shaanxi sprouts exhibited stronger light responsiveness and higher antioxidant capacity than Anhui ones. Photosystem I (PSI) of cryptophyte algae binds alloxanthin-chlorophyll a/c-binding proteins (ACPs) equivalent to the light harvesting complexes (LHCs) of green algae and higher plants. Here, we report a high resolution (2.25 Å) structure of the PSI-ACPI supercomplex purified from the cryptophyte Chroomonas placoidea (C. placoidea) by cryo-electron microscopy. The PSI-ACPI supercomplex consists of a monomeric PSI core, 14 ACPI subunits and a Chl-binding subunit, CCPI-S. The 14 ACPI subunits are divided into two layers, with an inner layer including 11 ACPIs, and an outer layer consisting of 3 ACPIs. The CCPI-S mediates the connections and energy transfer between the outer and inner ACPI layers. Due to the current higher resolution, a unique PsaQ subunit bound at the lumenal side was identified in the PSI core. Two new pigment molecules Chl a856PsaA and Mon 627ACPI − 8 as well as 18 new lipids were identified, compared to the previously reported PSI-ACPI structure from the cryptophyte alga C. placoidea (Zhao et al. 2023). These lipids mediate the interactions of subunits and the assembly of PSI-ACPI. The redox couple ascorbate/2,6-dichlorophenolindophenol (Asc/DCPIP) was shown to be more efficient, than the couple Asc/N,N,N’N’-tetramethyl-p-phenylenediamine (TMPD) both in photosynthetic studies and in biohybrid photovoltaic devices. We investigated the interactions of DCPIP with purified cyanobacterial PSI in the presence of Asc excess under laser flash excitation. Here we show that DCPIP, in contrast to TMPD, competes efficiently as an electron acceptor with the backward electron transfer from [FA/FB]− to P700+ even at micromolar concentrations, indicating accumulation of the oxidized DCPIP under aerobic conditions in the presence of Asc excess. The reduction of P700+ includes contributions both from reduced DCPIP and semiquinone DCPIP•−. The rates of P700+ reduction and [FA/FB]− oxidation by DCPIP demonstrate complex pH-dependencies, related to changes in protonation state of the mediator and probably to redistribution of electron density between the terminal cofactors FA and FB. The rate constants of the electron transfer from Asc, DCPIP and TMPD to P700+ and of the electron outflow from [FA/FB]− to the oxidized forms of these compounds are estimated by kinetic modeling. The obtained data reveal thermodynamic, kinetic and electrostatic factors responsible for the high DCPIP efficiency as electron donor and acceptor for PSI.
","PeriodicalId":20130,"journal":{"name":"Photosynthesis Research","volume":"164 1","pages":"1"},"PeriodicalIF":3.8,"publicationDate":"2025-12-18","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"145782543","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":3,"RegionCategory":"生物学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

In this contribution, we computationally explore an expanded set of over 250,000 Chl variants, looking for potentially interesting targets for synthetic biology. We focus on optical properties, employing a machine learning (ML) approach and subsequently verifying the corresponding predictions using time-dependent density functional theory (TD-DFT) and multireference DFT (DFT/MRCI). We find that (i) Chl f is the best monosubstituted red-shifted Chl, as no other Chl in our set exceeds Chl f in terms of both red shift and absorption intensity, (ii) Chl b is not the best Chl to harvest photons from the green region of the optical spectrum, as several other Chls with the same or better green absorbance were identified (most notably DVChl b) and (iii) the T1 energy of Chls can be slightly adapted. The latter would enable experiments to determine whether it is beneficial having the T1 transition energy located between the two lowest O2 singlet state transitions, as it is found for Chl a. This might be a prerequisite for stable, efficient oxygen generation. Our ML approach thus provides a comprehensive overview on an extensive subset of potential Chl modifications that could be used for tuning oxygenic photosynthesis, if suitable synthesis pathways can be found.
","PeriodicalId":20130,"journal":{"name":"Photosynthesis Research","volume":"163 6","pages":"67"},"PeriodicalIF":3.8,"publicationDate":"2025-12-11","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC12698835/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"145724694","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":3,"RegionCategory":"生物学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

To enhance cultivation efficiency and increase metabolite yield, it is essential to understand how photosynthetic induction varies with leaf developmental stage. This study examined the dynamic changes in photosynthetic parameters during induction across young (YL), fully expanded (FEL), and old leaves (OL) of foxglove, and identified the predominant limitations constraining photosynthetic efficiency. FEL exhibited the highest net photosynthetic rate (A), carboxylation capacity (Vcmax), and cumulative carbon gain. However, the time required to reach 90% of A and Vcmax did not differ significantly among the three leaf types. Steady-state stomatal conductance was also comparable. Despite similar induction kinetics, YL showed the greatest carbon loss ratio, whereas FEL and OL displayed similar levels of loss. Limitation analysis revealed that stomatal factors predominated in YL and OL, accounting for approximately 70% of total photosynthetic limitation, while FEL exhibited a more balanced contribution from stomatal and biochemical limitations. Since mesophyll conductance (gₘ) was not measured independently, its effects were likely included within the biochemical component, potentially leading to an overestimation of biochemical limitation in FEL. The consistent induction speed across developmental stages supports simplification in modeling canopy-level photosynthesis in foxglove. These findings also inform the development of dynamic lighting strategies in controlled environments. In summary, these findings highlight stomatal regulation as a primary constraint on photosynthetic induction, thereby establishing a physiological basis for enhancing productivity and cardiac glycoside accumulation in foxglove cultivation.
","PeriodicalId":20130,"journal":{"name":"Photosynthesis Research","volume":"163 6","pages":"66"},"PeriodicalIF":3.8,"publicationDate":"2025-12-10","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"145724613","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":3,"RegionCategory":"生物学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

At the present, structures of PBS with tricylindrical and pentacylindrical cores have been determined while the structure of the PBS with a bicylindrical core is yet to be revealed. Here we report the cryo-EM structure of PBS with bicylindrical core from Synechococcus elongatus PCC 7942 (Synechococcus 7942) at an overall resolution of approximately 3 Å. Similar to the PBS with a tricylindrical core, six peripheral rods are attached to the core by the rod-core linker protein CpcG in the PBS of Synechococcus 7942 even though the core lacks the top AP cylinder, which is important for the attachment of peripheral rods to the tricylindrical cores. We found that the C-terminus of ApcE in the Synechococcus 7942 was involved in interacting with both CpcG and CpcB of a top peripheral rod, compensating for the absence of the top AP cylinder of the core and maintaining PBS stability. Analysis of the bilin distribution reveals that distance of excitation energy transfer from top peripheral rods to the terminal emitters is approximately 15% shorter compared to the PBS with tricylindrical cores. Although there are 30% fewer bilin chromophores in the Synechococcus 7942 PBS core compared with the tricylindrical core, the aromatic residue ring in the Synechococcus 7942 PBS core is conserved, supporting the suggestion that these aromatic residues from AP and linker proteins are critical to the energy transfer of PBS.
","PeriodicalId":20130,"journal":{"name":"Photosynthesis Research","volume":"163 6","pages":"65"},"PeriodicalIF":3.8,"publicationDate":"2025-12-08","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"145701614","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":3,"RegionCategory":"生物学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

PCC 6803 immobilized onto a porous Millipore nitrocellulose membrane filter (MF) and contacting with indium tin oxide (ITO) semiconductor glass slides, termed ITO|PS I - MF|ITO have been directly measured. In the presence of the ascorbate/2,6-dichlorophenolindophenol redox couple and disaccharide trehalose, photoinduced generation of ΔV with a maximum amplitude was observed upon light irradiation for more than 1 h. After storing the ITO|PS I - MF|ITO system in a desiccator at 22 ± 1 °C and ~ 65% relative humidity for 7 days in the dark, the photovoltage amplitude decreased by ~ 15%. A significant drop in the ΔV amplitude was observed on the 14th day of storage under these conditions. The amplitude of the stable ΔV signal was fully restored after re-injecting fresh buffer directly into the system, although maximum amplitude was reached more slowly. It is clear that longer preservation of PS I functional activity during storage of the system can be achieved at a higher relative humidity in the desiccator, since the photovoltage was completely suppressed within 24 h when the system was stored in the dark in the laboratory room. The protective role of trehalose is also discussed.
","PeriodicalId":20130,"journal":{"name":"Photosynthesis Research","volume":"163 6","pages":"64"},"PeriodicalIF":3.8,"publicationDate":"2025-12-02","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"145655205","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":3,"RegionCategory":"生物学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
