Serotype and distribution of adhesion genes in Streptococcus mutans isolated from people with Parkinson's disease.

The virulence of Streptococcus mutans, a key caries pathogen, is partly explained by its rhamnose-glucose polysaccharide (RGP) and its sucrose-dependent and -independent adhesion mechanisms. Given their role in comorbidities, this study investigates the diversity of S. mutans through analysis of RGP and sucrose-independent adhesion proteins (SpaP, WapA, Cnm, Cbm) in Parkinson's disease patients. In the PARKIDENT clinical trial, strains were isolated from saliva samples of Parkinson's patients at baseline and during a follow-up visit. Strains were preanalyzed by multiplex PCR for serotype and collagen-binding proteins, followed by high-throughput sequencing and bioinformatics analysis using RAST® and BLAST®. Phylogenetic analyses and protein modeling were also conducted. Of 40 patients in the Parkident clinical trial, only 24 had Streptococcus mutans strains isolable from salivary swabs. Serotyping revealed that over 80% of the 44 strains isolated were serotype c, in line with prevalence data in the literature. Furthermore, only SpaP types A and B were identified in the strains. The low variability observed for the WapA protein underlines its functional importance. Finally, the cbm gene was not found in any strain, and only 5 strains possessed the cnm gene, all of them serotype c. The study of these 44 Streptococcus mutans strains showed characteristics similar to other populations. Serotype c predominates, with minimal peptide sequence variability in adhesion proteins (SpaP, WapA). Collagen-binding proteins Cnm and Cbm are rare or absent. Further research is essential to better characterize strain distribution in the Parkinson's-affected population due to S. mutans' growing role in extra-oral pathologies. Progress in the understanding of its pathophysiology has been hindered by the lack of standardized animal models. Existing models involving first molar or multiple molar extractions disrupt anatomic landmarks and face technical challenges, such as retained root fragments and inconsistent surgical procedures, which reduce reproducibility. To address these limitations, we developed a novel rat MRONJ model using maxillary second molar extraction combined with standardized palatal gingiva resection. By preserving the adjacent first and third molars, anatomic landmarks were retained, enabling precise and reproducible evaluations. The modified extraction technique incorporating wedge insertion improved the success rate and minimized root fractures. Notably, our findings revealed that suppressed bone metabolism in the MRONJ model inhibited natural tooth movement observed in the control group, highlighting a unique pathologic hallmark of MRONJ. The model effectively reproduced MRONJ-specific features, including persistent bone exposure, impaired bone healing, necrotic bone formation, and inflammation. Three-dimensional micro-computed tomography and histologic analyses provided robust and quantitative assessments of bone pathology. By integrating anatomic standardization and precise quantitative assessments, this model addresses the key limitations of previous approaches. It also provides a reliable platform for investigating the pathogenesis of MRONJ and for assessing preventive and therapeutic strategies. Two windows were created on the buccal surface of 72 molars for the erosion cycle. One of the windows was treated with fluoride varnish [(FV, Metroberry, Imicryl), varnish containing STMP-treated eggshell and membrane powder (EPV, BioViera, Imicryl) and bioactive glass varnish (BAG, Polimo, Imicryl)] while the other window was used as control. Acidic syrup (Atarax) was applied 3 times a day and acidic drink (Coca Cola) 4 times a day for 5 days. The samples were analyzed by SEM (n = 1), ATR-FTIR (n = 6) and Vicker's hardness (n = 5). Data were analyzed by one-way ANOVA and post hoc Tukey test. The protective effects of the varnishes were observed in the SEM images obtained. There was a difference between the FV and EPV groups in the 875 cm<sup>-1</sup> v<sub>2</sub> CO<sub>3</sub><sup>-2</sup> peak spectrum and microhardness values (p < 0.05). There was a difference between FV and FV-C in the carbonate v<sub>2</sub> band in the acidic beverage demineralised varnish groups (p < 0.05). Other band areas and CO<sub>3</sub><sup>-2</sup> /PO<sub>4</sub><sup>-3</sup> ratios of the varnish areas against the erosion and control areas showed similar results (p > 0.05). Microhardness analyses showed that the BAG group demineralized with acidic syrup and the FV group demineralized with acidic drink were similar to the control group (p > 0.05). It is characterized by inflammation, destruction, and resorption of mineralized tissues located around the dental apex. Although this is a local infection, pathogens and their products in the periapical area, as well as inflammatory cytokines, can reach other regions of the body and trigger a systemic immune/inflammatory response in the host. This condition has been associated with several types of systemic diseases. This systematic review aimed to explore the association between AP and neuroinflammation, providing insights into this relationship and discussing potential pathways used by AP microorganisms to reach the central nervous system. Extensive searches in electronic databases (PubMed, Embase, and Cochrane) and hand searches were conducted from January 2013 until May 2024, using the keywords apical periodontitis, endodontic infection, periapical abscess, periodontitis apical, brain inflammation, brain disorders, and neuroinflammation. Initially, 394 studies were selected. After exclusion and inclusion criteria were applied, 12 studies (7 case reports and 1 case series, 3 experimental studies using animal models, and 1 cohort study) were included in this systematic review. All studies showed the presence of AP in upper and/or lower teeth and brain disorders with neuroinflammation. It is important to mention that some cases reported the death of the patient. This systematic review indicates a significant relationship between AP and neuroinflammation based on animal and human studies. Moreover, this study suggests there may be pathways connecting upper and lower teeth with apical periodontitis to the brain, inducing neuroinflammation. Additionally, the oral cavity can become easily accessible when AP is present, allowing for the invasion of microorganisms that are usually non-invasive. This contributes to the development of systemic disorders and/or strengthens their effect, worsening brain pathologies by causing or enhancing neuroinflammation, potentially leading to death. Bovine dentin samples were subjected to the following five treatment conditions: control, demineralization, demineralization followed by remineralization, demineralization followed by brushing then remineralization, and demineralization followed by remineralization then brushing. X-ray fluorescence spectroscopy was then utilized to assess the elementary composition changes, while scanning electron microscopy provided microstructural analyses. Surface roughness and microhardness were also quantified to assess the physical changes in dentin. The control group retained significantly higher Ca/P content compared with all treated groups, indicating that demineralization, regardless of subsequent treatment, leads to a substantial loss of hydroxyapatite. Among the treated groups, those that underwent remineralization immediately after demineralization manifested higher Ca/P retention compared with those that included brushing before remineralization. Additionally, microhardness measurements indicated that post-demineralization brushing negatively affected dentin's microhardness. The sequence of demineralization, brushing, and remineralization treatments significantly affects Ca/P retention in dentin, along with its surface roughness and microhardness. Matrix metalloproteinases (MMP) are endopeptidases that play a role in extracellular matrix remodeling and pathophysiology of various diseases. The study aims to evaluate the gene polymorphism and protein expression of MMP-8 and their association with the severity of OSF. The cross-sectional study design involves patients diagnosed with OSF. DNA was extracted from saliva samples by the CTAB method followed by PCR and Sanger sequencing. The sequences were examined for MMP-8 polymorphism on Mega X software. ELISA was performed to assess the MMP-8 protein expression. The results of genetic polymorphism showed the alleles AG (rs11225395), TT (rs1861602574), CC (rs562617158), TT (rs2133511737), AA (rs1307439859), and CC (rs2133511706) consistent with disease severity. MMP-8 protein levels decreased with disease progression. Meanwhile, the association between the two showed a noteworthy significance with the presence of specific genetic variants, namely rs1320632, rs2133511732, rs1307439859, and rs2133511737, rs1307439859, rs562617158, rs11225395 shows a robust association with changes in MMP-8 protein expression. This study suggests a strong association between genotype and the modification of MMP-8 protein expression. Thirty single-canal lower human premolars were endodontically treated, filled using the single-cone technique, and prepared for fiberglass post luting. The teeth were positioned in the alveoli of human mandibles and cone beam computed tomography (CBCT) examination was performed. The specimens were randomly distributed (n = 10) according to the root canal access technique: control group (G<sub>C</sub>)-no access; conventional access group (G<sub>CA</sub>)-root canal accessed with spherical diamond burs and ultrasonic inserts; and EG group (G<sub>EG</sub>)-root canal accessed with bone graft/anchorage drills and EG. At the end of the root canal access, new CBCT examination was performed to assess the root canal walls' thickness and the volume of dental tissue removed during the fiberglass post removal. The time required for the access was also recorded. The data were submitted to statistical analysis (Kruskal-Wallis and Mann-Whitney tests, α = 0.05). G<sub>CA</sub> promoted greater deviation than the G<sub>EG</sub> (p < 0.05) and resulted in greater removal of the root canal walls. The volume of dental tissue/fiberglass post removed, deviations promoted, and the time spent were significantly lower (p < 0.05 and p < 0.001, respectively) in G<sub>EG</sub> than in G<sub>CA</sub>. These bisphosphonates are known to significantly impact the healing of soft and bone tissues due to their anti-angiogenic properties. Our study aimed to explore whether applying the basic fibroblast growth factor (bFGF), known for its ability to promote angiogenesis and bone remodeling, over extraction sockets could prevent the onset of BRONJ in rats administered with ZA. The experimental protocol involved administering each rat with two intravenous injections of 60 μg/kg of ZA, followed by tooth extraction. Rats were then divided into two groups: the test group (n = 12), which had a mixture of 3% hydroxypropyl cellulose (HPC) and 0.3% bFGF applied over their extraction sockets, and the control group (n = 12), which received only a 3% HPC application. Wound healing was monitored for 8 weeks post-extraction and assessed using micro-computed tomography, histological evaluations, and immunohistochemical analysis of CD31- and CD105-positive vessels. Results showed a BRONJ incidence rate of 100% (12/12) in the control group, whereas the rate was significantly lower in the test group at 8.3% (1/12). Furthermore, the test group demonstrated marked increases in both angiogenesis and new bone formation. This study aimed to synthesize an experimental composite resin modified with grapefruit seed extract-mediated titanium dioxide nanoparticles (GSE-TiO₂NPs) and evaluate its antibacterial activity and mechanical and physical properties. Green synthesis of TiO₂ nanoparticles was conducted utilizing GSE. The chemical profile of GSE was identified through gas chromatography-mass spectrometry. The as-prepared nanoparticles were incorporated into experimental composites at concentrations of 10 wt.% (10 wt.% GSE-TiO₂NPs group) and 20 wt.% (20 wt.% GSE-TiO₂NPs group), with an unmodified composite as the control group. Scanning electron microscopy (SEM) and transmission electron microscopy (TEM) confirmed the morphology and size of synthesized nanoparticles, with SEM revealing uniform particle distribution in the composite. Antibacterial activity against Streptococcus mutans was assessed using the agar disc diffusion method. Mechanical properties, including flexural strength (FS) and flexural modulus (FM), were evaluated according to ISO 4049 standards. Microhardness was tested according to the American Society for Testing Materials (ASTM E-384:1999) criteria. Polymerization shrinkage was measured using the strain gauge method. Modified composites exhibited significantly greater antibacterial activity against S. mutans compared to the control (p < 0.001). The 10 wt.% GSE-TiO₂NPs group demonstrated increased FS and FM values compared to the control group (p < 0.05). All groups surpassed the clinically acceptable microhardness threshold without significant differences (p = 0.588). Polymerization shrinkage was reduced in the modified groups (p < 0.01), with the 20 wt.% GSE-TiO₂NPs group showing the lowest value (13.06 ± 0.92%). Early detection of biomarkers associated with periodontal disease (PD) can significantly aid in timely diagnosis and intervention. Myeloperoxidase (MPO) is an enzyme abundantly present in neutrophils and has been associated in the pathogenesis of PD. Here, we present a novel approach for the electrochemical detection of MPO using printed screen graphene electrodes coupled with principal component analysis (PCA) for data analysis. We employed cyclic voltammetry to characterize the electrochemical behavior of MPO using potassium ferrocyanide and hydrogen peroxide. The process was controlled by species diffusion on the electrode surface using a scan rate spanning from 10 to 400 mVs<sup>-1</sup>. In addition, we investigated the detection of hydrogen peroxide, a substrate of MPO, as a method to indirectly asses MPO electroactivity, leveraging a redox potential of - 500 mV. Saliva samples were collected and analyzed using the developed electrochemical sensor, followed by principal component analysis to differentiate between healthy and diseased samples based on MPO levels. Our findings demonstrate the feasibility of using printed screen graphene electrodes for the sensitive and selective detection of MPO, offering a promising approach for early diagnosis and monitoring of periodontal disease.
