Iida","doi":"10.3314/JJMM.37.143","DOIUrl":"https://doi.org/10.3314/JJMM.37.143","url":null,"abstract":"病原性黒色真菌感染の中で最も多くみられる臨床型はクロモミコーシスである.そこで,本稿では,クロモミコーシスの発症機序について述べた.まず,最も頻度の高い原因菌であるFonsecaea pedrosoi(以下F.pedrosoiと略す)によるクロモミコーシスの病理組織所見の特徴を検討した.クロモミコーシスは他のスポロトリコーシスや白癬性肉芽腫のような皮膚の深在性真菌症と異なり,真皮上層~中層に限局して細胞浸潤が認められる.この理由は,腐生的には菌糸形で存在するF.pedrosoiが病変組織内では球状のsclerotic cellの形に変換して認められ,sclerotic cellは真皮の下方には侵入できないことと関連しているようである.またsclerotic cellは真皮上層にとどまって活発に増殖するため,菌要素に向かって遊走する多核白血球や微小膿瘍および組織球,巨細胞,リンパ球などの感染性肉芽腫の像を形成する浸潤細胞も真皮上層~中層に存在することとなり,これらの稠密な浸潤細胞の何らかの働きかけが表皮突起の延長を誘発し,本症に特徴的な経排除現象が引き起こされると考えた.そこで,F.pedrosoiの教室保存株を用いて寒天埋没法を行い,菌糸形菌要素からsclerotic cellに変換する過程を観察したところ,菌糸形からsclerotic cellへの変換は宿主側の多核白血球の遊走が重要な因子であり,またsclerotic cellは菌にとって環境条件が悪いときの耐性形態と考えられた.さらに,赤道部に隔壁を有する球状細胞も認められ,sclerotic cellの増殖する動態も観察された.次いで,腹腔内寒天埋没法と同様の操作を行った寒天ブロックをマウスの背部の表皮下に埋没させ,実験を行った結果,この皮膚寒天埋没法は本症の早期感染組織像の観察と経表皮排除現象の解明に有用な方法と考えられた.最後に皮膚寒天埋没法を用いた経表皮排除現象における免疫担当細胞について若干検討した.Nudeマウスにおいては,正常マウスと同様に菌を含んだ寒天ブロックの経表皮排除を認めたが,NK細胞の欠如しているbeigeマウスではこの現象は起こらなかった.本症の菌の経表皮排除にはNK細胞機能の関与が示唆された.","PeriodicalId":19301,"journal":{"name":"Nippon Ishinkin Gakkai Zasshi","volume":"28 1","pages":"143-146"},"PeriodicalIF":0.0,"publicationDate":"1996-07-30","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"78012560","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Horibe, Kenji Tanaka","doi":"10.3314/JJMM.37.167","DOIUrl":"https://doi.org/10.3314/JJMM.37.167","url":null,"abstract":"The levels of IgG antibodies specific for enolase, extracellular proteinase (EPR) and common antigens of Candida albicans, namely, the 67, 62, 29 and 25 kDa components in five children with acute lymphoblastic leukemia (ALL) were sequentially monitored by Western blot analysis. The total amounts of immunoglobulins in all sera of patients were below the normal range. Enolase-specific antibodies were detected in four of the five patients and in 38-57% of healthy children. EPR-specific antibody which was not detected in healthy children was shown only in one patient. In contrast, antibodies specific for the 67, 62, 29 and 25 kDa components were frequently detected in patients as well as in healthy children. In most cases in which specific antibodies were detected, the intensities of bands were sequentially decreased during chemotherapy, especially for the 29 and 25 kDa. Candida cells were recovered from four patients with enolase-specific antibodies. Hyphal development in media containing low serum concentrations is characterised by the formation of sparsely branched hyphae in which uninucleated, highly vacuolated intercalary compartments are arrested in the cell cycle, probably in G1. Re-entry into the cell cycle is accelerated by enrichment of the medium for nutrients and is preceded by reduction of the vacuolar space and concomitant increase in the size of the cytoplasmic volume. Hyphae exhibit contact guidance (thigmotropism) which may facilitate the invasion of tissues through local wounds, invaginations or sites of weakened surface integrity. Thus hyphal growth is adapted for nutrient scavenging and penetration of irregular surfaces.The genetics of dimorphism and virulence has been investigated through studies of differential gene expression, and by specific manipulation of gene expression by overexpression and the creation of null mutants. The expression of many genes is regulated during dimorphism however few of these play active roles in the regulation of morphogenesis. Disruption of genes encoding certain secreted aspartyl proteinases (SAP2) and a key step in the assembly of O-linked mannosylation (MNT1) led to attenuation of virulence. Manipulation of the expression of a hyphal specific gene HYR1 did not affect germ tube growth while overexpression of a constitutively expressed Candida gene HST7 stimulated pseudohyphal growth in Saccharomyces.
