CD271 has been identified as a marker of the most homogeneous MSCs subset. In this study, we separated CD271 positive MSCs (CD271-MSCs) from canine adipose tissue-derived MSCs (AD-MSCs) using immunomagnetic selection method. Both CD271-MSCs and AD-MSCs showed similar morphology and expressed common immunophenotypic markers (CD34, CD44, and CD45). However, CD271-MSCs and AD-MSCs exhibited difference in chondrogenic differentiation potential. Specifically, CD271-MSCs exhibited increased expression levels of chondrocyte specific markers, collagen type II alpha 1 (COL2A1) and Aggrecan compared to AD-MSCs. Also, electrical stimulation (ES)-treated CD271-MSCs have a greater capacity for expressing heat shock protein 70 (HSP70), COL2A1, and Aggrecan compared to control without exogenous differentiation condition. Complete depletion of 1 g/L 1,4-dioxane by CK-4 cultures growing on AMS media was achieved within 168 h. Both the Biolog system and phylogenetic analysis using 16S rRNA sequencing were utilized for identification, and the strain was designated as Shinella granuli CK-4, and registered in GenBank as [MN841914]. The mortality of CK-4 was not greatly affected by 1,4-dioxane concentrations below 10 g/L but was increased with increasing 1,4-dioxane concentrations. SDS-PAGE revealed that the amount of lipopolysaccharide in CK-4 increased or decreased according to the 1,4-dioxane concentration. The expression of 60-kDa GroEL stress shock protein (SSP) was increased in proportion to concentrations below 10 g/L 1,4dioxane but was not expressed at concentrations above 15 g/L dioxane. On the other hand, 70-kDa DnaK SSP was not expressed at any 1,4-dioxane concentrations (0~20 g/mL) tested. SEM demonstrated that the surface of CK-4 cells exposed to 10 g/L dioxane was not significantly different from that of normal cells, whereas cells exposed to 20 g/L 1,4-dioxane showed the presence of perforations and irregular rod forms with wrinkled cell surfaces. These results provide clues to understanding the resistance of S. granuli CK-4 to 1,4-dioxane and the promotion of 1,4-dioxane degradation, which can be applied to the treatment of industrial wastewater with higher concentrations of 1,4-dioxane. Biorenovation is a method of modifying the structure of a broad range of substrates such as chemical compounds and plant extract by microbial enzymes with the potential benefits of reduced cytotoxicities and enhanced biological activities relative to its parent substrates. we generated BN extract using biorenovation technology and tested the whitening effect properties of its biorenovation product (BNBR) in α-MSH stimulated B16F10 melanoma cells. Our results showed that BNBR significantly inhibited enzymes of melanogenesis such as TRP-1, TRP-2, MITF, tyrosinase in a concentration dependent manner without no noticeable toxicities. Therefore, it suggests that maybe various components that converted by biorenovation contained in the BNBR affect multiple factors involved in α-MSH-induced melanogenesis in B16F10 cells. The antidiarrheal activity was determined against castor oil induced diarrhea model, gastrointestinal transit and enteropooling tests in Swiss albino mice. The test groups received various doses (100, 200, and 400 mg/kg, body weight) of the extract, whereas positive control received loperamide (3 mg/kg, b.w.) and negative control was supplied with 1% tween-80 in distilled water (10 mL/kg, b.w.). At three test doses (100, 200 and 400 mg/kg), the extract showed significant (p < 0.001) and dose dependent antidiarrheal activity in all models. The delay and reduction in the onset, frequency, and weight of diarrheic feces as well as inhibition of the gastrointestinal transit and intraluminal fluid accumulation were observed in all models and highly comparable to the standard loperamide. The extract also exhibited high inhibitory ability to scavenge superoxide anion (O2 • ). In this study, we examined the whitening effect of Ligustrum japonicum (LJ) and LJ biorenovation extract (LJBR) in α-MSH-stimulated B16F10 melanoma cells. In the non-toxic concentrations of 25, 50, and 100 μg/ml, the inhibitory effect of LJ and LJBR on melanin synthesis and tyrosinase activity was observed. LJBR exhibited a stronger inhibitory effect on melanin synthesis and tyrosinase activity relative to its reaction substrate LJ. In addition, the protein expression level of MITF, TRP-1, TRP-2, and tyrosinase, which are known to play a critical role in melanin synthesis, was measured. As expected, a greater inhibitory effect on melanogenesis was observed in LJBR than in untreated group.
