One of the areas included in the flow of the Brantas River is the Jombang Regency. The Jombang Brantas River is used by the community to meet their daily needs in agriculture, plantations, industry, and fishing grounds. The input of waste generated from several community activities will result in the survival of fish in the waters. Fish as a biomarker agent is quite significant in estimating the level of heavy metal pollution. Red Bader Fish (Barbonymus altus) is often found and lives around the Jombang Brantas River. This study aims to determine the condition of the aquatic environment by observing the health of the fish through hematological observations of the Red Bader Fish (B. altus). Sampling was carried out at three stations with three replications every two weeks, which was carried out in February – March 2022 in the Jombang Brantas River. Data analysis used the Canonical Correspondence Analysis (CCA) method as an analytical method to determine the correlation between the dependent variable (hematological profile) and the independent variable (water quality conditions). In this study, the results showed that the water quality at Station 1 and Station 2 was classified as normal, while at Station 3, it was classified as polluted. Data analysis showed that erythrocytes, hemoglobin, and hematocrit had a relationship with temperature, pH, DO, BOD, ammonia, TSS, and TDS in low concentrations. There is an urge to develop an efficient Klebsiella vaccine, and epitope-based vaccines may offer a practical choice. Therefore, this study aims to design an epitope-based vaccine construct. YidR gene is expressed in all strains of Klebsiella with 97.6% of sequence homology, which makes it a potential vaccine candidate. In silico approach was used to make a multi-epitope vaccine construct consisting of immunogenic epitopes against the YidR gene of Klebsiella. Acquired multi-epitopes vaccine was analyzed for antigenicity, allergenicity, toxicity, and various physicochemical parameters. Molecular docking simulation was performed using TLR2 and TLR4 to asses binding affinity with the designed vaccine construct. The rice was damaged by N. Lugens sucking plant liquid in the form of carbohydrates in the form of starch. Environmentally friendly control methods are needed to reduce the increasingly high use of inorganic pesticides. Control of N. lugens with biotechnology can be done by inhibiting the metabolic cycle in these insects. This method utilizes the proteins available in rice seeds to stop the performance of enzyme α-amylase in the digestive system of N. lugens. α-amylase is an enzyme that plays a role in the process of starch degradation so that it becomes a simpler form both in microorganisms. The characterization of α-amylase enzymes in insects is an important first step to the determination of appropriate protein inhibitors so that they can be used to produce pest-resistant Genetically Modified Organisms crops. The research was conducted in the Agrotechnology laboratory, University of Jember. The research began with isolation and purification of α enzymes from brown stems and then continued with testing of α-amylase activity. Test parameters include the effect of temperature, pH, and substrate concentration on the activity of the α-amylase enzyme. The results showed that supernatant extracted from brown planthoppers produces a clear zone in the agar medium, which means the activity of enzymes α-amylase in the hydrolysis of starch. The pH value of 6 provides the most optimum conditions for the activity of α enzymes. The α-amylase enzyme is able to work optimally in the temperature range of 30°C - 45°C, and experiences a decrease in activity when the temperature reaches 50°C. However, over time this organic farming system experienced a decrease in fruit production, possibly from soil fertility and biofertilizers that were less available in the soil. This study aims to analyze the nitrogen-fixing ability and identify rhizosphere isolates that excel in nitrogen-fixing obtained from the rhizosphere of cacao (Theobroma cacao L) plant. Bacteria were isolated from the soil surrounding cocoa plant roots and grown on Nfb (Nitrogen free Bromothymol Blue) agar media. The nitrogen-fixing bacteria were analyzed with quantitative and qualitative methods. Six potential nitrogen-fixing isolates were identified based on the 16S rDNA sequence. The total number of isolates obtained from nitrogen-fixing isolation was six isolates. Reservoirs have different characteristics from other water bodies because they receive continuous water input from the river that flows through them. The river water contains organic and inorganic materials that can cause pollution caused by various kinds of human activities. The pollutant that is currently getting more attention is microplastics. Hence, various ways are developed to monitor the presence of microplastics in environments. The biotic component that may adsorb and accumulate microplastics is microbes that formed biofilm matrices as a predominant habitat. This study analyzes the presence of microplastics in water and biofilm matrices in Lahor Reservoir. The water parameters (pH, dissolved oxygen, temperature, and flow velocity) were also measured. Samplings were carried out at three stations, namely station A (inlet channel), station B (middle), and station C (outlet channel). This study revealed that water quality parameters were still relatively good according to the environment's quality standards. The average value of microplastic abundance in water at station A was 0.0013 particles.mL -1 , station B was 0.00083 particles.mL -1 , and station C was 0.00072 particles.mL -1 . The average abundance of microplastics in the biofilm at station A, station B, and station C was 7.55 particles.g -1 , 7.26 particles.g -1 , and 4.59 particles.gram -1 , respectively. This study indicates that the abundance of microplastics in the biofilm in the Lahor Reservoir was thousands of times higher than in the water. It has a high carbohydrate content and nutrients with a low glycemic index value. Taro is widely cultivated with intercropping techniques, especially in Kedungkandang District, Malang City. However, this technique caused a decrease in talas tubers production with only 710 tons ha-1. This number was lower than the total optimal production with 20.7 tons ha-1 of taro tubers. Plant Growth Promoting Rhizobacteria (PGPR), one of the most potential biofertilizers, can solve this problem with abilities such as phosphate-solubilizing, production of IAA, and nitrogen fixation. This research aimed to analyze the potency and identify the species of taro rhizosphere bacteria that had the best ability as PGPR agents. Taro rhizosphere bacteria were isolated using Pikovskaya medium and TSA (Tryptic Soy Agar) respectively. The PGPR abilities were evaluated on the phosphate-solubilizing, production of IAA, and nitrogen fixation. A total of 12 isolates for phosphate-solubilizing bacteria and ten isolates of IAA-producing bacteria. The highest concentration of phosphate solubilization was P1 isolate, with the concentration of 6.8 μg.mL-1; while I4 isolate had the highest potency for IAA production with the concentration of 23.11 μg.mL-1. Isolates P1 and I4 were selected for the nitrogen fixation ability test. P1 isolate shows the highest ammonia concentration of 2.52 μg.mL-1, and it was identified as Stenotrophomonas rhizophila with a similarity of 99.91% with Stenotrophomonas rhizophila R2A2 67. The purpose of this study is to determine the effect of steamcooling treatment on the characteristic of arrowroot starch and to investigate the feasibility of arrowroot-based RS application on wet noodles. The study used a Completely Randomized Design (CRD) with one factor which was the steam-cooling cycle. Later, the substitution of 5%, 10%, and 20% of modified arrowroot starches on wet noodles was used, and then the consumer test was carried out. The results showed that the RS content of arrowroot starch remained after steam-cooling treatment. The treatment, however, reduced the water content and the brightness of the starch. Subsequently, arrowroot starch substitution without or with modification also reduced the lightness and tensile strength of wet noodles. Also, wet noodles with modified arrowroot starch substitution were significantly different in color, taste, and aroma parameters but were still acceptable to the panelists at a maximum substitution of 20%.
