Klimenko","doi":"10.17537/ICMBB18.50","DOIUrl":"https://doi.org/10.17537/ICMBB18.50","url":null,"abstract":"Целью работы является исследование связи фактора роста эндотелия сосудов VEGF в сыворотке крови c гипоксией в группах пациентов, страдающих тяжёлыми неврологическими заболеваниями. Имеющиеся в литературе данные свидетельствуют об активации синтеза VEGF при гипоксии. Вместе с тем стандартный корреляционный анализ не позволил достоверно выявить по клиническим данным наличие связи между уровнем VEGF и параметрами оксиметрии, объективно характеризующими снабжение организма кислородом. В статье представлена методика, позволившая статистически достоверно доказать существование указанной связи, сводящейся к увеличению корреляции между VEGF и комплементом C4, а также между VEGF и белками S100 при снижении уровня оксигенации ниже некоторого порогового значения. Методика основывается на построении оптимальных разбиений интервалов значений показателей оксиметрии. Likhoshvai","doi":"10.17537/ICMBB18.18","DOIUrl":"https://doi.org/10.17537/ICMBB18.18","url":null,"abstract":"В кишечнике человека наиболее энергетически выгодным субстратом для бактерии Escherichia coli является нитрат, побочным продуктом восстановления которого является токсичный нитрит, используемый, хотя и с меньшей эффективностью, чем нитрат, в качестве акцептора электронов в дыхательной цепи. Нитрит восстанавливается до аммония периплазматической Nrf нитритредуктазой, кодируемой nrfABCD опероном, уровень эффективности транскрипции которого зависит от концентрации нитрита в среде Ранее нами была выдвинута гипотеза, что активность Nrf редуктазы при низких концентрациях нитрита в среде определяется не только механизмами регуляции экспрессии оперона nrfABCD, но и действием мембранного потенциала на процессы формирования активной формы фермента в периплазме. Эта возможность была теоретически доказана на математической модели, в которой мембранный потенциал представлен в виде феноменологической функции, зависящей от нитрита. Для окончательного подтверждения гипотезы была создана модель утилизации нитрита в хемостате, в которой величина мембранного потенциала была рассчитана на основе молекулярно-генетических механизмов его формирования. Поскольку структура дыхательной цепи на нитрите неизвестна, были реконструированы два гипотетических сценария формирования мембранного потенциала при культивировании клеток в хемостате и разработаны соответствующие модели. В сценариях рассматривается участие форматгидрогенлиазных комплексов FHL-1 и FHL-2, компонентами которых являются форматдегидрогеназа Fdh и гидрогеназы Hyd-3 и Hyd-4 соответственно. Было показано, что результаты математического моделирования не зависят от сценария формирования потенциала. Полученные результаты подтверждают, что мембранный потенциал играет важную роль в регуляции активности периплазматической Nrf редуктазы при низких концентрациях нитрита. Methods of Formal Concept Analysis allow clustering of multidimensional data under the single condition of partial ordering of such data sets. As a result, clusters are separate sublattices in the concept lattice, where each sublattice contains hierarchically related formal concepts. This solution of the clustering problem allows deep investigating both mutual influences of genes and their influence on other data obtained in experiments on gene expression. The paper describes a new information technology developed for the implementation of the proposed approach. The technology uses modern solutions in the field of Big Data processing, it has functions for communication with external data sources and other information systems.
