Elshafey, Rania Mayel, Mohamed Nasr, Noha Beder, Nagwa I El-Kasrawy, Asmaa Saleh","doi":"10.21608/djvs.2023.242255.1124","DOIUrl":"https://doi.org/10.21608/djvs.2023.242255.1124","url":null,"abstract":": Seven hundred-eighty dairy cows in Kafr-Elshikh and El-Beheira provinces were tested for subclinical ketosis (SCK) using qualitative determination of β -hydroxybutyric acid (BHBA) in urine from January to December 2022. All 280 positive cows were gathered in a questionnaire. The clinical examination, serum biochemical analysis, complete blood picture, and liver ultrasound of 89 subclinical ketotic cows were all performed. Our findings showed that both ruminal motility and milk production had significantly decreased while serum levels of BHBA, non-esterified fatty acid (NEFA), TAG, GGT, AST, ALT significantly elevated, blood glucose, total protein, albumin, and calcium levels decreased. Hb, RBCs, PCV, MCV, MCH, and MCHC, whereas WBCs, neutrophils, eosinophils, basophils, monocytes, and neutrophil-lymphocyte (N/L) ratio significantly increased. These cow's ultrasonography examinations revealed the typical signs of fatty liver. The prevalence of subclinical ketosis was 35.9 %. The development of SCK in dairy cows is influenced by various risk factors such as cows at the first 60 days of lactation, body condition score (3.5-5), age greater than 6 years, high milk production, and parity greater than three times. Early detection of SCK in early postpartum dairy cows is crucial to prevent financial loss to the farmers. A total of 350 raw milk samples were randomly collected from clinically mastitic bovine milk. Also, 140 oral swabs were obtained from patients attending external clinics of El-Delingat Hospital to be examined for the presence of Candida. The recorded results showed a high frequency of Candida spp. (36.9%) among the examined milk samples. Candida spp. isolated from cow milk (39.9%) was higher than that isolated from buffalo milk (29.3%). Also, the total prevalence of Candidiasis in human samples was 62.9% and the highest prevalence in relation to age groups was noticed in the age group > 2 - < 16 years (65.5%) and finally, the age group >40 years (58.8%). The identified Candida spp. in human samples were C. albicans (37.5%), C. glabrata (17.1%) , C. krusei (14.8%), C. parapsilosis (13.6%), C. tropicalis (11.4%), C. guilliermondii (4.5%) and C. kefyr (1.1%). It was observed that the highest prevalence of Candida species was among artificially fed infants (77.8%) and the lowest prevalence was detected in breastfed infants (53.3%). Based on the recorded results, Candida species constitute a major threat to both dairy animals and humans. This study aimed to assess the impact of substituting egg yolk with low-density lipoprotein (LDL) in Tris-Citric-Fructose (TCF) chilled extenders, with or without ascorbic acid, on the canine semen quality at 24, 48, and 72 hours of cooling. Semen from three German shepherd dogs was evaluated and extended at a 1:1 ratio using four extenders: Group A (TCF buffer, 20% egg yolk, with ascorbic acid), Group B (TCF buffer, 20% egg yolk without ascorbic acid), Group C (TCF buffer, 20% LDL, with ascorbic acid), and Group D (TCF buffer, 20% LDL without ascorbic acid). The extended semen was stored at 4°C and assessed after 24, 48, and 72 hours of cooling. The egg yolk extender resulted in less reduction in progressive sperm motility after 24, 48, and 72 hours of cooling; however, reduction in normal morphology, acrosome integrity, and cell membrane integrity were lower in TCF-LDL chilled extender than in TCF-EY chilled extender. Furthermore, after 24 hours of cooling, all evaluation parameters in both extenders were greater than 50%. The infection of monogeneans is the main cause of fish anemia leading to high mortalities. Thus, this study aimed to detect monogeneans parasites in Scoberomorus commerson and investigate the histopathological changes related to its natural infections. Furthermore, to research the potential parasite-host interactions. One hundred marine fishes ( S. commerson ) from the Ezbet-El Borg area, Damietta province, Egypt were examined from August 2022 to January 2023 for the presence of monogenean gill parasites. 38 fish were infected with monogenean species: Pricea multae 32% and Gotocotyla acanthura 19%. The structure of P. multae and G. acanthura were explained by using light and scanning electron microscope (SEM), the infected fish have no pathognomonic lesions, but for a few cases showed excessive mucus secretion. Six multiparous lactating Holstein cows were kept and examined twice weekly using ultrasound from day 11-19 PP until the appearance of a large mature corpus luteum (CL). The results showed that four of six cows (66.66%) exhibited dominant follicles (DF >10 mm) within 2 weeks PP (day 11, 15, 15, 16) and one cow (16.6%) needed 3 weeks (day 19) to develop DF. While only one cow (16.6%) displayed a delay (30 days pp) in the appearance of DF. The shortest interval between calving to the beginning of 1st luteal formation was 15 days recorded for one cow and the longest interval was 57 days and recorded also for one cow. While, in the other 3 cows, 29, 33, 43 days were recorded as the interval from calving to formation of first luteal tissue. The development of cysts following anovulated follicles with the size of 19-20mm occurred in two cows (33.3%). The persistence of cysts was longer in larger size cysts compared with the smaller one. Spontaneous recovery of the smaller cyst was faster than the larger cyst (10 vs. 23 days). The present study aimed to investigate the response of dairy cows with a syndrome of smooth inactive ovaries to different hormonal treatment protocols and their impact on reproductive performances. Lactating multiparous Holstein cows ( n = 75) having smooth inactive ovaries diagnosed by two successive ultrasonographic examinations performed at 10 days interval were used in this study. Cows were assigned randomly into 5 groups (15 each). Group A, treated with a single injection of 250 µg GnRH, Group B with a single injection of 2500 IU HCG, Group C, in which cows received CIDR for 7 days and PGF2α at day 6 (24 hours before removal of CIDR followed by injection of 250 µg GnRH (at day 8) 24 hours after removal of CIDR (at day 8)(CIDR, PGF2α and GnRH), Group D in which cows received CIDR for 7 days and PGF2α at day 6 (24 hours before removal of CIDR) followed by injection of 2500 IU hCG 24 hours after removal of CIDR (CIDR, PGf2α, and HCG), and Group E in which cows did not receive any treatment and kept as a control group. The results showed that CIDR+ GnRH and CIDR+ hCG protocols achieved the full (100%) estrus induction rate (EIR). hCG group achieved 93% and GnRH group 80%, while the control group only achieved 20% EIR. The shortest treatment estrus intervals (TEIs) were recorded in CIDR+ hCG and CIDR+ GnRH protocols. Mean±SD of TEIs were 1 ± 0, 1.4 ± 1.1days 10.4 ± 5.4, 16.7 ±7.1 and 23.7± 2.9 days in CIDR+ hCG . CIDR+ GnRH, HCG, GnRH and control groups respectively. The CIDR+ hCG group had the highest pregnancy rate (86.7%), followed by hCG (79.9%) and CIDR+ GnRH groups (80%), and GnRH group with 73%. The control group showed a 13.3 % pregnancy rate however, GnRH group showed the highest total conception rate (91.6%). It was concluded that the highest EIR and shortest TEI were obtained in CIDR+ GnRH and CIDR+ hCG protocols. Forty-five albino rats were divided into 9 equal groups. G1: Control, G2: Ochra 50 µg/kg b.wt, G3: Ochra + Nano Se 0.31 mg/kg b.wt, G4: Ochra+ vitamin C 1000 mg /L water, G5: Ochra+ Probiotic 400 g/ton of diet, G6: Ochra + Nano Se + vitamin C, G7: Ochra + Nano Se + Probiotic, G8: Ochra + Nano Se + vitamin C+ Probiotic, G9: Ochra + vitamin C + Probiotic. The obtained results showed significant increase in serum AST, GGT, ALT, and LDH activities, Creatinine, urea, TNF-α , IL-6, SOD, CAT, MDA, alpha 1, gamma 2, T. alpha and T4 concentrations in OTA intoxicated rats. Meanwhile, a significant decrease in GSH, TAC, total protein and its fractions including (albumin, Total beta, Total gamma, Total globulin, alpha 2, beta 1, gamma 1), albumin/globulin (A/G) ratio in addition to serum testosterone, estradiol, TSH and T3 concentrations were observed in OTA exposed rats. The results suggested that NanoSe, probiotics, and vitamin C are strong antioxidants agents that might ameliorate OTA-induced renal and liver damage in rats by combating free radical and attenuate oxidative stress. Oxidative stress is one of the aging associated mechanisms induced by pro-oxidants. Oxidative stress is an imbalance between free radicals and antioxidants in body cells. In the current study, we investigated the molecular docking scores of rutin with aging associated biomarkers, including human β -galactosidase, caspase-8, caspase-9, caspase-3, phosphatidylinositol-4,5-bisphosphate 3-kinase catalytic subunit delta (PK3CD), phosphatidylinositol-4,5-bisphosphate 3-kinase catalytic subunit gamma (PK3CG), and RAC-alpha serine/threonine-protein kinase (AKT1). All proteins were retrieved from RCSB PDB database (https://www.rcsb.org/), while rutin three-dimensional structure was obtained from PubChem database (https://pubchem.ncbi.nlm.nih.gov/). The docking process was achieved by InstaDock software (https://instadock.webs.com/) and visualized by BIOVIA Discovery Studio Visualizer software. Rutin exhibited binding affinity (p Ki ) against AKT1, PK3CD, PK3CG, β -galactosidase, caspase-8, caspase-9, and caspase-3 of 7.85, 7.04, 6.89, 6.82, 5.94, 5.57, and 5.50, respectively.
