This study investigated the synthesis of human IgG antibodies that specifically target HBsAg subtype adr in CHL-YN cells, a newly established cell line that grows faster than CHO-K1. To achieve the synthesis of human IgG antibodies, a plasmid vector encoding DNA sequences for human IgG antibodies against HBsAg was constructed and then transiently transfected into CHL-YN cells. The expression and antigen-binding capacity of the recombinant human IgG antibodies were analyzed using western blot and ELISA. The results showed successful expression and secretion of human IgG antibodies that recognize HBsAg subtype adr in CHL-YN cells. The ELISA test confirmed the specificity of the human IgG antibodies towards HBsAg subtype adr. Thus, this study concluded that human IgG antibodies that target HBsAg subtype adr were transiently expressed in CHL-YN cells.","PeriodicalId":16255,"journal":{"name":"Journal of Mathematical and Fundamental Sciences","volume":null,"pages":null},"PeriodicalIF":0.7,"publicationDate":"2023-05-07","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"78293281","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

One solution that can be applied is agriculture with hydroponic technology. To increase production efficiency, one must know the physical parameters that most influence the production process. This research used an IoT system to gather accurate and precise measurement data of physical parameters to be used as a dataset for machine learning. The dataset consisted of light intensity, humidity, air temperature, and total dissolved solids (TDS). Plant growth was measured by leaf area of the plant, number of leaves, and plant stem length every 3 to 4 days. The models used in the machine learning process were linear regression, polynomial regression, and random forest regression. The machine learning results showed that the best model for predicting plant growth was random forest regression with an MAE of 8.3% and an R2 of 0.93, for both bok coy and water spinach. The variables that influence growth the most are TDS and light intensity. According to the relationship between TDS gradient and plant growth gradient, the most optimal growth can be achieved by raising the TDS gradient or by maintaining a high TDS, which can be achieved by adding nutrient solution to the tank regularly.","PeriodicalId":16255,"journal":{"name":"Journal of Mathematical and Fundamental Sciences","volume":null,"pages":null},"PeriodicalIF":0.7,"publicationDate":"2023-04-17","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"78856238","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Other potential uses of the plant are being explored, including in food and medicine. Therefore, this study carried out antibacterial assay and gas chromatography-mass spectroscopy of G. versteegii fruit extracts in three solvents with different polarities, i.e., n-hexane, dichloromethane, and methanol. The agar well diffusion method was then used to test the extracts against Staphylococcus aureus and Escherichia coli. The dichloromethane extract showed the most effective antibacterial activity against S. aureus at a concentration of 40%, with a zone of inhibition of 13.17 mm compared to E. coli, for which it was 7 mm. Furthermore, the bacterial species showed different responses to the extracts. Total and partial inhibition was shown by the G. versteegii fruit extracts and E. coli, respectively, against S. aureus. Furthermore, GC-MS identified the following compounds in G. versteegii fruit extract: palmitic, oleic, and stearic acid, as well as bis-(2-ethylhexyl) phthalate, 2,3-dihydro-3,5-dihydroxy-6-methyl-4H-pyran-4-one, methyl octadec-9-enoate, squalene, and 2-monopalmitin derivates. The results showed antibacterial potency of G. versteegii fruit extract against pathogenic bacteria. Additionally, the antibacterial activity of the extract was stronger against S. aureus compared to E. coli.","PeriodicalId":16255,"journal":{"name":"Journal of Mathematical and Fundamental Sciences","volume":null,"pages":null},"PeriodicalIF":0.7,"publicationDate":"2023-03-04","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"88864739","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

From the time COVID-19 came into the limelight, people have been losing their loved ones and relatives as a direct result of this disease. Here, we present a six-compartment epidemiological model that is deterministic in nature for the emergence and spread of two strains of the COVID-19 disease in a given community, with quarantine and recovery due to treatment. Employing the stability theory of differential equations, the model was qualitatively analyzed. We derived the basic reproduction number  for both strains and investigated the sensitivity index of the parameters. In addition to this, we probed the global stability of the disease-free equilibrium. The disease-free equilibrium was revealed to be globally stable, provided and the model exhibited forward bifurcation. A numerical simulation was performed, and pertinent results are displayed graphically and discussed.","PeriodicalId":16255,"journal":{"name":"Journal of Mathematical and Fundamental Sciences","volume":null,"pages":null},"PeriodicalIF":0.7,"publicationDate":"2022-12-28","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"76153895","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Despite being known to be important for the host animal, the gut bacteria of blister beetles have not been investigated deeply. This study aimed to provide the first initial description of the gut bacterial community of Mylabris pustulata as one species of blister beetles, based on a culture-independent technique. Adult blister beetles of the same colony were sampled and confirmed as Mylabris pustulata Thunberg. The gut content was used in high throughput sequencing, targeting the V3-V4 regions of 16S rRNA gene, and in the cultivation of resident bacteria. The results showed that higher bacterial richness was present in the gut communities when compared to the grown bacterial culture. Proteobacteria was confirmed as the most abundant phylum in the gut of M. pustulata, whereby most reads belonged to the class of Gammaproteobacteria. The dominant bacterial genera were determined as Enterobacter, Acinetobacter, Enterococcus, Klebsiella, and Pseudomonas. In addition, our cultivation attempts led to successful isolation of members of Klebsiella and Enterococcus. The subject of this study could be explored further to find the potential roles of these microbiota in the gut of the specific beetles and their bioprospects.","PeriodicalId":16255,"journal":{"name":"Journal of Mathematical and Fundamental Sciences","volume":null,"pages":null},"PeriodicalIF":0.7,"publicationDate":"2022-10-18","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"81780192","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The model accommodates two types of interventions, namely infection detection and ground spraying. The dynamical system properties were thoroughly investigated to show the existence of equilibrium points, backward bifurcation, and how they are related to the basic reproduction number. We found that there is a chance that AAT may die out from the population if the basic reproduction number is smaller than one. However, the possible existence of backward bifurcation implies a condition where we may have a stable endemic equilibrium, even when the basic reproduction number is smaller than one. Hence, the basic reproduction number is no longer sufficient to guarantee the disappearance of AAT from the population. Our sensitivity analysis on the basic reproduction number showed that the interventions of infection detection and ground spraying have good potential to eradicate AAT from the population. To analyze the most effective intervention as time-dependent variable, we reconstructed our model as an optimal control problem. Numerical simulations on various scenarios were conducted for the optimal control problem. Cost-effectiveness analysis using the Average Cost-Effectiveness Ratio (ACER) and the Incremental Cost-Effectiveness Ratio (ICER) methods was performed. From the cost-effectiveness analysis, we found that ground spraying is the most cost-effective intervention to combat the spread of AAT in white rhino populations.","PeriodicalId":16255,"journal":{"name":"Journal of Mathematical and Fundamental Sciences","volume":null,"pages":null},"PeriodicalIF":0.7,"publicationDate":"2022-09-12","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"84540019","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

There is an existing subsurface geological model that is based on geophysical data. It is limited to the regional crustal scale and has a relatively low spatial resolution. The objective of our study was to delineate the sedimentary basin configuration of the area, both laterally and vertically, based on gravity data. Spectral analysis of the Bouguer anomaly allowed for anomaly enhancement by wavenumber domain filtering. Two main basins were identified from elongated low gravity anomalies that follow a SW-NE trend, i.e., the Central Basin and the Atambua Basin. The 2½D gravity modeling of selected profiles perpendicular to the regional structural direction revealed the sedimentary fills of the basins and the basement based on their densities. The Bobonaro mélanges and Viqueque sequences dominate and overlay the syn-rift (Kekneno sequences) and post-rift (Kolbano sequences) with varying lithology. These para-autochthon sediments are dominated by shale and carbonaceous rocks. Their respective thicknesses and depths of burial imply the possibility of hydrocarbon generation. The underlying basement may be associated with the Australian crust protruding from the south.","PeriodicalId":16255,"journal":{"name":"Journal of Mathematical and Fundamental Sciences","volume":null,"pages":null},"PeriodicalIF":0.7,"publicationDate":"2022-09-02","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"86341345","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The aim of this study was to invert Time Domain Electromagnetic (TDEM) data using the Flower Pollination Algorithm (FPA) as a new inversion scheme technique. FPA was originally inspired by the fertilization process of flowers, in which pollen transfer grains from male flowers to female flowers. The modeling of TDEM data was done by combining the FPA and elitism (eFPA) techniques. The applicability was tested on forward modeling data and observed data in MATLAB 2017a. In testing the algorithm, we used a model from homogeneous half space to a multi-layer model using different parameters (resistivity and thickness). In addition, in the inversion process, we used field data with various starting model approaches. Based on the results of the TDEM data modeling, FPA and eFPA can both be applied as algorithmic techniques for inversion modeling of TDEM data. The eFPA technique gave better results than FPA.","PeriodicalId":16255,"journal":{"name":"Journal of Mathematical and Fundamental Sciences","volume":null,"pages":null},"PeriodicalIF":0.7,"publicationDate":"2022-08-30","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"73195978","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Free radicals are the major cause of reactive oxygen species (ROS) generation, which affects the balance of the liver metabolism. Piper crocatum, commonly called red betel, is widely used in ethnomedicine because it has biological capabilities, including antioxidant and anti-inflammatory properties. The present study was undertaken to examine the possibility of hepaprotective activity of red betel extract on acetaminophen (APAP)-induced HepG2 cells as the cirrhosis in vitro model. Assessment of red betel extract (RBE) was performed using the colorimetric method for the quantification of LDH, AST, and ALT. As preliminary study, a cytotoxicity  assay was performed at various RBE concentrations (100.00; 50.00; 25.00; 12.50; 6.25; 3.13 μg/ml) using an MTS assay to decide the safe concentration based on cell viability. The RBE treatment did not affect cell viability, even at a high concentration (100 µg/ml, p > 0.05). RBE at concentrations 25 and 100 µg/ml successfully reduced LDH, AST, and ALT activities in the hepatotoxic model in a dose-dependent manner. In the APAP-induced hepatotoxicity model, RBE positively indicated a hepatoprotective effect and cell amelioration by a decrease in hepatotoxic markers.","PeriodicalId":16255,"journal":{"name":"Journal of Mathematical and Fundamental Sciences","volume":null,"pages":null},"PeriodicalIF":0.7,"publicationDate":"2022-06-21","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"84859565","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
