Phenolic compounds in this by-product (ultrafiltration permeate of protein purification step) were almost exclusively CGA (mainly 5-CQA isomer). Five different macroporous resins including XAD4, XAD7, XAD16, XAD1180, and HP20 were screened for CGA capture. XAD16 had the best massic adsorption capacities (15.32 ± 0.04 mg/g), while XAD7 had a better surface adsorption capacity (0.027 ± 0.00146 mg/m²). CGA adsorption on both resins followed the pseudo-second-order kinetic model with a similar intra diffusional pattern. Adsorption isotherms of the two resins better fitted the Langmuir model with Qmax for XAD7 and XAD16 of 0.054 and 0.040 (mg/m²), respectively. The adsorption process of phenolic compounds revealed to be exothermic, physical adsorption, and spontaneous. Better adsorption results were observed at 25°C. Maximal CGA desorption ratio was observed from 70% (v/v) ethanol. The high values were reached with both the resins (88.09 ± 0.13 and 86.16 ± 0.32% for XAD7 and XAD16, respectively). The CGA purity in the desorption phase was surprisingly high (77.56 ± 0.99% and 74.59 ± 0.12% for XAD7 and XAD16, respectively).

Quantification of acetaminophen in pre-term and term neonates and small children requires the availability of highly sensitive assays in small volume blood samples. We developed and validated an LC-MS/MS assay for the quantification of acetaminophen in human whole blood. The drug molecule was extracted from whole blood samples followed by protein precipitation extraction technique followed by LC-MS/MS analysis. An excellent analyte separation was achieved by using a phenomenex Gemini® C18 column (50× 3.0 mm, 3 μm) using mobile phase A-Water:Formic Acid (100:0.1) and Acetonitrile:Formic acid (100:0.1) as mobile phase B. With 3.40 minutes as analytical run time, this analytical method demonstrated linearity between 50.0 to 50,000 ng/mL and the achieved correlation coefficient (r2) was greater than 0.9996. Accuracy, precision, matrix effect and robustness were all acceptable according to FDA guidelines. In addition, acetaminophen was stable up to 179 days at both -20°C and -70°C temperatures. Hence, the acquired results proved that the LC-MS/MS was precise and accurate for quantification of acetaminophen in whole blood, and eventually this method can be applied to micro-sampling technique which is the recent trend towards sample miniaturization, collection of the sample from home and sending the devices for laboratory testing. The main aim of this study is to reduce the time consumption and to develop and validate a less expensive method by using HPLC. The chromatographic separation of Dolutegravir and its related impurities is carried out by using C8 column (150 × 4.6 mm), 5µm with 0.1% trifluoroacetic acid in water as mobile phase A, methanol as mobile phase B. The flow rate is 1.0 mL/min with gradient elution mode and the wave length for detection is 240 nm (UV detector). The developed method was validated and proved that the method was specific, accurate and precise as per ICH. The system suitability criteria found to be within the limits. The limit of detection and limit of quantification demonstrate that the method is sensitive. The linearity curve was found to be linear and the correlation coefficient obtained is not less than 0.998. The average percentage recoveries of impurities were in the range of 97 to 101%. The proposed method was found to be suitable and accurate for quantitative determination of impurities in Dolutegravir drug substance.

Paroxetine, a unique small molecule with a single secondary amine, posed significant challenges in the development of reproducible chromatography. Substantial retention time shift was observed for paroxetine using acidic mobile phases with either new or used columns. Buffer strength played an essential role in reducing the observed column-to-column retention time variation. A 96-well plate-based liquid-liquid extraction was developed. The method was validated and used to support clinical studies for the measurement of paroxetine in human plasma in the range of 0.250-50.0 ng/mL using paroxetine-d6 as its internal standard. We at the University of Central \u0000Lancashire (UCLan) proposed in a previous publication a sequential chromatographic separation process, Alternative \u0000Reprocessing Technology (ART) for Fission Products (FPs) and Minor Actinides (MAs) separation from spent \u0000fuel dissolver liquor, as an improvement to/replacement for PUREX (Plutonium Uranium Redox Extraction). This \u0000publication addresses the removal of one particular fission product, cesium, and its impact on waste management, \u0000and down-stream PUREX operations. Although our proposed process is still in its infancy, its impact on the PUREX \u0000process could be significant, with major gains in the separation circuit, waste management of High Level Waste \u0000(HLW) and subsequently waste disposal to and design of a geological repository. Therefore, the study of the levels of this protein or its gene could be useful for the diagnosis of these pathologies, in addition to other uses. The objective of this work was the development of a new detection method of the COL3A1 gene based on the quantification of mRNA in real time. 10 healthy men volunteers were injected intravenously for 3 consecutive days with 0.026 mg-1kg-1day-1person-1 of rhGH, and serum and blood samples were collected for 7 days at different times. mRNA extraction was performed after lymphocyte purification following standard protocols, and samples were quantified by quantitative real-time PCR using TaqMan probes designed specifically for this gene. The results showed an increase in COL3A1 expression that was maintained during 7 days of sampling after the last administration, observing more sensitive and robust results when compared to quantification of the PIIINP protein in serum from the same individuals. After the trial set-up, a partial validation was carried out in which parameters such as linearity, inter and intra-assay variation, influence of sex and age, stability of blood samples storage at 4°C and amount of blood sample, were studied.

Lactic acid, malic acid and the hydroxy analog of methionine (HMB) are important members of the AHA group. The chemically synthesized formulations of alpha hydroxy acids are a racemic mixture comprising of 50% L and 50% D forms. It is a well-documented fact that enantiomeric forms of chemicals can possess different activities. Enantio purity of chemicals is important in their use in pharmaceutical applications and it may play a significant role in nutritional and cosmetic applications as well. However, this role has not been fully documented because of the lack of enantio pure HMB and other AHAs. In this work, papain and chymotrypsin catalyzed peptide capping was evaluated for obtaining enantio pure alpha hydroxy acids. Chymotrypsin catalyzed peptide capping was found to be a very fast and efficient tool to obtain enantiopure HMB. The enantio purity of products was evaluated with Reverse Phase liquid Chromatography and Chiral Liquid Chromatography-Mass Spectrometry. The results indicate that enantio-purity of greater than 95% was obtained in less than ~30 minutes with chymotrypsin catalyzed capping HMB capping reaction.
