In this study, the effects of hesperetin on seizures behavior and its function on total antioxidant capacity and lipid peroxidation has been investigated in pentylenetetrazol (PTZ)-induced seizures model. METHODS: In this experimental study, thirty-five NMRI mice were divided into five experimental groups (n=7) as control, saline and hesperetin at doses of 10, 20 or 50 mg/kg. Animals received orally the related interventions for 7 days. On day 7, 30 minutes after oral gavage, convulsion was induced by single intraperitoneal (i.p.) injection of PTZ at dose of 60 mg/kg. After recording of convulsion behaviors including latency to myoclonic jerks, latency and duration of generalized tonic-clonic seizures, time to death, measuring of Ferric Reducing Antioxidant Power (FRAP) and Thiobarbituric acid reactive substances (TBARS) was carried out in hippocampus tissues. FINDINGS: Pretreatment with hesperetin at dose of 50 mg/kg significantly increased the latency of myoclonic jerks (hesperetin 50: 22±3.35 s, p=0.032) and generalized tonic-clonic seizures (hesperetin 10: 1±21.48 s, p=0.0003, hesperetin 20: 35.2±83.6 s, 0.001, hesperetin 50: 34.5±2.30 s, p=0.004). The use of hespertin at dose of 10 mg/kg significantly reduced TBARS values compared to saline (p<0.003) and doses of 20 and 50 mg/kg hespertin (p<0.0001). Any significant difference in FRAP levels was not observed between different experimental groups. Recently, a new perspective has been developed to better utilize stem cells using nanofiber scaffolds. The present study was conducted to investigate the effect of polycaprolactone/gelatin (PCL/GT) nanofiber scaffold on the therapeutic function of hematopoietic stem cells. METHODS: In this experimental study, 15 male BALB / c mice were divided into three groups of five, including the control group, the group receiving stem cells in the wound site (cell group) and the group receiving cell + PCL nanofiber scaffold. PCL/GT nanofiber scaffold was prepared by electrotherapy. After hematoxylin and eosin staining, the parameters of epidermal repair and hair follicle formation in the wound site were evaluated by fluorescence microscope and Image J and SPSS programs. FINDINGS: On day 28 after transplantation, the highest and lowest epidermal thicknesses were observed in the cell + scaffold group and control group, which were 10.5±0.3 and 27.3±0.9 μm, respectively, which was significant (p<0.05). Moreover, the highest and lowest number of hair follicles were observed in the cell + scaffold group and control group, respectively; 5.2±0.2 and 4.2±0.3. The aim of this study was to evaluation the effect of deprivation of maternal care on LTP induction in neurons of hippocampal CA1 region in morphine-dependent rats. METHODS: This experimental study was conducted among 40 45-day-old male rats in control group, morphinedependent group, and 3 groups of rats that were deprived of treatment for one, two and three weeks after birth for 3 hours daily. Except for the control group, the rest of the groups received 10 mg/kg body weight morphine sulfate subcutaneously every 12 hours for 10 days. On the eleventh day, the symptoms of deprivation syndrome were investigated by the Gellert-Holtzman method, and on the following day, the synaptic plasticity of neurons in CA1 region was studied. FINDINGS: The Gellert-Holtzman score in the morphine-dependent group was 14.98±4.16 and increased to 31.79±5.12 in the group that was deprived of maternal treatment for 3 weeks (p<0.001). Although morphine dependence did not affect basic responses of CA1 region neurons and LTP induction, deprivation of maternal care reduced the range of basic responses from 1.01±0.04 in the morphine-dependent group to 0.68±0.09 mV in the group that was deprived of maternal care for 3 weeks (p<0.001) and prevented LTP induction (p<0.001) in a time-dependent manner. The correlation between severe tissue eosinophilia with optimal and undesirable prognosis, or even an ineffectiveness effect was related. The aim of the present study was to investigate the correlation between tissue eosinophilia and prognosis in patients with head and neck squamous cell carcinoma. METHODS: In this cross-sectional study, 33 patients with squamous cell carcinoma of the pathology department of Shahid Beheshti hospital of University of Babol University of Medical Sciences (age, sex, tumor site) were collected and histopathologic in the vascular and nervous invasions was studied. Numbers of eosinophils (eos.) were counted in 15 successive fields at invasive fronts of HNCSCCs (area of 15 fields=1.2mm) at×400 magnification with optical microscope. The correlation between the mean number of eosinophils per Mm2 and the severity of eosinophilia (mild and severe) and clinical-histopathologic factors was analyzed. FINDINGS: Mean eos/mm was 99.1913±104.39897. Overall survival rate, disease-specific survival rate and diseasefree survival rate were %72.72, %61.53% and %84.84, respectively. There was inverse significant correlation between disease-free survival with eos/mm (p=0.043). There were no significant correlation between other clinicalhistopathologic factors with mean eos/ mm.There were inverse significant correlations between 5-year overall survival rate and 5-year disease-free survival rate with severity of tissue eosinophilia (p=0.02 and 0.013 respectively).There were not significant correlations between 5-year disease-specific survival and other factors with severity of tissue eosinophilia. ABSTRACT BACKGROUND AND OBJECTIVE: Candida albicans infection is a problem of growing clinical importance, particularly in immunocompromised populations. This study aimed to investigate the inhibitory effect of carvacrol on the hyphae formation and expression of C. albicans HWP1. METHODS: This cross-sectional study was done over a 6-month period in 2016-2017. Vaginal, mouth and skin surface swabs were obtained from immunocompromised patients. Colonizing clinical isolates of C. albicans were identified and drug susceptible isolates detected using WHONET software. The susceptibility test for carvacrol (range 6.25–300 μg/ml) were carried out with a broth microdilution according to the CLSI guidelines against drug susceptible C. albicans. The time kill assay of carvacrol (range 2 × MIC to 1⁄4 × MIC) was determined. Hyphae inhibition was evaluated by light microscopy. We determined the expression levels of HWP1 implicated in hyphae formation of C. albicans ATCC 14053 cells by quantitative RT-PCR. FINDINGS: Ten colonizing clinical isolates of drug susceptible C. albicans were identified. Carvacrol was inhibited the growth of all drugs susceptible isolates of C. albicans (MIC range, 25-200 μg/ml). Time kill curve assay demonstrated that carvacrol could significantly inhibit the growth of C. albicans (p≤0.05). Carvacrol efficiently prevented hyphae formation in drug susceptible C. albicans. The expression levels of HWP1 gene were down-regulated by 1.82and 1.62fold at concentrations of 2 × MIC and 1×MIC of carvacrol, respectively.
