The judgment to assess whether an examinee recognizes crime-relevant information is based on the diŠerence in autonomic responses between the crime-relevant information and the crime-irrelevant information. The present study examined the within-questionnaire consistency of the diŠerential physiological responses, which is a key point in the above judgment. The mock theft CIT data ob-tained from 167 participants were modeled into three possible models, and Bayesian estimation of parameters and model selection were performed. Results of model selection implied that participants in the encoding group were not clearly divided into high consistency responders and non-responders. Bayesian estimates of parameters revealed that the within-questionnaire consistency of the diŠerential physiological responses had diŠerent characteristics among the physiological measures. Regardless of the high toxicity, they have been used in some cases such as attempted murder cases due to the easy accessibility of castor beans. For these cases, detection of ricin is important for case control and proper remediation or decontamination. In this work, we have investigated the applicability of Bio-Threat Alert (BTA) test strips for the detection of ricin in beverages. By using BTA test strips, standard ricin protein in various beverages as low as 0.1 mg/mL could be detected by visual detection or by Guardian BTA Test Strip Reader. The calibration curves for ricin in tomato juice and cafe au lait were constructed using sample values obtained from Guardian BTA Test Strip Reader, which is the intensity of the color developed by the reaction of ricin and antibody. At high concentrations, the response seemed to be saturated. From the beverages added with mashed castor beans, ricin could also be detected by BTA test strips. Today, quantitative real-time PCR ( qPCR ) targeting the autosomal D17Z1 region, which cannot distinguish between male and female DNA, is the main method for forensic human DNA quantiˆcation for the Japanese police. For more e‹cient testing Y-STR type, we examined the forensic evaluation ( sensitivity, repeatability, speciˆcity, inhibi-tion, comparison with commercial qPCR reagent and simulated mixture study ) of qPCR assays targeting Y-chromosomal Sex determining region Y ( SRY ) and DYZ5 regions. We evaluated the eŠectiveness of simple and practical male DNA quantiˆcation methods in male and female mixture samples. The sensitivity of SRY and DYZ5 assays were 0.015 ng / m L and 0.0015 ng / m L. Both assays could quantify male DNA in the presence of high concentrations of female DNA. The SRY assay had the same sensitivity as the commercial qPCR reagent, but had more resistant to inhibitors. The DYZ5 assay tended to have higher DNA concentration than the commercial qPCR reagent and the SRY assay, but could detect trace amounts of male DNA. The reason for this is that DYZ5 region is multi-copy locus and there are individual diŠerences in the copy number. The DYZ5 assay was considered to be one of the eŠective methods for proving the presence of male DNA in the sample. In this study, we examined the STK sheet and STK spray for their speciˆcity to detect human semen, sensitivity, eŠects on DNA, and response to old semen samples. Additionally, STK sheet was examined for optimal crimping time depending on the type of fabric. STK spray was examined through assessing the eŠect of ultraviolet ( UV ) radiation after spraying. As a result of this study, STK sheet and STK spray were found to ex-hibit high speciˆcity for the detection of human semen. These reagents did not aŠect the yield of extracted DNA or its degradation. STK sheet and STK spray can detect semen up to 1 / 25 th and 1 / 100 th dilutions, respectively. Both STK sheet and STK spray can react with semen samples left at room temperature for over a year. We found that the crimping time on STK sheet diŠers depending on the type of fabric, and the exposure to UV radiation for an hour after using STK spray does not aŠect DNA degradation.
