Kidney regeneration in Aphanius furcatus (Teleostei: Aphaniidae) after damage induced by gentamicin and evaluation of vitamin C in toxicity reduction 庆大霉素致肾损伤后的肾再生及维生素C的降毒作用

The kidney regeneration of the killifish Aphanius furcatus was studied after gentamicin-induced oxidative stress. In addition, we evaluated the possible role of vitamin C as an antioxidant in the reduction of renal toxicity. A total of 24 specimens (3.5-4cm in total length) were divided into four groups: In group-I two fish specimens treated with 7.5µl of normal saline injection; in group-II, 10 specimens received 10µg/g body weight (b.w) gentamicin; in group-III, two fish were treated with totally 60µg/g b.w vitamin C; and in group-IV, 10 specimens received both 10µg/g gentamicin and 60µg/g b.w vitamin C. The kidney tissues of group-I, III and IV were sampled during the nephrotoxicity phase (i.e., 10 hours post injection (hpi) to 5 days post injection (dpi)) and kidney tissues of group-II sampled daily for 10 dpi until regeneration completed. The kidney regeneration of A. furcatus through nephron neogenesis was detected by newly formed nephron on 8 dpi, and it is completed on 10 dpi. Our histological results in gentamicin-induced oxidative stress (group-II) clearly showed that the most severe damage is occurred in the kidney tissue within 1-4 dpi. However, in the specimens that antioxidant therapy supplied with vitamin C (group-IV), damage effect of gentamicin has remarkably repressed within these days. The outcome of this study revealed that administration of vitamin C is effective in reduction of gentamicin-induced nephrotoxicity in A. furcatus . Vitamin C reduces the toxicity effect of gentamicin by reduction of free radicals and reactive oxygen species (ROS) through its antioxidant activity. This study evaluated the health status of Catla catla, Labeo rohita, Cirrhinus mrigala and Labeo bata cultured in West Bengal, India by determining the Health Assessment Index (HAI) and coefficient of variation (CV) of HAI values of each species as well as the superoxide anion production. The annual mean HAI value was the highest for L. rohita (77.51) and least for L. bata (55.30). The highest and lowest CV of HAI values were recorded for L. bata (95.80) and C. mrigala (52.55), respectively . The HAI values correlated negatively ( P <0.05) with temperature. The HAI distinguished successfully among the species, seasons and sampling areas on the grounds of temperature and water quality, with higher index values recorded in carps grown in poorly managed ponds. Labeo bata exhibited higher immune responsiveness compared to other carps. Labeo rohita was the most susceptible to diseases. The results on the high production of oxygen radicals by phagocytic cells of L. bata together with low HAI and high CV values suggested that this species may resist biotic and abiotic stressors well under culture conditions. In this study, otolith (Sagitta) morphology of four fish species viz. Pomatomus saltatrix , Sarpa salpa , Trachurus trachurus , and Belone belone was investigated. The length (mm), width (mm), area (mm 2 ) and perimeter (mm) measurements of the otoliths were made and four different shape indices, including form factor (F F ), aspect ratio (A R ), roundness (R D ) and circularity (C I ) were calculated. In addition, relationships between total fish length and otolith morphology were investigated. Measurements of sagittal otoliths of four species were obtained by image analysis using tri-ocular microscope. Based on the results, the morphometric measurements and four examined shape factors of otolith varied among species. Coefficients of correlations (r) between total fish length and otolith morphology and otolith length-otolith morphology were generally highly significant ( P <0.05) for studied fishes. The results show that the shape indices were significantly different in analyzed species even they indicated a similar pattern with maximal otolith length. Otolith shape indices, including Form Factor (FF), Circularity (C), Roundness (RO), Rectangularity (RE), Aspect Ratio (AR) and Ellipticity (E) were used for otolith shape analyses and multivariate analyzes (Canonical Discriminant Analysis) were used to assess interspecies variations. The FF and C were found statistically significant for lapillus, but not for asteriscus otoliths in the four habitats. Furthermore, RO was similar for all localities for both asteriscus and lapillus. CDA results showed that 66.5% of the chub individuals were correctly classified. Age, growth, sex ratio, spawning season, fecundity, gonado-somatic, modified gonado-somatic and Dobriyal indices were estimated. The sex ratio (F: M) was obtained as 1.00:2.63, and the percentage of males was greater than females. The mature males and females were longer than 88 and 148 mm in total length, and 1 + and 2 + in age, respectively. The spawning of C. razii took place from late May to late August when the water temperature was between 19.7 and 22.8 ◦ C. At the beginning of the reproduction period, the average gonado-somatic index values were 10.0% ranging from 4.03% to 15.90% in ripe females. The estimated gonadal indices showed that the ripe males were ready to spawn earlier than females. The average egg diameter was 0.99±0.65, ranging from 0.2 to 2.3 mm. The averages absolute and relative fecundity were 4386±2174 and 66.3±37.8, respectively. Two species, Sabanejewia aurata and Sabanejewia caspia are recognized and diagnoses are presented for both species. They are distinguishable through body pattern, length of caudal peduncle, snout and post orbital distance, shape of scale and suborbital spine, caudal complex skeleton and also the number of fixed, diagnostic nucleotide substitutions. After that, there are very rare reports on this particular species especially on its morphometry after McCosker (1972). This paper extends the range of A. concolor McCosker, 1972 from eastern Andaman Sea and Northern Territory, Australia to Indian waters.
