This study aimed to investigate the therapeutic effects of applying GTE as a topical agent for oral leukoplakia and the subsequent histopathological changes. We examined the absorption of GTE in rat oral mucosa. In addition, we examined 30 patients diagnosed with oral leukoplakia between April 2009 to March 2011. We administered an oral ointment composed of 1% GTE kneaded into a base material. A single dose(0.5 g) was applied to the affected area thrice a day for 90 days. Macroscopic observations and serum catechin levels were examined before, during, and after administration. The comparative histological effects and Ki-67-positive cell rates were examined using hematoxylin and eosin(H-E)staining and Ki-67 immunohistochemical staining. Nine patients in the treatment group exhibited disappearance, shrinkage, or thinning of the leukoplakia. After administration, effective cases exhibited histopathological normalization of the mucosa, and Ki-67 immunohistochemical staining suggested normalization of epithelial growth dynamics. GTE ointment applied to the dorsum of the rat tongues were examined for tissue permeability of GTE in frozen sagittal sections. Imaging of the rat tongue sections showed GTE penetrating the epithelium over time. Together, our results indicate that topical administration of GTE may be effective against oral leukoplakia. verify its inhibitory effect on cell growth in human oral squamous cell carcinoma lines(HSC - 4, Ca9 - 22). After confirming a decrease in cell activity, we examined its application as a therapeutic agent for oral leukoplakia. immunohistochemical suggested the GTE ointment into oral mucosa acts on - related factors(5). The objective of this study was to evaluate the wound healing ability of oral mucosa and skin and to compare differences in their healing responses. Human gingival fibroblasts(HGF) and human skin fibroblasts(HSF)were used to characterize cell migration ability in wound healing assays. In oral mucosa and skin wound healing mouse models, mRNA levels of several inflammatory markers, IL-1β, IL-6 and IL-17, were examined in tissues collected around the wounds using RT-PCR. The healing ability of the oral mucosa as well as the skin was observed using immunohistochemistry at day 3 post-wound healing, and the expression level of IL-1β was also compared in oral mucosa and skin tissues collected around the wounds. Our findings showed that the cell migration of HSF cells was slower than HGF cells. In vivo experiments suggested that wounding induced a more significant up-regulation of IL-1β, IL-6 and IL-17 in healing oral mucosa compared with healing skin tissue. Tissue repair in oral mucosa wounds was more rap-id that in skin wounds. The objective of this study was to investigate the possibility of increasing the lateral expansion limit in orthodontic treatment by induce bone formation in the buccal alveolar bone through local application of fluvastatin(statins). The effectiveness of statins was evaluated by using MC3T3-E1 cells to cell proliferation ability, osteoblast differentiation ability, and mRNA levels of mineralization maker genes. Further, we mounted a lateral enlargement device to the maxillary molar region of a 10-week-old male C57BL/6J mouse. After 14 days, we used specimens of the first molar alveolar bone to carry out width diameter measurement, measurement of the amount of root exposure, and histological evaluation of specimen sections. For the statin group, we injected optimal concentration statins into the gingiva. As a result, mRNA levels of mineralization maker genes expressions in the MC3T3-E1 cells increased in the administered 10 nM statins compared to the control. Further, the calcification ability of the 10 nM statins increased compared to the control. In vivo, there was no significant difference in the amount of lateral enlargement, but the amount of root exposure was significantly less, and the amount of bone added to the buccal alveolar bone was higher in the statin group than in the control group. Further, most patients discontinue wearing an oral appliance(OA)for improving respiration due to nasal obstruction. We aimed to develop a nasal breathing stimulator(NBS)for use in combination with an OA to promote nasal breathing. Methods: Thirteen OSA patients wore a NBS, Max-Air Nose Corns ® , Mute with hole ® , which expands the nasal cavity from the inside, and Breathe Right ® , which expands the nasal cavity from the outside, and compared the resultant changes in respiratory measures with those when a device was not used. The inspiratory flow rate(peak nasal inspiratory flow: PNIF)and visual analogue scale(VAS)scores were measured under five conditions. Furthermore, PNIF was measured under four conditions: OA(-)NBS(-), OA(+)NBS(-), OA(-)NBS(+), and OA(+)NBS(+), and a sleep test was performed. Results: PNIF was significantly higher when wearing an NBS compared with the other four devices(p < 0.001). In addition, PNIF was significantly higher in OA(+)NBS(+) than that in the other three conditions(p < 0.001), and OA(+)NBS(+)improved the respiratory disturbance index(p < 0.001), lowest SpO 2( p < 0.001), and Epworth sleepiness scale(ESS)score(p < 0.001)on the sleep test.
