Asmaz","doi":"10.30782/ULUVFD.415592","DOIUrl":"https://doi.org/10.30782/ULUVFD.415592","url":null,"abstract":"The objective of this study was to investigate the effect of Tamoxifen (TAM) treatment on epidermal growth factor receptor (EGFR) expression in the pubertal mice ovary. In this study, 80 female mice (8 week-old) were used. Animals divided four groups; non-injected (control A), injected with TAM’s vehicle solution (control B). The mice in groups TAM 0.5 and TAM 1.5 were treated with tamoxifen at a dose 0.5 and 1.5 mg/mouse/day respectively. TAM was dissolved with 10% ethanol: 90% corn oil. Mice were given daily subcutaneously injections for 5 days. Ovarian sections were immunostained with EGFR antibody and triple staining for examine the general structure. We observed that follicular atresia was increased, follicular cysts were formed in stroma, and interstitial cells were increased with TAM dose treatment. In the present study, there were no significant differences in the EGFR expression in mouse ovary of all groups. EGFR expression was not determined in the granulosa cells. While strong EGFR immunoreaction was observed in the interstitial cells and oocyte cytoplasma, weak EGFR immunoreactions was observed in theca cells of follicles. Oglaklarda hastalik tablosu ‘Floppy Kid Disease’, sigirlarda ‘Ruminal Drinkers’ son olarak kuzularda ‘Drunken Lamb Sendromu’ olarak adlandirilmistir. Bu uc hastaligin etiyopatogenezinde, rumende veya barsak florasinda ani ve cok miktarda bakteriyel fermantasyon sonucu artan D-laktik asit’in sistemik dolasima katilmasi ve anyon acikli metabolik asidozun olusumu rol almaktadir. D-laktik asidoz, depresyon, dehidrasyon ve norolojik belirtilerin olusmasina yol acabilmekte ve tanisi zor konulmaktadir. Keci yetistiriciliginde dol, sut ve et verimi onem tasir. Dolverimi tum diger verimlerin temeli olup et verimini dogrudan etkilemektedir. Ureme organlarinin gelismesi ile genc hayvanlarda ureme faaliyetlerinin basladigi doneme puberta denir. Bu donemde genc erkeklerin ejakulatinda ilk spermatozoid, genc disilerde ilk ovulasyon meydana gelir fakat hayvanlar cinsel olgunluga tam ulasmis degildir. Cinsel faliyetlerin optimum duzeye ciktigi doneme ise cinsel olgunluk cagi denir ve ayni zamanda bu doneme “damizlikta ilk kullanma cagi” anlaminada gelir. Kecilerde yasam boyu verim performansinin belirlenmesinde onemli faktorlerden birisi puberta yasidir. Besleme, canli agirlik, irk, dogum mevsimi ve buyume orani gibi bircok faktorun puberta yasini etkiledigi bilinmektedir. Fertility traits are important for economic values in animal breeding. However these traits exhibit only in females and they are determined in the later stages of lifes. Hence the genetic perspective is more effective way to confirm the fertility capacities early period of life time. Fecundity genes such as BMP15 regulate the fertility traits by increasing the ovulation rate due to the occuring mutations. Despite some prolific breed were investigated for FecX B mutation; the researches carried out to determine that mutation in Chios sheep breed are limited. The objective of the current study was to estimate the the genotype frequencies of FecX B mutation of BMP15 gene in prolific Chios sheep. We investigated the FecX B mutation by PCR-RFLP method in Chios sheep (n=77). According to result, the investigated Chios breed was found monomorphic for FecX B mutation. All individuals were digested by DdeI restriction enzyme and showed wild-type genotype and did not carry FecX B mutation nin present study. In conclusion it’s thought that the high prolificacy in Chios sheep may be based on another region of BMP15 gene or different major gene. Bakteriyel etkenlerin teshisine yonelik cok sayida calisma yapilmis, ancak bakterilerin kesin teshisinin detayli biyokimyasal testlerle desteklenmesi gerektigi belirtilmistir. Bu amacla, calismamizda Y. ruckeri ve L. garvieae etkenlerinin ayrintili biyokimyasal farkliliklarinin belirlenmesi ve hizli teshis kitleri kullanilarak identifikasyonun kisa bir surede yapilmasi amaclanmistir. Turkiye’nin farkli bolgelerinden izole edilmis olan Y. ruckeri ve L. garvieae izolatlarindan 30’ar izolat incelenmis, konvansiyonel mikrobiyolojik testler ve API RAPID test kitleri ile biyokimyasal ozellikleri belirlenmistir. Biyokimyasal ozellikler acisindan Y. ruckeri izolatlarinin L. garvieae izolatlarina gore daha heterojen oldugu belirlenmistir. Izolatlar arasinda biyokimyasal farklilik olmasina ragmen, tum izolatlar hizli teshis kitleri kullanilarak 5-7 saat icerisinde basarili bir sekilde identifiye edilmistir. Hayvanda dogumu izleyen donemde rektal muayene yapilmak istendiginde rektum cevresinde zamanla sayi ve hacimleri artan kitleler gozlendi. En buyugu 6,5 cm capinda yuvarlak ve 4x3x1,4cm buyuklugunde oval, cesitli buyuklukte 14 adet kati esnek kivamda, beyaz renkli kitlelere rastlandi. Kitlelerin bazilari deri ve cogunlukla mukoza ile kapli oldugu dikkati cekti. %10’luk tamponlu formaldehit ile tespit edilerek parafine gomulen ve 5 µm kalinliginda kesitler alinan dokular, Hematoksilen-Eosin (H-E) ve Van Gieson ile ayrica immunohistokimyasal (IHK) olarak sitokeratin ve vimentin ile boyandi. H-E kesitli dokulardaki tumoral kitlenin tipik anafor tarzinda yerlesmis fibroblast ve fibrositlerden olustugu, yapilan IHK boyamada ise vimentin pozitif ve sitokeratin negatif olarak gozlendi. Bu olgudaki gozlenen makroskobik, mikroskobik ve IHK’sal bulgular kaudal rektumda fibromatozis olgusu ile uyumlu bulunmustur. Anahtar kelimeler: Inek, rektum, fibroma Rectal fibromatosis in a Simmental cow. Abstract: In this case, it was aimed to investigate the clinical and pathological aspects of fibromatosis which make difficult rectal examination due to multiple tumoral masses observed in a 4-year-old Simmental–breed at rectum. Period of after the birth, the masses were seen around the rectal area of the cow when rectal examination was carried out. The number and volume of masses increased with time.The shapes of masses were round and oval and the largest size were 6.5 cm in diameter and 4 x 3 x 1.4 cm respectively. Totally 14 masses, covered with skin and mucosal membrane were found. Cut surface were whitish colored and solid-flexible in consistency. The tissues were fixed with 10% buffered formaldehyde and embedded in paraffin and sections with a thickness of 5 μm were stained with hematoxylen-eosin (H&E) and Van Gieson and immunohistochemical (IHC) with cytokeratin and vimentin.Tumor tissue in H&E sections consisted of fibroblasts and fibrocytes located in the typical herringbone pattern. Tumoral cells were vimentin-positive and cytokeratin-negative in IHC staining.Macroscopic, microscopic and IHC findings, in this case, were found to be consistent with the presence of fibromatosis in the caudal rectum.
