El-Sharkawy, Amira Mira","doi":"10.21608/jampr.2024.285984.1068","DOIUrl":"https://doi.org/10.21608/jampr.2024.285984.1068","url":null,"abstract":"The present study aimed to the in vitro investigation of the ethanol extracts of Washingtonia filifera roots (W.R), barks (W.B), leaves (W.L), fruitless bunches (W.F) and seeds (W.S) for their neuroprotective activities including the phenolic content, antioxidant activity, anti-inflammatory activity and the potentiality to protect mouse neuroblastoma (neuro-2A) cells against H 2 O 2 and A β 1 - 42 -induced neurotoxicity. Among the extracts, W.L and W.F had the highest phenolic content; 226.7 and 172.3 GAE, respectively. Consequently, W.L and W.F showed pronounced DPPH radical scavenging activity with IC 50 values of 19.95 and 30.46 µg/mL, respectively. The highest inhibitory potential of COX-2 was achieved by W.F then W.L with IC 50 values of 111.62 and 191.77 µg/mL. The most efficient extract for the neuroprotection against H 2 O 2 and A β 1 - 42 -induced neurotoxicity was W.F which had more potent activity than the positive control in A β 1 - 42 assay. S. aureus's ability to build biofilms and other virulence variables determines whether it can cause superficial or invasive infections. It can also rapidly alter its virulence and metabolic responses in various tissues. Anti-virulent medications that stop the development of biofilms and virulence factors are therefore becoming increasingly necessary. As a result, we focused on an antibiotic that had already been approved and investigated whether changing the dosage would have an impact on S. aureus virulence. Most of the studied isolates exhibited a noteworthy decrease in S. aureus biofilm and virulence factors production (protease, hemolysin, and coagulase) upon treatment with 1/4 and 1/8 MICs of ceftriaxone, according to the data. These fake preparations are usually harmful to users. So, it is always necessary to develop specific as well as sensitive analytical procedures to detect sildenafil in such preparations. HPLC that equipped with modern sensitive detectors such as diode array detector (DAD) can play a great role in solving such problems. In this work, a new HPLC method was established for the detection of sildenafil in some marketed dietary supplements. Thermo scientific ODS HYPERSIL column (250 x 4.6 mm, 5µm) was used. The mobile phase was composed of methanol and potassium di-hydrogen phosphate buffer 0.05M (pH 3.5) at a ratio of 70:30 (v/v). Triethylamine (0.3%) was added to the mobile phase. The flow rate was 1mL/min. Detection was performed at 290 nm using a diode array detector. The method showed good linearity (5.0-80.0 µg/mL). The retention time was 5.3 min. The validation parameters were evaluated as per ICH guidelines. The findings given by the developed method were compared statistically to those achieved by a reported one. There was a good statistical agreement. Compared to traditional nanocarriers, bilosomes have the advantage of being able to withstand disruption caused by physiological bile salts normally secreted in the gastrointestinal tract. In addition to nonionic surfactants and bile salts, cholesterol is one important ingredient in bilosomes composition. This work investigated the effect of cholesterol concentration on the release rate of the entrapped drug. Tamoxifen was used as a model drug that suffers from poor oral bioavailability due to poor solubility and extensive pre-systemic degradation. Bilosomes composed of Span 60, cholesterol, and bile salts were prepared. Cholesterol was used at two different concentrations of 0.4% and 0.8% w/v, producing formulations BiL1 and BiL2, respectively. The entrapment efficiency and in vitro drug release were evaluated using Franz diffusion cells. Increasing cholesterol concentration reduced drug release. The release efficiency values after 24 hours of release study were 9.7 and 6.8% for BiL1 and BiL2, respectively. This indicates that increasing cholesterol concentration increased the rigidity of the bilosomal membrane and enhanced drug encapsulation. Methods This pilot single-center retrospective cohort research study involved 25 patients with muscle-invasive bladder cancer who were admitted to the Urology and Nephrology Center (UNC), Mansoura University, Egypt between January and December 2016. The study tried to reveal the association between SNP rs11615 in ERCC1 gene and the change from baseline characteristics in a median of 96 weeks follow-up in blood, renal, and hepatic biochemical parameters. Results Univariate analysis of the association between rs11615 and post-chemotherapy changes in uric acid and hemoglobin levels showed a statistically significant relationship. Conclusion These findings hint at possible genetic influences in response to chemotherapy, necessitating further research to understand the mechanisms and potential clinical implications. Natural products and herbal medicines with accepted safety profiles are considered promising sources for discovering new medication leads. Genus Marrubium was reported to exhibit potential antiviral and antioxidant activities. Thus, this study aims to evaluate the inhibitory activity of the total methanolic extract and its soluble fractions: (petroleum ether, methylene chloride, and ethyl acetate) of the aerial parts of Marrubium alysson L . against SARS-CoV-2 main protease (M pro ). Also, they were quantitatively assayed for their phenolics and flavonoids contents, in addition to evaluating their antioxidant activity. The results showed that the petroleum ether fraction has the highest inhibitory activity against SARS-CoV-2 main protease (M pro ) enzyme with IC 50 value of 3.959±0.17 µg/mL compared to the reference tipranavir (IC 50 1.89±0.08 µg/mL). The ethyl acetate fraction showed the highest total phenolics and flavonoids contents as well as the highest antioxidant activity (IC 50 33.81±0.15 µg/mL) compared to the reference L-ascorbic acid (IC 50 30.43±0.14 µg/mL).
