Vitrification of female gametes is the most important tool for solving the strategic tasks of cellular reproductive technologies in animal husbandry, veterinary medicine and biomedicine: replication of elite individuals (cloning, transgenesis), biodiversity conservation, organ xenotransplantation. The aim of this study was to evaluate the parameters of nuclear cytoplasmic maturation (chromatin status and mitochondrial activity) and developmental competence of donor porcine oocytes subjected to intra- (IOV) or extra-ovarian (EOV) vitrification and cultured in various maturation systems. Methods. Donor pig oocytes (6–8 months old) were vitrified extra- or intraovarially using cryoprotective agents (DMSO, ethylene glycol, trehalose, sucrose). The composition of cryoprotective agents was supplemented with 0.001 % highly dispersed silica nanoparticles (HDSns). Oocytes were thawed in trehalose solutions in TS-199 with 10 % of fetal bovine serum (FBS). Oocytes were cultured for 44 hours in media supplemented with granulosa cells (106 cells per ml of medium) and/or 0.001% of HDSns at 38.5 °C, in an atmosphere of 5 % CO2. Fertilization in vitro was carried out in accordance with the recommendations presented by us earlier [17. p. 17]. The functional activity of mitochondria and the status of chromatin in oocytes were assessed by double staining using MitoTracker Orange CMTMRos and Hoechst 33258 dyes. Results. The level of matured oocytes was 61 % at EOV and 29 % at IOV, and the level of cleavage embryos in EOV was in 11 % (31 % vs. 20 %, P < 0.05) higher than at IOV when HDSns with granulosa cells were added in culture media. The maximum indicators of the functional activity of mitochondria among devitrified oocytes were in the group of EOV oocytes have cultured in media supplemented with HDSns and granulosa cells (152 ± 14.9 μA). Scientific novelty. The protocols for intra- and extra-ovarian vitrification of porcine oocytes have been modernized. Addition to cryoprotective agents and culture media of HDSns and co-culture of devitrified oocytes with granulosa cells significantly increased the fertility rates of devitrified gametes.","PeriodicalId":125083,"journal":{"name":"Agrarian Bulletin of the","volume":"109 6","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-01-23","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"139605730","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

In conditions of increasing environmental load from accumulated synthetic packaging materials in environmental objects, there is a need to develop new compositions of biodegradable films that additionally have functional properties to increase the shelf life of products. The scientific novelty of the research lies in the production of new types of biodegradable active films with the addition of protein hydrolyzate as an active component, as well as data on their properties. The purpose of the study is to establish the properties of biodegradable films based on agar with the addition of a suspension of protein hydrolyzate and CMC as active components. Research methods. The control (composed of agar and glycerin) and test samples of films (composed of agar, glycerin and a suspension of MCC and protein hydrolyzate in an amount of 5, 10 and 15 % by weight of the biocomposite) were determined for mechanical properties, microstructure, antioxidant properties, vapor permeability, solubility, and moisture absorption. Results. The introduction of a suspension into the composition of the biocomposite had a negative effect on the mechanical strength of the film – the tensile strength decreased from 9,71 MPa for the control sample to 3,35 MPa for the test sample with 15 % suspension, while the relative elongation was maximum for the sample with 10 % suspension – 54,9 %. The antioxidant properties of the films increased with the addition of the suspension: the antiradical activity of DPPH increased from 57.65 % in the control sample to 63.81 % in the test sample with 15 % suspension, and the phenol content – from 0.253 in the control sample to 0.502 mEq of gallic acid per 1 g for a prototype with 10% suspension. It was noted that the addition of the suspension had a significant effect on vapor permeability and moisture absorption. Thus, the results obtained showed the need for further research into the storage capacity of food products with different humidity levels when packaged in tested films.","PeriodicalId":125083,"journal":{"name":"Agrarian Bulletin of the","volume":"48 19","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-01-23","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"139603478","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The purpose of the study is to develop a model for the modernization of conceptual approaches to state regulation of the agro-food market of the region, which includes tools and mechanisms that eliminate imbalances in its development. The existing unilateral mechanism of state support for the agricultural sector in the face of agricultural producers does not ensure the maintenance of the total supply of food at the level of consumer demand, which, on the one hand, leads to a crisis of overproduction of certain types of food, on the other hand, does not allow to increase their economic accessibility. Methods. The instrumental and methodological apparatus of the study consisted of methods of comparative, structural-target, scenario analysis, expert assessments, cognitive modeling and others. The processing of analytical material, which ensures the reliability of calculations, was carried out using the IGLA decision support system. The scientific novelty lies in the substantiation of additional tools and mechanisms aimed at stimulating solvent demand, among which the development and implementation of a regional program of domestic food aid and a mechanism for stimulating solvent demand in the context of the regional State Program “Development of agriculture and regulation of agricultural products, raw materials and food markets” are proposed. Results. It is argued that the variety of factors affecting the functioning of the agri-food market, the high level of dynamism, multi-aspect and uncertainty of the processes taking place in it, the complexity of obtaining relevant information necessary to predict its development, allow us to attribute the agri-food market to complex poorly structured systems, the study of which by traditional methods is significantly difficult. To solve the problems of regulating the agri-food market, it is proposed to use cognitive modeling methods, which allows us to justify the need and adequacy of the use of additional tools of state regulation aimed at stimulating consumer demand.","PeriodicalId":125083,"journal":{"name":"Agrarian Bulletin of the","volume":"6 9","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-01-23","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"139604411","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The yeast Saccharomyces cerevisiae is widely used in many fermentative processes of food production. At the same time, the spent yeast biomass obtained after production processes generates significant amounts of waste. Thus, in the brewing industry residual brewer's yeast is the second largest volume of waste, which is about 3% of the volume of brewed beer. Utilization of these wastes is rather complicated and requires additional costs from the enterprises. Under these conditions, the directions of additional utilization of spent yeast mass are of high interest. One of such directions can be the use of plasmolysis of spent yeast and its further use for encapsulation of biologically active substances. Among biologically active compounds, plant polyphenols – substances with pronounced antioxidant properties – occupy an important place. Scientific novelty. This study examines dihydroquercetin, rutin and curcumin in their original and nanostructured forms. For these compounds an extensive list of pharmacological properties such as anti-inflammatory, antioxidant, capillaroprotective and others has been established. At the same time, these compounds are characterized by a low level of bioavailability. The purpose of the study was to investigate the possibility of using spent, plasmolysed cells of brewer's yeast Saccharomyces to encapsulate plant polyphenols: dihydroquercetin, rutin and curcumin, and to evaluate the effect of such an approach on the bioavailability of biologically active substances in an in vitro digestion model. Results. As a result of these studies, it was found that plasmolysed yeast cells are able to act as a “delivery system” of plant antioxidants. Using the proposed approaches, it is possible to achieve an encapsulation efficiency of approximately 57–64 %, provided that the polyphenols are ultrasonically nanostructured beforehand. Analysis of the potential bioavailability of encapsulated forms of plant antioxidants in an in vitro gastric digestion model showed that the technology of encapsulation into yeast cells allowed to ensure the preservation of plant antioxidants about 80 %. In comparison with the initial forms of biologically active substances, encapsulation allowed to provide an increase in bioavailability by about 30–40 %.","PeriodicalId":125083,"journal":{"name":"Agrarian Bulletin of the","volume":"140 25","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-01-23","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"139604532","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The purpose is to identify infradian rhythms in the seasonal dynamics of apical growth of shoots of Salix ‘Bullata’ plants and to establish the features of morphogenesis associated with the existence of endogenous (genetic and hormonal) mechanisms for the occurrence of nonlinear changes, including fluctuations in the growth characteristics of shoots. Methods. A quantitative approach was used to study the dynamics of morphogenesis processes. There was carried out a comparative research of seasonal dynamics of velocities and accelerations of apical and radial growth of Salix ‘Bullata’ plants shoots, which differ in origin and location on the plant. Results. In the seasonal dynamics of the characteristics of apical growth of branching and plant formation shoots of Salix ‘Bullata’ plants there are observed infradian rhythms, similar to those identified for other willow species. These rhythms are most pronounced for the growth accelerations (about 5 maximums within 3.5 months). Seasonal dynamics of apical growth accelerations of two types of shoots are correlated and almost synchronous. Seasonal dynamics of speeds and accelerations of radial growth of the studied types of shoots are similar and differ from the seasonal dynamics of apical growth. The weak dependence of growth characteristics on weather conditions confirms the concept of the endogenous nature of the growth of plant shoots periodicity. Scientific novelty. The results complement the existing information on biological rhythms and processes of morphogenesis in woody plants. The growth of shoots of willows and, possibly, other species of woody plants has an oscillatory character. Seasonal dynamics of apical and radial growth are different. The occurrence of oscillations in the rate of shoot apical growth is mainly due to the processes of apical meristem development itself. Apparently, the oscillatory nature of shoot growth is the result of the superposition of several processes of different nature in the meristem tissues, including division, extension and differentiation of cells, and also the production and distribution of phytohormones. The results can be applied to the use and modification of methods of woody plants nursing – the grafting, the pruning, the rejuvenation and the vegetative propagation.","PeriodicalId":125083,"journal":{"name":"Agrarian Bulletin of the","volume":"5 7","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-01-23","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"139602986","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The purpose of the study is to assess the current regulatory framework for the regulation of organic agriculture for the analysis of existing shortcomings and the degree of its compliance with the real needs of domestic agribusiness. The main method of research was the analysis of the legislative framework at the federal and regional levels, as well as domestic and foreign experience in organizing budget support for organic agricultural producers on its basis. Results. The current regulatory legal acts regulating the production of organic food in Russia and the world are considered. The assessment of measures to support organic agriculture in the context of individual regions of the Russian Federation was carried out. The characteristics of the development features and necessary adjustments of the current regulatory framework of organic production are given. Taking into account foreign practices, proposals are formulated to reduce the costs of organic producers for certification by developing systems of guarantee participation and group certification. In order to more actively develop the organic food market in Russia and, in particular, in the North-West, measures are proposed to make adjustments to the legal regulation of organic farming and animal husbandry, the transition to mass certification of small businesses. This will strengthen the competitive position of domestic organic agricultural producers primarily in the domestic market and strengthen the overall position of the industry even in conditions of increased external restrictions for export supplies. Domestic industrial rabbit breeding after 30 years of stagnation is beginning to gain momentum. Rabbit breeding is a well-developed branch of animal husbandry all over the world. An essential condition for the success of animal husbandry is the correctness of the organization of feeding, since in the cost of production, the cost of feeding reaches 60–70 %.The basis of the organization of feeding of all types of farm animals, including rabbits, is to determine their nutritional and energy needs. The feeding of young rabbits attracts special attention of scientists, since its energy demand is 3.4 times higher than that of adult animals during reproduction. Scientific research of domestic and foreign scientists on feeding young rabbits is very fragmented. The scheme of three-breed rabbit crossing has been developed at the Afanas`ev Research Institute of Fur – Bearing Animal Breeding and Rabbit Breeding on the basis of which the first domestic precocious and highly productive cross “Rodnik” was created. The relevance of the study lies in the need to create optimal diets for highly productive rabbit crosses. The purpose of the study is to study and analyze the achievements of domestic and foreign rabbit scientists contributing to the development of optimal diets for highly productive rabbit crosses. The novelty of the research lies in the fact that the analysis of new knowledge obtained by different scientific schools makes it possible to choose the direction of creating optimal diets for young rabbits. The results of the study. It has been established that the physiological features of rabbit digestion indicate the direction to the creation of diets optimized in terms of the ratio of nutrients, allowing to reduce the cost of feeding by 34 %. Probably, the need for nutrients and energy of new highly productive and precocious rabbit crosses will differ from the needs of meat-skinned breeds, as a result of which there will be a need to create individual diets.","PeriodicalId":125083,"journal":{"name":"Agrarian Bulletin of the","volume":"63 17","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-01-23","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"139603198","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

To form an optimization model in terms of fertility indicators relative to the requirements for the cultivation of potatoes. Methods. The object of the study is the territory of the northern forest-steppe of the Tyumen region. The studies were carried out on the basis of an analytical method using the following tools: at the first stage, the technology for creating a digital soil map of the South of the Tyumen region was carried out; at the second stage, a modeling method was used to develop an optimization model of soil fertility indicators; data of long-term research of the Department of Soil Science and Agrochemistry of the State Agrarian University of the Northern Trans-Urals. Scientific novelty. Work has been carried out to create a digital soil map of the south of the Tyumen region. 52 genetic types of soils were identified, the maximum area in the region falls on bog high-moor peat soils – 32.1 %, the minimum meadow-chernozem solonetsous and podzolized chernozems – 0.1 %. Taking into account the parameters of soil fertility, using the modeling method, an optimization model has been developed in the context of soil grouping by fertility. Results. The most favorable in terms of fertility for the cultivation of potatoes are the following types of soils - gray forest podzolized, dark gray forest solodized, ordinary solonetzic chernozems, podzolized chernozems, leached chernozems and meadow chernozem soils, since their initial fertility indicators are closer to the required conditions for potato cultivation. The purpose of researches is influence legume crops and mineral fertilizers on crop rotation productivity and soil fertility elements in the conditions of Kaluga region. Methods. The research was carried out in a long-term stationary field experiment on gray forest medium loamy soil. The laying of field experience, observations, records and generalization of research results were carried out in accordance with generally accepted methodological recommendations. Results. It was found that the studied factors had an insignificant effect on the weed propensity, which, against the background of mineral fertilizers and with an increase in the share of legumes in the structure, was slightly higher. On average, during the fourth rotation, the highest crop productivity was obtained in a crop rotation with 40% saturation with legumes, both against the background of fertilization (38.6 c/ha of grain units) and without their use (32.6 c/ha of grain units). Applying of mineral fertilizers contributed to an increase in crop rotation productivity by 18–24 %. On average, for crop rotations, 6.4 c/ha of grain units or 22 % were obtained by applying mineral fertilizers in addition to a background without fertilizers.
