Properties of I(NaP) were studied on neurons isolated by mechanical treatment (control group) and by mechanical and enzymatic treatment using pronase E (from Streptomyces griseus) or protease type XXIII (from Aspergillus oryzae). We show that in neurons isolated with pronase E kinetic of activation and density of I(NaP) was unaltered. Enzymatic treatment with protease type XXIII did not alter I(NaP) activation but result in significant decrease in I(NaP) density. It has been shown that an inhibition of CSE by propargylglycine (PAG) results in restoration of a decreased ACh-induced relaxation of aorta in old rats. This effect of PAG was removed by blocking nitric oxide (NO) synthesis in the endothelial cells. Age-related changes in the levels of H2S, NO2- and enzyme activity of the constitutive synthesis of NO (cNOS) in the heart, were determined. It has been shown that PAG introduction elevates a decreased levels of HzS, NO2- and stimulates the suppressed activity of cNOS in old rats. These results suggest that PAG activates alternative ways of H2S synthesis and stimulates the constitutive synthesis of NO. It was shown that subcutaneous administration of amylin at the dose 1 mg/kg body weight doesn't affect the volume of secreted bile. Under these conditions, the concentration of taurocholic acid was increased and the concentration of tauroconjugates of chenodeoxycholic and deoxycholic acids was decreased in the bile. At the same time, the concentration of glycocholates remained constant and of unconjugated bile acids was decreased. This redistribution of bile acids spectrum leads to an increase in the coefficient of conjugation. Amylin changes the ratio of trygydroxy- and dygydroxycholates in secreted bile leading to an increase in the coefficient of hydroxylation. These results suggest that amylin enhances the processes of conjugation and hydroxylation of bile acids in hepatocytes that results in improvement of detergent properties of the bile, particularly, the ability of the bile to maintain the cholesterol in dissolved state. Minchenko","doi":"10.15407/FZ60.05.014","DOIUrl":"https://doi.org/10.15407/FZ60.05.014","url":null,"abstract":"We studied the levels of lymphocyte surface-associated sialylglycans and the expression of neuraminidase NEU1 and sialyltransferase ST6GAL1 genes in lymphocytes in erythremia patients and healthy donors as well as the levels of sialic acids in plasma and sialylation of alpha-acid glycoprotein and fibronectin. Moreover, we also investigated the type of sialic acids binding with its glycans using sialospecific lectins MAA and SNA. fibronectin protein in lymphocytes and its cell surface in erythremia disease as compared to healthy donors. It was shown that the levels of free sialic acids and neuraminidase activity in plasma are increased in erythremia disease as compared to healthy donors; however, MAA-II-binding activity of tested glycoproteins is decreased, fibronectin-1 mRNA expression in lymphocytes is increased in patients with erythremia. The decreasing of plasma fibronectin concentration and its heparin-binding activity as well as increasing of lymphocyte content with surface-associated and intracellular fibronectin were revealed in erythremia disease in comparison with healthy donors. Positive correlation between plasma fibronectin level and its heparin-binding activity and negative correlation between plasma fibronectin level and quantity of lymphocytes which express fibronectin inside the cell and on cell surface was detected. Enhanced levels of α2,3- and α2,6-linked residues of glycocojugates were detected on lymphocyte cell surface in erythremia disease using sialospecific lectins and flow cytometer as well as fluorescent confocal microscope. The level of NEU1 and ST6GAL1 mRNA expressions is significantly increased in lymphocytes in erythremia disease. At the same time, the activity of both inducible and constitutive isoforms ofNO-sythase decreased. The parameters of lipoperoxidation remained at the level observed during water-restrained stress. It was shown the advantages of the use of H2S-releasinfg nonsteroidal anti-inflammatory drug ATB-346, which are associated with its cytoprotective effect of the drug manifested by a decreased total area of gastric damage. However, parameters of lipoperoxidation and NO-syntase system did not differ substantially from those in the group treated with napoxen, indicating the prevalence of parent molecule (naproxen) in regulation of function of NO-system Administration of dual COX/LOX inhibitor, the compound 2A5DHT, caused a decrease of gastric damage as compared to the effect ofnaproxen. Ca2+-accu- mulation was carried out in the presence of Mg-ATP2- and succinate. Transport of Ca2+ recorded using Ca2+-sensitive fluorescent probe Fluo-4 AM. It is shown that acidification of extramitochondrial medium is accompanied by stimulation of Ca2+ release from mitochondria. This process is insensitive to the tetraphenylphosphonium which is relatively specific Na+-Ca2+-exchanger inhibitor of mitochondrial inner membrane, but inhibited in the presence of monoclonal antibodies directed against the protein LETM1 (Anti-LETM1). LETM1 protein in some tissues is the molecular basis of the H+-Ca2+-exchanger functioning on mitochondria. It was found that the H+-Ca2+-exchanger is stimulated by 100 μM amiloride (diuretic) and inhibited by Mg ions in milimolar concentrations. The transport system was completely resistant to the action of nitric oxide (sodium nitroprusside and sodium nitrite), but was stimulated by macrocyclic compounds of Calixarenes (C-97 and C-99) in submicromolar concentrations. Thus, the mitochondria of rat myometrium probably not have a system of Na+-Ca2+-exchanger, and provide the maintenance of the matrix Ca2+-homeostasis with H+-Ca2+-exchanger. Experimental trials were performed under conditions of chronic experiment on model chrome-induced ECD. Both deltaran and melatonin either alone or in combination were used for complex pharmacological correction of humoral and cell-mediated immunity and also for stability of leukocytes. The data obtained showed the expressed disturbances of humoral and cell-mediated immunity and neutrophils' functional stability damage under conditions of chrome-induced ECD in rats. The revealed alterations in functional activity of the immune system were successfully corrected using the combined administration of deltaran and melatonin. The cross-section area of acini reduced by 12%, the cross-section area of acinocytes nuclei increased by 10%, the length between the lobes of the gland has grown by 48%. Kosterin","doi":"10.15407/FZ60.02.012","DOIUrl":"https://doi.org/10.15407/FZ60.02.012","url":null,"abstract":"The influence of the donor and the precursor of NO, namely 100 mM sodium nitroprusside and sodium nitrite on the energo-dependent Ca(2+)-transport in isolated mitochondria from rat myometrium was investigated. Changes in the mitochondrial matrix Ca(2+)-concentration was evaluated by spectrofluorimetry using Ca2+ sensitive probe Fluo-4 AM. Mg(2+)-ATP-dependent Ca(2+)-accumulation on mitochondria in the presence of succinate significantly stimulated by nitric oxide, in particular, 100 microM sodium nitroprusside amplified the transport by 1.6 times relative to its control values. NO effect becomes significant only when the incubation of mitochondria with the compounds was performed. Ca(2+)-accumulation in the presence of sodium nitroprusside effectively suppressed by protonophore (CCCP) and ruthenium red (10 microM). It was concluded that inner mitochondrial membrane Ca(2+)-uniporter stimulated by nitrogen oxide. Ca(2+)-accumulation in mitochondria in the presence of sodium nitroprusside was not sensitive to the action of a specific permeability transition pore inhibitor cyclosporine (5 microM). This data indicates that the role of permeability transition pore is less significant than Ca(2+)-uniporter in the processes of Ca(2+)-transport in mitochondria under the nitric oxide action.
