According to the data obtained, severe hypoxia in the prenatal period induces remarkable disturbances of GR expression in the neurons of neocortex of adult males but not females, that correlates to the disruption of working memory in adult males exposed to hypoxia on the prenatal 14-16th days. Elevation of stress plasma corticosterone levels have been observed only in the females subjected to hypoxia on the prenatal 17-19th days. Administration of MI-I1 at 2.7 mg/kg for 20 weeks on DMH-induced carcinogenesis prevents anemia, which is a consequence of cancer and complicates it. This is confirmed by a reduction in the numberofreticulocytes (0,19(0,15;0,21)x 10(12)/1)and restoration of mean corpuscular hemoglobin (18,02 (17,44;19,03) pg) and mean corpuscular hemoglobin concentration (309,42 (292,38;318,27) g/L) to a control value (0,17 (0,15;0,19), 18,31 (17,95;18,45), 310,78 (306,25;316,18), respectively) in contrast to the group DMH (0,28 (0,24;0,39); 17,50 (17,00;17,96); 288,10 (284,71;303,73), respectively). MI-I normalizes the number of monocytes (1,40 (0,95;2,50)x 10(9)/L) and platelets (646,32 (575,23;700,50)xl10(9)/L) in the blood after 26 weeks ofexperiment; in the DMH group, the values are significantly higher (1,97 (1,52;2,58), 783,90 (687,64;922,27), respectively) as compared to control group (1,23 (0,94; 1,68), 629,34 (590,19;711,48), respectively). MI- 1 reduces the involvement of these cells in the tumors progression and metastasis. It was shown that blockade of ERN1 finction in U87 glioma cells is induced the expression of RYBP and SESN1 genes, but decreased the expression of TP53BP1, TP53TG1, TOPORS, NME6, genes. Moreover, the expression levels ofRYBPI SESN1, TP53BP1, and ZMAT3 genes is increased in control glioma cells by L-glutamine deprivation condition but blockade of ERN1 signaling enzyme function significantly enhances this effect on the expression all of these genes. At the same time, the ERN1 knockdown eliminates the response TP53TG1 and TOPORS genes to L-glutamine deprivation condition. Pyshnov, Ie V Moĭseienko","doi":"10.15407/FZ60.01.070","DOIUrl":"https://doi.org/10.15407/FZ60.01.070","url":null,"abstract":"The study revealed that the Antarctic territory in the area of the Ukrainian Antarctic station \"Academician Vernadsky\" can be referred to the zone of the \"super high\", risk in developing ophthalmological pathology, caused by ultraviolet radiation. The results of the comprehensive dynamic study of 24 winterers of the XVI and XVII Ukrainian Antarctic expeditions (2011-2013) showed the developing of specific changes in the organ of vision under the exposure to the excessive natural ultraviolet radiation (UVR), such as parenchymatous-endothelial keratopathy, nuclear-posteriorcapsular phacopathy, maculopathy. Morphological changes were recorded using the cornea densitometry method: a decrease of the central cornea thickness by 14.3%, a decrease of the density and the number of endothelial cells of hexagonal type by 10.1% and 16.6%, respectively (P < 0.05). Using densitometry method, we detected morphological changes in the lens. Specifically, we found an increase of the optic lens density: in median zone by 10.1%, in maximal zone by 21.6%, in posterior part by 13.1%, P < 0.05. The results of densitometry recordings point to the development of local opacity in the posterior lens parts (in the nucleus and in the capsule). The data of the computed quantitative macula perimetry showed a significant decrease of light sensitivity of retina conicalis cells by 6 times (P < 0.05). The morphological signs of the endured acute solar retinopathy were found in more than 50% winterers. Our results indicate that treatment of diabetic rats with agmatine causes a suppression of erythropoiesis and increases the resistance of erythrocytes against HCl-induced hemolysis. It was shown a 10% increase in the number of young red blood cells and 35% reduction in the number of structurally transformed erythrocytes that are capable of restoring normal biconcave disc shape under physiological condition. Dosenko","doi":"10.15407/FZ60.03.003","DOIUrl":"https://doi.org/10.15407/FZ60.03.003","url":null,"abstract":"To determine the role of proteasome proteolysis in the pathogenesis of hypertension, we have studied the proteolytic activity of the proteasome in the aorta and heart tissues of rats with spontaneous hypertension (line SHR), and used quercetin, the drug that can inhibit the activity of this multicatalytic complex. In the aorta of SHR, the activities of the proteasome were not significantly different from that observed in Wistar rats. At the same time, in the heart tissues the trypsin-like (at 40%, P > 0.05), and chymotrypsin-like (by 1.7 times, P < 0.03) activities were significantly less in SHR. Significant morphological changes (fibrosis of the left ventricle was 4.7%, aorta intima width was increased and heart weight index was higher by 21.6% (3.7 +/- 0.6 mg/g) compared with Wistar rats (2.9 +/- 0,4 mg/g, P < 0.004) were observed in these animals functional disorders (reduced stroke volume by 3 times (P < 0.0001), ejection fraction by 2.5 times (P < 0.0001), increased end diastolic pressure by 6.5 times (P < 0.005), end systolic pressure by 15% (P < 0.004)) were revealed. Pharmacological drug \"Qvercetin\" effectively inhibited trypsin-like and chymotrypsin-like proteasome activities in the aorta (2.7-fold (P < 0.005) and 2-fold (P < 0.003), correspondingly) and trypsin-like, and peptidyl-glutamyl peptide-hydrolyzing-like activities (2.4-fold, P > 0.05 and 9.3-fold, P < 0.02, correspondingly) activities in the heart, leading to a significant improvement of morphological and functional parameters of the heart.
