Bahi","doi":"10.21157/J.KED.HEWAN.V13I4.12890","DOIUrl":"https://doi.org/10.21157/J.KED.HEWAN.V13I4.12890","url":null,"abstract":"The objective of this study was to characterize the morphology of ovary from slaughtered Aceh cow and measuring the concentration of estradiol in the follicular fluid to examine the potential utilizing for in-vitro fertilization (IVF) development in Aceh cattle. In total, 30 pairs of ovaries were collected from 30 Aceh cattle slaughtered at Banda Aceh abattoir. Morphological characteristics of ovaries observed were the size of ovaries, weight of ovaries, number of follicles, the diameter of follicles, corpus luteal (CL), and volume of follicular fluid. The follicular fluid was aspirated using a disposable syringe and analyzed the estradiol concentrations using enzyme-linked immunosorbent assay (ELISA). The follicles were grouped into 3 categories based on the diameter (Ø) of follicles: small follicles (Ø 1 - 5 mm), medium follicles (Ø 5 - 8 mm), and large follicles (Ø 8 mm). The morphological characteristic of ovaries varied between right and left ovaries in which right ovaries showed bigger size and weight compared to the left ovaries (P0.05), whereas the number of follicles and follicular fluid did not significantly different (P0.05). The concentrations of estradiol in follicular fluid of large and medium follicles were significantly higher compared to small follicles (P0.05), whereas the concentration of estradiol between large and medium follicles, did not significant different (P0.05). In conclusion, the follicles from abattoir can be used as potential oocytes resources to support IVF development for Aceh cattle in the future. This study used a completely randomized factorial pattern design with the first factor was the type of egg yolk (purebred chicken, kampung chicken, duck, and quail) and the second factor was the concentration of egg yolk (5%, 10%, and 15%). Semen was collected from twelve kampung roosters using massage method. Immediately after collection, the semen was evaluated macroscopically and microscopically. Semen with more than 70% motility was used in this study. The semen was diluted, packed in a ministraw, equilibrated, and frozen using liquid nitrogen vapor and stored in a liquid nitrogen container for 24 hours. Observation of spermatozoa motility was carried out in fresh semen, diluted semen, after equilibration and after thawing with four replications. The results showed that the type of egg yolk treatment had no effect (P0.05) on the recovery rate and motility of spermatozoa before and after cryopreservation, but egg yolk concentration had a highly significant effect (P0.01) on the quality of spermatozoa. Egg yolks in 10-15% concentration had spermatozoa motility and recovery rate higher than egg yolk with 5% concentration. A total of 15 broiler chickens were used in this study and divided into 5 groups, K- (without treatment), K+ (given heat stress), P1 (given heat stress and 0.56mL BCO/400kg body weight, P2 (given heat stress and 1.11 mL BCO/400 g body weight), and P3 (given heat stress and 2.22 mL BCO/400 g body weight). Heat stress was given for 5 hours with temperature range of 34-35° C for 7 days. Cortisol was measured using the cortisol enzyme-linked immunosorbent assay (ELISA) kit. Liver histopathology was stained with hematoxylin eosin and observed with electron microscope. The data were analyzed using one way analysis of variance (ANOVA). This study found that application of heat stress to broiler chickens increased cortisol levels and induced histopathological changes in the liver. The BCO administration reduced cortisol level significantly (P0.05) in heat-stressed broilers. BCO administration also significantly reduced (P0.05) the degenerative changes in liver histopathology such as fat degeneration, hemorrhage and necrosis in broiler chickens under heat stress, but did not significantly influence the inflammatory cells infiltration. The in vitro method was carried out using well diffusion method. For in vivo evaluation, the effect of L. acidophilus on the survival rate of Pangasianodon hypophthalmus (P. hypophthalmus) infected with A. hydrophila was evaluated. The well diffusion method showed a significant inhibition ability of L. acidophilus cells against A. hydrophila compared to the cell free extract. The inhibition diameters obtained with cells and cell free extract were 17.23 mm and 15.17 mm, respectively. P. hypophthalmus injected with L. acidophilus cells and cell free extract following challenged with A. hydrophila cells showed survival rate of 70% and 60% respectively, at 2-week post challenged. The gas chromatography-mass spectrophotometry (GC-MS) result revealed that a diverse of compounds was detected in both the L. acidophilus cells and cell free extract, among them the most abundant component was pyrrolo[1,2-a]pyrazine-1,4-dione, hexahydro-3-(2-methylpropyl), which showed a promising anticancerous activity and might be played a significant role in the recovery of the infectious P. hypophthalmus.
