Pitt","doi":"10.1016/S0007-1536(88)80038-X","DOIUrl":"10.1016/S0007-1536(88)80038-X","url":null,"abstract":"<div><p>The effects of water activity (<em>a</em><sub>w</sub>) and temperature on germination and growth of <em>Eurotium rubrum, Aspergillus flavus, A. wentii</em> and <em>A. penicillioides</em> were studied on media in which <em>a</em><sub>w</sub> was controlled by glucose/fructose or NaCl. All species grew most rapidly on glucose/ fructose and were partly or highly inhibited by NaCl. <em>Aspergillus wentii</em> was surprisingly tolerant of NaCl at 25 °C, with high growth rates on both NaCl and glucose/fructose media near 0.95 <em>a</em><sub>w</sub>. Decreasing spore density and the amount of accompanying matrix increased germinability of freshly gathered spores, especially those from < 10-day-old cultures. Removing the matrix altogether by washing and then decreasing spore density significantly increased germinability. When subjected to drying, germinability of matrix-free (washed) spores was negligible, whilst that of unwashed spores remained the same as for freshly harvested spores, especially with increasing spore density and culture age. No interfertility was detected corresponding to a low DNA homology of 63%. DSM 4018 displayed the tetrapolar mating system. The GC content of the nuclear DNA was 43.2 ± 0.4 mol%. The activities of several enzymes of intermediary carbon metabolism were only slightly affected by removal of carbon dioxide. However, depending on the time and duration of a lowered carbon dioxide level, sexual differentiation was strongly inhibited, possibly through inhibition of α-1,3 glucan synthesis. If the cultures were deprived of carbon dioxide at the end of the growth phase, the activity of the developmentally regulated enzyme α-1,3 glucanase was very low and sexual differentiation was inhibited. Addition of carbon compounds to the medium in some cases alleviated the negative effect of lack of CO<sub>2</sub> on sexual differentiation.
