La capacité de ces anticorps à réagir avec les 4 sous-classes d'IgG a été évaluée dans les conditions expérimentales décrites pour les mesures. La capture des anticorps monoclonaux anti sousclasse est réalisée par un anti Fc d'IgG de souris. La technique est sensible : capable de détecter des concentrations de l'ordre du ng/ml et précise : le cœfficient dev ariation inter-essais est de 14 %. En utilisant le sérum de référence OMS 67/97, nous avons étalonné un pool de sérums réalisé à partir de 100 donneurs. De plus, la concentration de chaque sous-classe a été évaluée dans 69 sérums d'adultes sains afin d'étudier la distribution du taux des différentes sous-classes. Un cœfficient de corrélation de 0,78 a été obtenu entre le taux des IgG mesurées par immunonéphélémétrie et la somme des sous-classes d'IgG chez ces mêmes 69 sujets.</p></div><div><p>We developped a enzyme linked immunosorbent assay (ELISA) for measuring IgG subclasses concentration in serum. For this we used monoclonal antibodies. The specificity of these antibodies was evaluated with a panel of myeloma proteins belonging to the 4 IgG subclasses. The ELISA was sensitive (allowing the detection of subclasses at ng level) and accurate (inter-assay cœfficient of variation of 14 %). Using the WHO serum 67/97 as reference, we determined the concentration of IgG subclasses in a pool of sera. In addition concentrations were measured in 69 healthy adults to study the distribution of each IgG subclass. L'application des règles simples de gestion de la qualité permet d'atteindre plus facilement ce but. Après la recherche de la qualité, après sa mesure efficace et régulière et après la définition d'objectif qualité, la formation et l'information du personnel favorisent la confiance indispensable à une bonne gestion de qualité. Enfin, la standardisation de l'automatisation des procédés de fabrication constituent les seules voies pour l'amélioration de la qualité réelle, pour la diminution des déclassements et rejets et tout naturellement pour l'augmentation de la productivité.</p></div><div><p>The quality controls of blood components, realised in Transfusion Centres, are necessary to check the true conformy of products in accordance with official standards. This controls are unsufficient to enhance blood components quality. The pratical application of the quality management allows us to reach this aim easier. After the quality display, after it efficiency and regulary measurement and after a quality objectives selection, we consider that the professionnal training and the data presentation contribute to the necessary confidence for a better quality management. L'analyse des profils électrophorétiques des sérums contenant ces variants, réalisés à 3 pH différents (8,6, 5,0 et 6,9) après addition d'une protéine témoin, la transferrine, permet d'identifier chacun des variants par l'estimation quantitative de ses mobilités relatives. La précision et la reproductibilité de la technique en a fait une méthode de référence couramment utilisée pour l'étude des variants européens.</p><p>Les échantillons servant de témoins de référence de cinq types de variants génétiques, les proalbumines Christchurch et Lille, les albumines Vanves, B et Reading ont été soumis à des analyses de séquences afin de déterminer la nature et la localisation de leur modification structurale.</p><p>Ces données, complétées par la mutation déjà décrite des variants d'albumine Gent et Roma, permettent de disposer de sept témoins de référence, sur les huit existants, dont la mutation a été caractérisée et confirmée.</p></div><div><p>Eight different types of genetic variants of albumin are observed in the French population. The analysis of electrophoretic patterns of sera containing these variants, performed a three different pHs (8.6, 5.0 and 6.9) after addition of a reference protein (transferrin), allows the identification each variant by a quantitative estimation of its relative mobilities.
