{"title":"Establishment and Validation of Prognostic Model of Lung Adenocarcinoma Based on Apoptosis-Related Genes","authors":"Yu Liu","doi":"10.31579/2690-4861/307","DOIUrl":"https://doi.org/10.31579/2690-4861/307","url":null,"abstract":"Objective: Apoptosis is an autonomous cell death process controlled by genes that can keep the internal environment of cells relatively stable. Lately, many studies have shown that apoptosis plays an important role in the construction of tumor microenvironment, mainly through the release of a series of regulatory factors to control cell growth. Therefore, it is very valuable to find and explore the possible role of apoptosis in the pathogenesis of lung cancer and its impact on the prognosis of patients. Materials and Methods: We selected 10 apoptosis-related genes and used different statistical methods to construct a model to predict the survival and prognosis of patients with lung adenocarcinoma. Univariate and multivariate Cox analysis were used to evaluate the predictive power and value of prognostic models, as well as receiver operating characteristic (ROC) curve analysis. Results: We defined two subgroups (cluster1 and cluster2). Compared with cluster1, cluster2 was named \"apoptosis inhibition subgroup\". The survival prognosis of cluster1 was significantly better than that of cluster2, and the infiltration of immune cells in cluster2 suggested that its immune system was inhibited. Then, we established a new prognostic model of apoptosis-related genes to identify high-risk patients with lung adenocarcinoma. Two types of Cox proportional hazards analysis and ROC curve showed that our model can be used as a new independent factor for the prognosis of lung adenocarcinoma patients, and had strong predictive ability. Conclusion: Apoptosis is involved in the formation and development of lung adenocarcinoma and plays an important role. {"title":"Comparison of Serum Levels of SIL-2R, IL-6, IL-10, TNF-α, CRP, ESR and Fibrinogen in Patients with Active and Inactive Behçet’s Disease","authors":"Ali Osman AVCI","doi":"10.31579/2690-4861/312","DOIUrl":"https://doi.org/10.31579/2690-4861/312","url":null,"abstract":"Behçet's disease (BD) is a chronic inflammatory illness that affects the entire body and is characterized by recurring episodes of oral aphthae, ocular and cutaneous lesions, and scrotal or vaginal ulcerations. The involvement of other organs and systems increases mortality in addition to the significant morbidity. This study involved a total of 40 participants, 20 of whom were healthy controls and 20 of whom were patients (of the 20 Behçet's patients, 12 (60%) were in the active phase and 8 (40%) were in the inactive phase). There was no therapy being given to any of the 20 patients that would have affected their cytokine levels. Only young men made up the patient and control groups because both early onset and male sex are signs of poor prognosis. The ELISA method was used to measure the levels of serum cytokines. The statistical analysis of the derived numerical values employed the Mann-Whitney U Test. We found a significant correlation between serum cytokine levels and classical acute phase markers in active Behçet’s patients. ESR (P < 0,001), CRP (P < 0,001), fibrinogen (P < 0,001), IL-10 (P < 0,001), IL-6 (P < 0,001), SIL-2R (P < 0,001) and TNFα (P < 0,001). There was no statistically significant difference in serum levels of classical acute phase markers ESR (P = 0,746), CRP (P = 0,476) and fibrinogen (P = 0,940) when inactive Behçet’s patients and healthy controls were compared. However, serum levels of IL-10 (P < 0,001), IL-6 (P = 0,001), SIL-2R (P < 0,001) and TNFα (P = 0,001) were statistically different between inactive Behcet's patients and the control group. Our research shows that even in the inactive phase, serum cytokine levels of Behçet’s patients are much higher than the healthy control group. However, the levels of ESR, CRP and fibrinogen, which are classical acute phase markers, were found at normal levels in Behçet’s patients in the inactive phase. This research aimed to develop a risk level prediction model and clinical decision support system for CVD in Ethiopia using data mining techniques. A total of 4004 datasets were used to develop the model. Moreover, primary data was collected from the domain experts via interviews and questionnaires. The domain experts identified thirty-one risk factors, of which only eleven attributes were selected after experimentation to develop the model. Based on the result of experimentation, the model was developed by an unpruned J48 classifier algorithm which produced F-Measure 0.877, which is comparatively the best algorithm. The prototype system was developed by Visual C# studio tool. The developed prototype system helps health care providers to identify risk level CVD diseases. It was developed using a data mining technique, which can efficiently predict cardiovascular disease risk levels. There are, however, important limitations: increasing antibiotic resistance of Helicobacter pylori, medical contraindications, and an increasing unwillingness among patients to use antibiotics. Case presentations: We present three exemplary case reports of successful antibiotic-free Helicobacter pylori eradication therapies, one for each of these limiting categories: a 61-year-old female Caucasian patient refractory to prior eradication therapy, an 80-year-old female Caucasian patient with contraindication to antibiotics, and a 41-year-old male Caucasian patient refusing antibiotics. A review of the literature attempts to provide a scientific rationale for proton pump inhibitor-based and probiotics-supported Helicobacter pylori eradication. Conclusion: Antibiotics-free Helicobacter pylori eradication may offer a therapeutic option for patients in whom classical eradication therapy cannot be conducted or who are refractory to antibiotics.
