Here, we report the use of a biomineral magnetic nano-contrast agent, Fe-doped calcium phosphate nanoparticles (nCP:Fe), to label mesenchymal stem cells (MSCs) and track their migration towards brain tumor using magnetic resonance imaging (MRI). nCP:Fe labelled MSCs implanted intra-ventricularly in a rat C6-glioma model showed longitudinal migration towards tumor-region over 14 days, detected as hypointense signals in T2/T2* weighted-MR images, which was confirmed ex vivo using histological staining. Although, a small fraction of implanted MSCs migrated to the tumor-site, the study demonstrates that, MRI tracking of stem cells using nCP:Fe nano-contrast agent will facilitate better assessment of the efficacy of stem cell-guided anti-tumor therapy.","PeriodicalId":92706,"journal":{"name":"Precision nanomedicine","volume":"95 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-02-20","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"140445724","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

This gradual deterioration presents significant challenges in early diagnosis and treatment. As a solution, iron oxide nanoparticles (IONPs) have emerged as promising materials due to their diminutive size and inherent biocompatibility, which facilitates bypassing the blood-brain barrier (BBB), thereby serving as effective targeted nanocarriers for neuroprotective therapeutics, offering efficient delivery to the brain. Furthermore, IONPs demonstrate efficacy as T1/T2 contrast agents (CAs), offering a novel diagnostic tool for various NDDs, emphasizing GB. Despite the expanding potential of IONPs in the management of NDDs, a clear gap exists in developing environmentally sustainable synthesis methods. Such methods must adhere to ecological standards and enable the production of nanoparticles capable of efficiently crossing the BBB. This is essential for precisely delivering and monitoring therapeutic interventions in diseases such as AD, PD, and GB. This review systematically explores the potential of green-synthesized IONPs in the context of NDD treatment. In this review, we emphasize different functionalization techniques for IONPs, tackling how these functionalized IONPs can be effective therapeutic approaches for NDDs. Moreover, we discuss biosafety and biocompatibility by covering a range of in vitro biocompatibility studies to analyze the safety of green-synthesized IONPs for the human body. Furthermore, our thorough analysis discussed the current state of green-synthesized IONPs using plant extracts polyphenols, which also act as neuroprotective agents, as proved by studies mentioned in the review.","PeriodicalId":92706,"journal":{"name":"Precision nanomedicine","volume":"25 18","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-01-20","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"139610713","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Monoclonal antibodies have emerged as a potential disease-modifying therapy targeting amyloid-β (Aβ), a hallmark of AD pathology. This study offers a comprehensive overview of the efficacy and safety of lecanemab, a monoclonal antibody, focusing on its binding properties and clinical trial results. Lecanemab’s unique binding profile, with a strong affinity for toxic Aβ protofibrils, sets it apart from other monoclonal antibodies. Clinical trials have shown promising results, including reductions in amyloid burden, improvements in cognitive measures, and a reduction in the rate of cognitive decline. However, limitations such as small sample sizes, short-term follow-up, and the exclusion of certain patient groups must be considered. While lecanemab shows potential in slowing the progression of AD, a more extensive study is required to provide robust evidence of its efficacy, especially in diverse patient populations. Adverse effects and long-term safety also warrant further investigation. The results of this study have the potential to inform clinical guidelines and benefit healthcare professionals, caregivers, and researchers in their efforts to improve the treatment of AD. This review is a crucial step in addressing the growing challenge of AD and enhancing the well-being of affected individuals and their families.","PeriodicalId":92706,"journal":{"name":"Precision nanomedicine","volume":"16 11","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-01-08","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"139446754","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Studies on Boschniakine are sparse, also, taking into consideration that poor absorption, a rapid metabolic rate, and rapid systemic elimination could limit bioalkaloids’ pharmacological efficacy. To recognize such limitations, we propose the use of highly bioavailable Boschniakine-Solid Lipid Nanoparticles (Bos-SLNPs). This study examined the potential molecular mechanisms underlying the anti-diabetic effects of Bos-SLNP. Bos-SLNP (25 and 50 mg/kg) was evaluated for 30 days for its antidiabetic activity in streptozotocin-induced Type 1 and Type 2 diabetic rats. In addition, the RT-PCR technique was used to determine messenger-RNA expression of Interleukin-1, Interleukin-6, and Tumor Necrosis Factor-alpha in renal tissue. haematoxylin and eosin staining and Western blotting were used to find differences in the nuclear factor erythroid 2-related factor 2/heme oxygenase/Nuclear Factor-κB signaling pathway and how its proteins were expressed. Bos-SLNPs treatment resulted in an increase in the expression of nuclear factors erythroid 2-related factor 2 and heme oxygenase-1 and a decrease in the expression of Nuclear factor-B. This was because Bos-SLNPs have a protective mechanism, which comes from their anti-inflammatory and anti-oxidant properties. Bos-SLNPs exhibited a protective effect against diabetic nephropathy in rats induced with streptozocin, possibly via the nuclear factor erythroid 2-related factor pathway 2 / heme oxygenase 1/ nuclear factor B.","PeriodicalId":92706,"journal":{"name":"Precision nanomedicine","volume":"21 37","pages":""},"PeriodicalIF":0.0,"publicationDate":"2023-12-28","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"139148496","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Green synthesis attempts to utilize products from natural agents like plants and fungi because of their profound availability of bio compounds. The NPs derived from them may exhibit anticancer, antibacterial, antimicrobial, antioxidant, anti-diabetic, and immunomodulatory properties because of their unique physical and chemical properties. These extraordinary properties make them promising agents in medicine and agriculture areas. The review summarizes mushroom-derived NPs and bioactive compounds, including the nutritional content of mushrooms with their multipurpose properties, followed by encapsulation and delivery of the biotherapeutics. These NPs have found significant applications in advancing industrial and biomedical ventures. A complete understanding of the synthesis mechanism will help optimize the synthesis protocols and control the shape and size of NPs.","PeriodicalId":92706,"journal":{"name":"Precision nanomedicine","volume":"28 3","pages":""},"PeriodicalIF":0.0,"publicationDate":"2023-12-11","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"138978812","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

However, the patient remained with two active lesions. An experi-mental topical treatment with liposomal Miltefosine was tested. The efficacy was determined from the MD’s inspection of the patient and analysis of the lesions’ photographic images, le-sions samples culture, and anti-Leishmania antibodies quantification. The parasites from the patient were isolated. The parasites were typified as Leishmania (Viannia) braziliensis, and their susceptibility to Miltefosine, Meglumine Antimoniate, and Amphotericin B was measured. The wound healing was initially good with the topical treat-meant, but the clinical cure was achieved only with a combination of the topical liposomal Miltefosine and intravenous Amphotericin B. Of note, the treatment efficacy was strongly corre-lated with the degree of stress suffered by the patient. To our knowledge, this is the first report on using a topical Miltefosine formulation on a CL patient.","PeriodicalId":92706,"journal":{"name":"Precision nanomedicine","volume":"8 7","pages":""},"PeriodicalIF":0.0,"publicationDate":"2023-12-08","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"138589702","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

While several anti-TB drugs are currently commercially available, the main problem with these drugs is the requirement of prolonged treatment duration and frequent dosing. They are non-specifically distributed in the whole body through the blood circulation system, resulting in lower therapeutic efficacy and also causing other severe complications with-in the body. This review article focuses on the currently available various nanotechnolo-gy-based therapeutic approaches, including lipid nanoparticles, polymeric particles, carbon nanotubes, glucan, and alginate-based anti-TB drug-delivery systems, that allow the delivery and targeting of anti-TB drugs to host macrophage. Finally, we also present some promising recent nanotechnology-based mycobacterial detection systems using silver nanoparticles, silica nano-particles, magnetic nanoparticles, quantum dots, and biosensors that have recently emerged as the latest diagnostic tools against TB. Functionalized nanomaterials for emerging aggregation-induced photodynamic therapy as next-generation theranostics have also been discussed as a novel option for TB diagnosis and therapy.","PeriodicalId":92706,"journal":{"name":"Precision nanomedicine","volume":"7 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2023-11-29","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"139214398","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The study utilized a co-culture spheroid model comprising MIA PaCa-2 cancer cells and patient-derived cancer-associated fibroblasts (CAF-98) to mimic pancreatic cancer conditions. The spheroids underwent treatment with GNPs (7.5 μg/mL), LNPDTX-P (99 nM of DTX pro-drug), and 2 Gy of RT. Cell viability of the spheroids was evaluated using the CellTiter-Glo 3D assay. At the same time, DNA double-strand breaks (DSBs) were assessed by examining the expression of the DNA damage marker 53BP1 through an immunofluorescence assay. Alt-hough GNPs/RT and RT/LNPDTX-P showed a reduction in spheroid size and an apparent in-crease in DNA DSB damage, the combination of the two nanoparticles, GNPs, and LNPDTX-P, with RT, significantly enhanced the anticancer efficacy, resulting in a 28% decrease in spheroid size and an estimated 39% increase in DNA DSB. The combination of GNPs and LNPDTX-P with RT showed a synergetic effect due to their radiosensitizing properties, improving the ther-apeutic efficacy of each treatment modality alone. This triple modality offers a hopeful strategy to enhance cancer treatment efficacy while reducing adverse effects.","PeriodicalId":92706,"journal":{"name":"Precision nanomedicine","volume":"45 5","pages":""},"PeriodicalIF":0.0,"publicationDate":"2023-11-22","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"139250051","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
