Ketonic monosaccharide and disaccharide have begun to replace larger sugars in insulin resistance, obesity, LDL cholesterol, triglycerides, metabolic syndrome, type 2 diabetes and cardiovascular disease because they can impart different properties such as biochemical, glycobiological and biological properties, at the same time, provide property enhancements at lower loadings sugars form a variety of ordered structures on nanometer length scales, allowing them to severe as a ordered matrix for Fructose, Glucose and Galactose which add functionality to the sugars [11–21]. Discontinuous molecular dynamics simulation is used to study the phase behavior of Fructose, Glucose and Galactose [22– 25]. Considerable effort has been devoted to theoretical and computational simulation studies of sugars phase behavior [26–31]. This editorial describes the models used for phase diagrams of self– assembled mono–tethered nanospheres (TNS) from molecular simulation and comparison to larger sugars. An impressive variety of Fructose, Glucose and Galactose of different sugars and geometries has been synthesized. Also, in this editorial, we aim to predict the assembled structures formed from Fructose, Glucose and Galactose as well as to compare with larger sugars. Both lectins LA and LB showed by two dimensional gel electrophoresis very similar apparent molecular mass, and their isoelectric point for both lectins were also very close. Mass spectrometry analysis confirmed these similarities showing high sequence homology among them, and when these sequences were compared to the Mirkov´s deduced sequence of the phyto-hemagglutinin from Phaseolus acutifolius, LA showed higher homology than LB. On the other hand, when their biological activity was determined, only LA possessed affinity to type A erythrocytes, and when these lectins using a semi pure fraction were measured for their cytotoxic effect on colon cancer cells, they showed a differential effect. In the semi pure protein fraction tested, the rest of no-lectin proteins separated by electrophoresis presented no activity. Wei","doi":"10.4172/2168-958X.1000116","DOIUrl":"https://doi.org/10.4172/2168-958X.1000116","url":null,"abstract":"Objective: To investigate the relationship between different glucose metabolism status and Pulse Wave Velocity (PWV)and further explore the relationship between PWV, HOMA-IR and the Framingham risk score of 10-year (FRS). \u0000Methods: 105 subjects were divided into three groups: normal glucose tolerance group (G1, n=47); pre-diabetes group (G2, n=32)and type 2 diabetes mellitus (DM) group (G3, n=26)General clinical data and biochemical parameters of each subject were collected. PWV was measured using the Complior SP. FRS was calculated with the lipids excel spreadsheets from Framingham Heart Study homepage. The correlation of different glucose metabolism status, PWV and FRS were analyzed and compared. \u0000Results: A positive correlation was found between carotid-to-femoral PWV (cf-PWV) and age, (Systolic Blood Pressure) SBP, 2-hr plasma glucose (2h PG). There is a direct connection between homeostasis model assessment of insulin resistance (HOMA-IR) and HOMA-IR (CP). FRS was difference among three groups. A correlation was found between FRS and cf-PWV, 2h PG, HOMA-IR (CP). \u0000Conclusion: HOMA-IR (CP) and cf-PWV are positive correlation with FRS, which is a predictor of cardiovascular disease (CVD). Their relative uptake in normal hearts varies with the metabolic milieu (fed/fasting state), plasma levels and insulin levels. Myocardial ischemia results in a profound up-regulation of glucose and down regulation of FFA metabolism. Fluorine-18 labeled deoxyglucose (FDG); a glucose analogue is used extensively in clinical practice for imaging glucose metabolism in heart, tumors and other organs. Alternations in glucose metabolism accompanying several disease conditions can be used to diagnose these conditions. Exercise results in ischemia and up-regulates glucose uptake in regions perfused by diseased coronary arteries. This can be used for developing a non-invasive diagnostic imaging test for coronary artery disease (CAD). We evaluated the diagnostic potential of exercise FDG imaging for CAD and compared it with exercise-rest myocardial perfusion imaging (MPI), an established and routinely used test. FDG imaging had higher diagnostic sensitivity compared to MPI. Furthermore, increased regional FDG uptake is observed only on exercise images. Persistence of FDG uptake 24 hours later is seen in less than one third of cases and is indicative of more severe CAD. To reduce both the time and the temperature of the analysis, wood powder samples are first totally disrupted in 1-ethyl- 3-methylimidazolium under controlled microwave irradiation. The solubilized extracts are then directly spotted onto nitrocellulose membranes in order to specifically detect their polysaccharide contents with a set of monoclonal antibodies. Compared to the long sequential method classically used for biomass analysis, the technique described in this article allows the simultaneous treatments of higher numbers of smaller samples. {"title":"Role of HSGAGs in Regulation of FGF Signaling Pathway: Insights from Mathematical Modeling","authors":"J. Kanodia, G. These cues act as regulators of how cells undergo biological processes like development and normal tissue homeostasis among others. Errors in environmental cues or the ability of cells to respond to such cues leads to erroneous cellular behavior and potentially disease states like cancer. Accordingly, insghts into cell-signaling and processing can be instrumental in design of effective therapies against malignancies. A central paradigm of signaling pathways is that ligands bind to and activate cell-membrane bound receptors, which in turn leads to activation of intracellular cascades. However, to target receptor mediated signaling effectively, it may not be enough to understand the biology of the target receptor in isolation. Interactions with other receptors of the same receptor tyrosine kinase (RTK) family or other families might be critical for understanding the signaling diversity of a particular pathway from the systems perspective. For instance, the role of kinase-dead ErbB3 receptor in the regulation of EGFR and ErbB2 has been critical for a deeper understanding of the ErbB-pathway Along similar lines, heparan sulfate glycosaminoglycans (HSGAGs) have been shown to serve as co-receptors essential for signaling through cell-surface interactions on multiple receptors such as MET and FGFRs. We have recently investigated signaling responses in the FGFR system and found a thus far unappreciated signaling mechanism. We have leveraged a data-driven mechanistic modeling approach to examine the role of FGF2-dependent FGFR signaling driven by FGF2-HSGAG-FGFR1 trimeric complexes.
