A polymorphic change from methionine (Met) to valine (Val) at amino acid position 129 of the alpha 2 heavy-chain categorize MICA alleles into strong and weak binders for the NKG2D receptor. We investigated here whether MICA-129 alleles are associated with gastro-intestinal tract (GItract) carcinomas in Tunisian affected patients as compared to healthy controls (HC). \u0000Material and methods: 181 patients affected by colorectal cancer (CRC) and 61 patients affected by gastric cancer (GC) along with 203 healthy controls (HC) were genotyped by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) procedure. \u0000Results: We found that the MICA-129 Val/Val genotype was statistically more prevalent in patients affected both by CRC and GC as compared to HC. After stratification with poor prognostic parameters, we observed that MICA- 129 Val/Val genotype is significantly associated with advanced tumor extension (T3-T4), lymph node metastasis (N+), and distance metastasis (M+). In both cases, the MICA-129 Val/Val genotype seems to behave as a risk genotype and a poor prognostic biomarker in our population. \u0000Conclusion: Our findings suggest a potential tumor escape possibly due to an inability to activate NK cells and/ or to stimulate specific T lymphocytes subsets particularly active in the GI-tract.","PeriodicalId":89808,"journal":{"name":"Journal of molecular biomarkers & diagnosis","volume":"8 1","pages":"1-8"},"PeriodicalIF":0.0,"publicationDate":"2017-01-27","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"41443637","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The present study investigated the possible biochemical and molecular mechanisms underlying the hepatoprotective and testoprotective effects of SBEAB in diabetic rat. Biomarkers of hepatic and testicular damage, histological and immunohistochemical techniques were used. The expression of cyclooxygenase (COX-2) and inducible nitric oxide synthase (iNOS) were also estimated. SBEAB administered orally at dose of 100 mg/kg for 14 days significantly lowered the activities of serum transaminases and MDA levels induced by single intraperitoneal administration of streptozotoxin (STREP) (80 mg/kg) and preserved the integrity of both hepatocytes and spermatocytes. Also, SBEAB elevated the STREPinduced reduced activities of Δ5-17β-HSD and Δ5-17β-HSD with corresponding decrease in the activity of CAT. SBEAB inhibited the STREP induced expression of COX-2 and iNOS. The protective effect of SBEAB was compared to that of metaglomide (METAG), an established anti-diabetic drug. METAG treatment on hepatic damage was most efficacious in diabetic rats; followed by post and pre-treatment respectively while pre-and post-treatment were more efficacious on testicular damage than anti-diabetic drug. Furthermore, pre and post-treatment were more efficacious in preventing pro-inflammation and testicular cancer in diabetic rats than METAG-administration. We therefore concluded that the repression of genes encoding COX-2 and iNOS proteins by SBEAB validates the molecular basis of testicular protection and further suggests the links between the hepatocellular damage and male reproductive dysfunctions in diabetic individuals.","PeriodicalId":89808,"journal":{"name":"Journal of molecular biomarkers & diagnosis","volume":"8 1","pages":"1-8"},"PeriodicalIF":0.0,"publicationDate":"2017-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"70335025","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Since the current subjective clinical diagnostic systems were not designed to assess depression in patients with chronic pain, they fail to adequately capture the nature of mood states of these patients. It is difficult to apply these assessment tools to segregate unipolar depression (MDD) from the demoralization inherent in chronic pain states. MDDScore™, a multivariate biomarker blood test for depression, was used to determine if it was possible to identify biomarker patterns consistent with major depressive disorder in multiple chronic pain states. Three study groups were analyzed, and included: (i) patients (n=93) with centralized Chronic Intractable Pain (CIP), (ii) patients (n=20) with chronic pain of diverse origin from Scripps Pain Clinic (SPC) and (iii) prospectively collected patients (n=28) with comorbid arthritis and depressive symptoms. A very distinct bimodal pattern was observed. Forty-nine of 93 CIP patients (52.7%), 18 of 28 (64.2%) of the arthritis patients, and 9 of 20 (45%) patients with chronic pain of diverse origin from Scripps Pain Clinic had MDDScores of ≥5. Thus, the biomarker panel could segregate patients into two major groups based upon MDDScores. These data suggest but not prove we can objectively identify chronic pain patients with a higher probability of comorbid major depression. Importantly, we can use the biomarkers on the MDDScore panel to gain insight into and gauge the residual (post-treatment) level of inflammation in these intensively treated patients. To this end, we determined and compared the serum concentrations of alpha-1 antitrypsin (A1AT), Myeloperoxidase (MPO) and soluble tumor necrosis factor receptor type 2 (sTNFR2) in each of the patient populations studied.","PeriodicalId":89808,"journal":{"name":"Journal of molecular biomarkers & diagnosis","volume":"8 1","pages":"1-5"},"PeriodicalIF":0.0,"publicationDate":"2017-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"70335417","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Fear increases likelihood fixation in memory behaviorally important information associated with a traumatic event. For healthy psychological functioning is crucial the ability to update the content and emotional charge of memories (cognitive plasticity) contributing to fading and extinction of fear memories once this event has been passed. Memory performance and its plasticity are associated with ability of synapses to change of number α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid glutamate receptors (AMPAR) on the post-synaptic membrane surface. AMPAR undergo exocytosis/endocytosis (AMPAR trafficking), thus changing the strength of neural connections (synaptic plasticity) and contributing to formation, maintaining and transformation of memories. Impairment of AMPAR trafficking may become a cause of deficiency of fear extinction, and development of posttraumatic stress disorder (PTSD). AMPAR trafficking is controlled by the nervous and the immune systems jointly. Kidney and brain expressed protein (KIBRA) and interleukin (IL) -1β are important regulators of AMPAR trafficking. Data show that genes encoding of these proteins (KIBRA-WWC1 gene and IL-1β gene) and their alleles mediates of stress impact on synaptic plasticity and are a bridge between memory and diseases. Disease susceptibility in humans is most commonly associated with single nucleotide polymorphisms (SNPs), when corresponding sequences of DNA from different individuals differ at one DNA base. PTSD is polygenic disorder, so can be identified the combination of allelic variations of SNPs, associated with strength and plasticity of memory that distinguish PTSD patients from the control. Our preliminary results showed that the combination of the certain allelic variants of WWC1, SNP rs17070145 (T/C), and the IL-1β gene, SNP rs16944 (A/G), is characteristic for PTSD compared to depressive and healthy groups. We assume that this combination might be used for pre-clinical diagnosis PTSD and in clinical practice.","PeriodicalId":89808,"journal":{"name":"Journal of molecular biomarkers & diagnosis","volume":"8 1","pages":"1-6"},"PeriodicalIF":0.0,"publicationDate":"2017-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"70335697","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Enrolled women were randomized into 3 intervention arms, screened by VIA/ VILI, pap smears, and careHPV assay respectively. Women had positive primary screening results were referred to colposcopy and/or biopsy. All the clinical and lab work was performed by local health providers. The final diagnoses of histopathology were based on the diagnosing of a senior histopathology expert from Cancer Hospital, Chinese Academy of Medical Sciences. Questionnaires about the knowledge and attitudes towards the HPV and the screening program of the health providers at village clinics were collected. Results: 894 women had careHPV DNA test, 552 underwent VIA/VILI and 547 had Pap smears. The positive rates for careHPV assay, VIA/VILI and Pap smears were 10.6%, 18.1%, and 4.9% respectively (χ2=48.647, P<0.001). The overall CIN2+ detection rate was 0.5%, the CIN2+ detection rate for the three arms were not significantly different (0.7% for careHPV assay, 0.5% for VIA/VILI, 0.2% for pap smear, χ2=1.648, P=0.439). The knowledge of the health providers about HPV, HPV assay in screening was unsatisfactory. However, their attitudes about implementing HPV assay into the national program were positive. The Human Microbiome (all of our microbes' genes) can be considered a counterpart to the human genome (all of our genes). The genes in our microbiome outnumber the genes in our genome by about 100 to 1 [1]. The human microbiota consists of the 10-100 trillion symbiotic microbial cells harbored by each person, primarily bacteria in the gut; and the human microbiome consists of the genes these cells harbor [2]. The microbiome may be considered a “new organ system,” because its existence and contributions to human health and disease was uncovered by researchers between 15 and 20 years ago [3]. The composition and function of the microbiome of the human gut evolves during the first years of life and stabilize within the first 3 years of life [4]. Specifying the definition of the human microbiome has been complicated by confusion about terminology: for example, “microbiota” (the microbial taxa associated with humans) and “microbiome” (the catalog of these microbes and their genes) are often used interchangeably [1]. Microbial colonization of the infant occurs during a critical time window for immune and gastrointestinal development. Infant colonization sets the stage for the adult microbiome [5]. Major outcomes of colonization include the timing-dependent education of the neonatal immune system, which is interconnected with barrier function and metabolism. The infant is born not completely sterile, depending on mother state of health prenatally. As soon as the infant faces real environment “In a matter of thirty minutes” the colonization of the gut and the skin starts. The type and diversity of gut bacteria during the first hundred days of an infant's life has a profound influence on the developing immune system and on the risk of atopy, infection and inflammation. It dramatically influences this period of immune maturation. Manipulating an infant's \"dysfunctional\" microbiome might soon be a therapeutic approach for preventing atopy in a predisposed infant.!!! Factors such as breastfeeding, mode of delivery, antibiotic exposure, and pet exposure, have been known to be factors affecting allergy and asthma, by influencing the infant microbiome. The microbiome affects the development of regulatory T-cells, immunoglobulin E responsiveness, and the development of atopy symptoms. The association between microbial diversity, in the infant gut and regulatory T-cell development, is crucial in prevention of allergy and other non-communicable diseases. With breast feeding, microbes in mother’s breast milk seed the infant gut, [6] including those associated with beneficial effects, underscoring the importance of breastfeeding in maturation of the infant gut microbiome where the breast milk as bifido genic effect start colonization on the infant mucosa.
