Lactobacillus species are lactic acid bacteria that have great potential as probiotics, in which cow’s milk is a great source of lactic acid bacteria. In this study, Lactobacillus spp. were isolated from fresh local Indonesian cow’s milk. The methods used to characterize the Lactobacillus spp. were (i) biochemical tests including catalase, hemolytic and sugar fermentation tests; (ii) tolerance tests against salt (NaCl 2%, 4% and 6%), low pH (2, 3, 4, 5, 6 and 7) and temperature (7°C, 37°C and 45°C); and (iii) 16S rRNA sequencing. The isolation yielded 14 isolates matching the criteria of Lactobacillus spp. colony and cell (i.e., Gram positive rods that did not produce endospores and did not have a waxy layer covering its cell wall). Based on subsequent biochemical tests, 5 isolates were suspected as potential probiotic Lactobacillus spp. The 16S rRNA sequencing revealed that the isolate L was Limosilactobacillus fermentum. Namun, banyak tanaman jeruk di indonesia yang terkena penyakit Citrus Vein Phloem Degeneration (CVPD). Penelitian ini bertujuan untuk mengidentifikasi secara lebih pasti keberadaan bakteri Liberobacter asiaticus penyebab penyakit CVPD pada tanaman jeruk yang menunjukkan gejala klorosis dan juga keberadaan bakteri L. asiaticus penyebab penyakit CVPD pada tanaman jeruk yang tidak menunjukkan gejala klorosis. Untuk mencapai hal tersebut, teknik PCR akan digunakan untuk analisis molekuler bakteri L. asiaticus. Dengan mengamati sampel daun dari tanaman jeruk siam Kintamani yang menunjukkan gejala klorosis dan yang tidak menunjukkan gejala klorosis, maka metodologi penelitian eksploratif diterapkan dalam penelitian ini. Pita DNA pada 1.160 bp tidak ditemukan pada sampel daun jeruk yang diuji, baik pada sampel daun jeruk yang sehat maupun pada sampel daun jeruk yang terindikasi klorosis. Tanaman jeruk yang menunjukkan gejala klorosis tidak ditemukan bakteri L. asiaticus pada sampel daunnya. Hal ini mungkin disebabkan oleh kurangnya unsur hara di dalam tanah, konsentrasi bakteri yang rendah, atau distribusi bakteri yang tidak merata di dalam jaringan daun. {"title":"Pattern of Antibiotic Resistance in Salmonella sp. One of them is caused by improper use of antibiotics. This study aims to determine the pattern of resistance of Salmonella sp. isolated from laying hens to antibiotics. The samples used in this study were thirty samples of fresh faeces of laying hens from Kediri District, West Lombok Regency. The samples were isolated and identified through Gram staining and biochemical tests (indole, methyl red-Voges Proskauer, triple sugar iron agar, citrate, and urease), resulting in ten positive samples of Salmonella sp. The study continued with antibiotic resistance testing using disc diffusion or Kirby-Bauer methods. The antibiotics used in the study were aztreonam, ciprofloxacin, streptomycin, chloramphenicol, and tetracycline. The results showed that Salmonella sp. has been resistant to some antibiotics. The highest resistance levels are streptomycin and tetracycline at 50% (5/10), while the lowest is ciprofloxacin at 10% (1/10). Antibiotics that are still sensitive are aztreonam 90% (9/10) and chloramphenicol 100% (10/10). {"title":"Karakterisasi Molekuler Tumbuhan Sidingin (Kalanchoe sp.) Sidingin asal Riau belum dapat ditentukan nama spesiesnya. Penentuan nama spesies tumbuhan dapat dilakukan menggunakan beberapa barkode DNA, seperti matK, rbcL, dan ITS. Penelitian barkoding DNA pada sidingin asal Riau belum pernah dilakukan. Penelitian ini bertujuan menganalisis empat barkode DNA pada tumbuhan sidingin (Kalanchoe sp.) asal Riau, yaitu ITS, trnL-trnL-trnF intergenic spacer (IGS), ndhC-trnV IGS, dan rpl16 intron. Pada penelitian ini telah diperoleh sekuens DNA untuk ITS, trnL-trnL-trnF IGS, ndhC-trnV IGS, dan rpl16 intron dengan panjang basa secara berturut-turut 662 pb, 909 pb, 949 pb, dan 944 pb. Keempatnya telah didaftarkan dengan nomor registrasi, secara berturut-turut, MW297180, MW297177, MW297178, dan MW297179. Komposisi nukleotida pada ITS berbeda dengan ketiga barkode DNA lainnya. Tidak ada aksesi di GenBank yang memiliki kemiripan 100% dengan sidingin sehingga nama spesies dari sidingin belum dapat ditentukan. Variasi nukleotida paling sedikit dijumpai pada daerah trnL-trnL-trnF IGS (9,65%) dengan jumlah mutasi subtitusi dan indels relatif mirip. Namun uniknya, nukleotida kritis yang merupakan nukleotida penciri paling banyak dijumpai pada daerah trnL-trnL-trnF IGS (1,41%). Modification of starch with Amylosucrase can increase the degree of polymerization, degree of crystallinity, heat stability, and increase the proportion of slow digestible starch (SDS) and resistant starch (RS). Thus, Amylosucrase (ASase) modified starch has enormous potential to be developed in the food industry because the consumption of ASase modified starch can improve insulin sensitivity and reduce blood glucose response, making it suitable for consumption by people with Diabetes Mellitus. Keberhasilan regenerasi protokorm anggrek dipengaruhi oleh penambahan ZPT dan air kelapa ke dalam medium. Oleh karena itu, penelitian ini bertujuan untuk menentukan pengaruh konsentrasi air kelapa dan BAP terhadap regenerasi protokorm Dendrobium hibrida. Penelitian menggunakan Rancangan Acak Lengkap dengan dua faktor yaitu konsentrasi air kelapa dan BAP. Paramater yang diamati meliputi persentase tunas, jumlah tunas, tinggi protokorm, dan persentase akar. Hasil menunjukkan bahwa penambahan air kelapa dan BAP ke dalam medium VW berpengaruh nyata terhadap tinggi dan persentase akar protokorm. Konsentrasi air kelapa (10%) tanpa penambahan BAP dapat meningkatkan persentase tunas, akar, jumlah tunas, dan tinggi protokorm Dendrobium hibrida. Hasil penelitian menunjukan bahwa penggunaan air kelapa secara tunggal maupun kombinasi dengan hormon sitokinin (BAP) signifikan dalam mempercepat regenerasi protokorm Dendrobium hibrida. Tujuan penelitian ini adalah untuk mempelajari struktur komunitas satuan lanskap yang dimanfaatkan oleh Masyarakat Lereng Gunung Slamet. Penelitian dilakukan pada bulan Juli 2019 — Januari 2020 di desa Lereng Gunung Slamet yaitu, (1) Desa Ragatunjung, (2) Desa Cipetung, dan (3) Desa Pandansari Kecamatan Paguyangan, Kabupaten Brebes. Pengumpulan data botani dilakukan dilakukan secara purposive sampling menggunakan transek kuadrat. Analisis vegetasi diolah menggunakan Indeks Nilai Penting (INP), Indeks Keanekaragaman (H’), Indeks Kekayaan (DMg), Indeks Kemerataan (e’), dan Indeks Kesamaan komunitas. Berdasarkan Analisis vegetasi ditemukan 136 spesies yang dikategorikan ke dalam 11 genus dan 55 famili. Analisis struktur komunitas dapat dilihat pada H’ berkisar antara 1,57—28,9 yang termasuk dalam kategori sedang. DMg berkisar antara 11,82 – 28,8 dan Indeks kemerataan berkisar antara 0,11—0,92. Indeks kesamaan antar satuan lanskap, wanah dan majegan merupakan yang tertinggi yakni 62,67% yang termasuk kategori tinggi. Tujuan dilakukannya penelitian yaitu untuk mengetahui keanekaragaman ikan dan tumbuhan air tawar di Sumber Gentong, Kecamatan Pakis, Kabupaten Malang. Waktu dan tempat dilaksanakannya penelitian yaitu 2 November 2022 di Wisata Sumber Gentong. Metode penelitian yang digunakan yaitu penelitian deskriptif dengan cara survey, observasi dan wawancara yang dilakukan kepada penjaga di tempat wisata tersebut. Data yang diperoleh kemudian di susun dan dianalisis tingkat keanekaragamannya menggunakan indeks keanekaragam Shannon-Wiener. Hasil penelitian menunjukkan bahwa ditemukan 4 spesies ikan yaitu ikan wader, ikan cakul, ikan platy pedang, dan ikan mas. Tumbuhan air yang dapat ditemukan yaitu cabomba caroliniana, kangkung, eceng gondok, dan kayu apu. Indeks keanekaragaman ikan dan tumbuhan air tawar di Sumber Gentong, tergolong sedang dengan nilai (H’ = 1,14) untuk ikan air tawar dan (H’ = 1,00) untuk tumbuhan air tawar. Metode yang digunakan dalam penelitian ini adalah metode deskriptif dengan Teknik observasi lapangan, Hasil dari penelitian ditemukan tiga belas individu dan delapan jenis satwa liar mamalia yaitu Bandikut (Echymipera kalubu), Bandikut (Echymipera rufescens), Rusa timor (Rusa timorensis), Babi hutan (Sus scrofa), Kuskus cokelat (Phalanger orientalis), Kuskus totol (Spilocuscus maculatus), Kelelawar codot kecil (Microglossus minimus), Kelelawar kalong besar (Pteropus neohibernicus).
