Donelaičio g. 58, Kaunas 44248, Lithuania African swine fever (ASF) is a contagious disease of key importance that occurs in wild boar population and domestic pigs. To date, there is no vaccine or treatment available, hence disease surveillance and stamping out in case of an outbreak is the only way to control it. A duplex ASF-IC real-time PCR assay was optimised using field samples obtained in Lithuania and results of singleplex and duplex real-time PCR assays allowing the identification of the ASF virus were compared. The specificity and sensitivity of ASF virus detection was validated using a reference sample of positive and negative virus nucleic acid and ASF virus positive and negative samples obtained from animals in Lithuania. Results prove that duplex ASF-IC real-time PCR assay provides a rapid, sensitive, and reliable molecular tool for ASF virus detection in wild boar population and domestic pigs. In general, it was found that duplex ASF-IC real-time PCR assay proves to be faster and more sensitive than singleplex real-time PCR and therefore has a high potential to be applied in routine analysis.","PeriodicalId":8858,"journal":{"name":"Biologija","volume":"6 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2020-03-29","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"75203232","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Donelaičio g. 58, 44248 Kaunas, Lithuania Climate change significantly affects biological diversity around the world. Trends of this phenomenon have also been noticed in Lithuania: in the past decades, 55 new bird species have been recorded. The recent record of a new species was done on 3 December 2019. During ordinary birds ringing carried out in Ventės Ragas Ornithological Station in Lithuania, an individual Siberian northern shrike (Lanius borealis sibiricus) was caught and ringed. Additional blood sample was collected for species confirmation. Based on identification keys and molecular mitochondrial cytochrome b analysis, we report that this is the first record of the Siberian northern shrike in Lithuania.","PeriodicalId":8858,"journal":{"name":"Biologija","volume":"13 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2020-03-29","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"91210030","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The decrease in the linear dimensions of the aubergine was established for all preparations. The most significant decrease in height was observed after the application of 2-HEPCs. The retardants increased the number of leaves, the dry matter mass of leaves, the leaf area, chlorophyll content, and the surface density of the leaf. EW-250 and ССС-750 retardants strengthened carbohydrate accumulation in aubergine fruits, mainly due to reducing forms. During the growing season, the total and protein nitrogen content decreased in roots and stems both in control and in experiment variants. EW-250 and CCC-750 accelerated total and protein nitrogen outflow from roots and stems and increased its content in leaves. Increasing the carbohydrates content in fruits the retardants simultaneously reduced the content of various nitrogen forms. Such changes in the dynamics of total nitrogen under the influence of tebuconazole led to enhanced fruit formation on each plant. Thus, retardants EW-250 and ССС-750 changed the nature of the donor-acceptor relations in them by increasing the outflow of plastic substances from vegetative organs to fruits and increasing the culture productivity.","PeriodicalId":8858,"journal":{"name":"Biologija","volume":"20 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2020-03-29","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"73821589","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The present work is devoted to the study of the amorphous state stability of plant vitrification solutions (PVS) by differential scanning calorimetry method. The objects of the study were PVS1 modified, PVS2, PVS3 modified, PVS4 and PVSN. The thermograms of PVSs, frozen in liquid nitrogen, were recorded at a warming rate of 0.5 deg/min. The glass transition temperatures of PVS1, PVS2 and PVS3 were –109.0°C, –115.3°C, and –93.9°C, respectively. Neither exonor endothermic peaks have been recorded, indicating no crystallization, at the cooling and the warming stages. PVS4 and PVSN thermograms, besides glass transition (–111.5°C and –110.0°C, correspondingly), showed crystallization from the amorphous state (devitrification) and ice melting peaks. Crystallization enthalpy of PVSN was 2.8 times higher compared to PVS4. This fact testifies that the crystalline phase was larger in PVSN compared to PVS4. It should be noted that crystallization in PVS4 and PVSN may be related to the low warming rate used during the experiment. At higher warming rates, this crystallization is likely to be avoided. Thus, PVS1, PVS2 and PVS3 have a high glass-forming ability and a stabile amorphous state. The amorphous state stability of PVS4 and PVSN is significantly lower compared to PVS1, PVS2, and PVS3.","PeriodicalId":8858,"journal":{"name":"Biologija","volume":"74 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2020-03-29","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"85902226","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The investigated salmonid species included: the rainbow trout Oncorhynchus mykiss, the brown trout Salmo trutta morpha fario, the brook trout Salvelinus fontinalis, and the European grayling Thymallus thymallus. The fish were sampled from aquatculture farms as well as natural water bodies and their diseases included invasive (parasitic) and infectious (viral and bacterial). The most frequently encountered invasions in the brown and brook trout were ciliates: Chilodonella pisci cola and Apiosoma conicum, Trichodina (in particular T. truttae and T. nigra), Ichtyophthirius multifiliis; monogenea Gy rodactylus birmani, and diplostoma Diplostomum spathaceum. Aeromonas and Flavobacterium bacteria were isolated from fry and young-of-the-year rainbow trout and brook trout. Y. ruckeri positive samples were isolated from the fish with the signs of yersiniosis. As for viral diseases, IPNV isolates (the first time in Ukraine) of the rainbow trout were first isolated in fish farms in the western regions of Ukraine. A phylogenetic analysis of these IPNV isolates was performed, which showed that they belonged to Sp strain and the European genotype.","PeriodicalId":8858,"journal":{"name":"Biologija","volume":"69 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2020-01-04","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"75746634","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

It was established that under the action of gibberellin, stimulation of the aboveground part and root system growth of maize, beans and pumpkin seedlings was more intensive in comparison with the control. This process was faster in the dark. The use of tebuconazole and chlormequat chloride significantly inhibited the process of germination both in the light and in the dark. At the same time, the coefficient of the use of reserve substances was maximal under the action of gibberellin, and minimum under the action of retardants, both under the conditions of skotoand photomorphogenesis. The change in the growth characteristics and the coefficient of the use of reserve substances of bean seeds was accompanied by a decrease in the content of total nitrogen, indicating the use of reserve nitrogen-containing compounds in the processes of morphogenesis. The content of protein nitrogen in the control was lower under the conditions of skotomorphogenesis than in the photomorphic seedlings, and the opposite effect was noted for the actions of gibberellin and the retardant. In the later stages of germination, the largest reserve oil of pumpkin seed remained in cotyledonary leaves of photomorphic plants under the effects of chlormequat chloride, which clearly correlated with the least intense growth rates of seedlings in this variant, both in the light and darkness. Under conditions of skotomorphogenesis the growth stimulating effect of gibberellin significantly increased, and the light blocked the action of this phytohormone.","PeriodicalId":8858,"journal":{"name":"Biologija","volume":"33 1-2 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2020-01-04","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"78191941","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

One of the prospective approaches to remedy the current state and halt the loss is the international project “Barcode of Life”. The report describes the results of molecular genetic research on the insect representatives of such orders as Coleoptera (beetles), Lepidoptera (butterflies), and Trichoptera (caddisflies) using the cytochrome c oxidase subunit 1 of the mitochondrial region (COI). We proposed a technique for the sampling of biological material to isolate DNA from the hind legs of individuals. The technique prevents removal of the individuals of the most valuable species collected on the territory of the Republic of Belarus from their habitats. The DNA sample collection of order representatives was investigated. For the subfamily Cetoniinae (flower chafer), significant differences in haplotypes among the representatives of the Belarusian and European parts of their areal were found. For Trichoptera, it was shown that the COI gene has high variability to differentiate species. It was also revealed that some species of caddisflies, which Belarusian researchers believe to be synonymous ones, have sequences with big differences according to the BoldSystem database. This fact should be explored in the future. So, our analysis allows considering the COI gene as a satisfactory marker for the species identification in the Belarusian insects’ taxa studied.","PeriodicalId":8858,"journal":{"name":"Biologija","volume":"58 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2020-01-04","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"84400048","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The investigation of chromosome regions and genes involved in chromosomal aberrations leading to malignization in these lines was the aim of our work. Cytogenetic analysis was performed at different stages of cultivation in vitro. To determine the oncogenes and tumour suppressor genes located on aberrant chromosomes, data on mapping rhesus macaque genes, and high similarity of human and monkey karyotypes were used. We found that, in the line obtained from lymphomatous baboon tissue, the inactivation of tumour suppressor gene RB1 on chromosome 17 after chromosomal rearrangement is one of the most probable causes of in vivo malignization. Chromosomal aberrations at the region of oncogene c-Ki-ras and tumour suppressor gene TP53 change the proliferative status and differentiation in established cell lines obtained from healthy but EBV-seropositive primates. The other cause of malignization in these lines is an increase in expression of the oncogene c-myc caused by trisomy of chromosome 8 where c-myc is located. Structural aberrations in established primate cell lines affecting several chromosomal loci were identified as: (1) causing the proto-oncogene activation – the central event in the tumour clone occurrence, and (2) deactivating tumour suppressor genes. The change in the chromosome number leads to increase in oncogenic products and to damage of regulatory functions associated with cell proliferation.","PeriodicalId":8858,"journal":{"name":"Biologija","volume":"14 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2020-01-04","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"88924551","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

It has been established the growth processes of P. ostreatus, can be best stimulated by applying laser irradiation of mycelium with a green spectrum of light for 10 s. According to this mode of exposure, the best increase in the growth rate of mycelium of 38.3% and the appearance of the largest number of rudiments of the fruiting bodies were recorded. Laser irradiation of mycelium for 10 s with red and blue light spectrum increased the growth rate of mycelium from 7.41 to 20.4%, respectively, and the number of rudiments of the fruiting bodies increased by 1.5 to 2 times. Laser irradiation of the mycelium with 5 s, 15 s, and 20 s with red, blue, and green light spectra did not have a significant effect on the growth processes of P. ostreatus.
