In this study, effects of EGCG on the expression of IL-1beta gene and the IL-1beta level at wound sites during cutaneous wound healing were investigated with histological stain, ELISA and RT-PCR technique. In order to exploit effect of bFGF on MEF, we collected a series of MEF conditioned medium (bFGF-MCM) by co-culturing MEF with increasing bFGF concentrations: 0.03, 0.1, 0.3, 1 and 4 ng/ml. The primitivity of hESC cultured in bFGF-MCM was estimated by morphology and alkaline phosphatase staining. Compared with the control medium (medium conditioned without bFGF: MCM), percentage of undifferentiated colony was increased from 23% to 29%, 44%, 74%, 77% and 78%, respectively. However, percentage of undifferentiated colony in the blank medium (medium conditioned with bFGF but without MEF: bFGF-SR) was from 13% to 31%. This indicated that low concentration of bFGF acted on MEF and stimulated MEF producing effective conditioned medium for maintaining hESC. By monitoring the change of plastids and the expression of FtsZ protein, which functions in the progress of chloroplast division, we show that chloroplasts turn into amyloid and hypostasis when explants dedifferent and at the same time the expression of FtsZ proteins decreases. When explants redifferent and bud, hypostasis turn back into the chloroplast and the expression of FtsZ protein restores. Oral vaccination of animals and man, to provide effective mucosal and/or systemic immunity, is a good delivery system, but it remains poor bioavailablity, one of the most promising strategies to improve the oral delivery relies on polymeric nanoparticles carriers to encapsulate vaccine. Nanoemulsion was used as the carriers of oral immunization of Lugurus Lugurus zona pellucida 3, the immune response and antifertility potential of this vaccine is valuated. Nanoemulsion-encapsulated plasmid pcDNA3.0-Aat-COMP-lzp3-C3d3 (pACLC DNA vaccine) was prepared and evaluated the encapsulation efficiency by anion exchange chromatography. The mice were orally delivered with nanoemulsion-encapsulated plasmid, to detect specific IgG in serum and IgA in vaginal fluid by indirect ELISA, and to perform fertility experiment, to do histological analysis of immunized mice ovary. The results showed that the specific IgG and IgA antibody is elicited, the fertility rate is deceased at a certain extent. In this article, Random Amplification Polymorphic DNA (RAPD) technology was used to increase the Humulus scandens female and male genome teams. The results show that two of the 100 primers, S1519 and S2142, respectly produced a sex-linked band only found in tested males, with the length of 1207 bp and 762 bp. These fragments were cloned and sequenced and then aligned with the GenBank database by BLAST, finding that they are abundant in AT (individually 64%, 54.7%), and no sequence was significantly similar. {"title":"[The expression of MKP-1 and p-ERK(1/2) in primary ovarian epithelial tumor tissues].","authors":"Jian Wei Zhou, Ning Yue Gan, Wei Jiang Zhang","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>To investigate the expression of mitogen activated protein kinase phosphatase-1 (MKP-1) and phosphorylation extracellular signal-regulated kinases (p-ERK(1/2)) in primary ovarian epithelial tumor tissues, and provide experiment's foundation on the new treatment in ovarian cancer. Expression of MKP-1 and p-ERK(1/2) in tissues from 64 patients with primary ovarian epithelial tumor, 35 patients with ovarian epithelial bordline tumor, 32 patients with ovarian epithelial benign tumor and 26 normal ovarian tissues was detected by immunohistochemistry. Western-blot was also used for detecting the expression of MKP-1 and p-ERK(1/2) protein in these tissues. Immunohistochemistry and Western-blot assay showed that the expression of MKP-1 was gradually decreased in normal ovarian tissues, benign tumor, bordline tumor and carcinoma respectively, and there were significant differences among them (P < 0.01). The MKP-1 expression level in the carcinoma tissues of stage III/IV patients was significantly lower than that of stage I/II patients. However, the expression of p-ERK(1/2) was gradually increased in normal ovarian tissues, benign tumor, bordline tumor and carcinoma respectively, and there were also significant differences among them (P < 0.01), the p-ERK(1/2) expression level in the carcinoma tissues of stage III/IV patients was significantly higher than that of stage I/II patients. Expression of MKP-1 and p-ERK(1/2) in same ovarian carcinoma tissues detected by immunohistochemistry and Western-blot assay showed significant negative correlation (r = -0.90, P < 0.01 and r = -0.78, P < 0.01 respectively). The expression changes of MKP-1 and ERKs may play a role in the development of ovarian carcinoma. With the variance of calcium ion concentration and pH, the results obtained by using polarizing microscope showed that there was no significant difference on calcium ion containing of epithelim cells between nucleus rotundus present and absent. Meanwhile, the reproduction of epithlim cell in mantle was occurred after incubation and it was further transferred to form pearl sac. The complete pearl sac is formed when connective tissue cells around epithlim cell of pearl sac. An obvious birefringence was observed by using polarizing microscope and its strength was positively affected by the incubation duration and calcium ion concentration of culture medium. The results demonstrated that with the development of cell and tissue of mantle, the absorption of calcium carbonate from culture medium was occurred and there existed the significant correlation between surrounding calcium ion concentration and absorption or secretion of cultures. The largest quantity of calcium sphere was occurred when the calcium ion concentration was 200 ml/L, which was very significantly higher than that of others (p < 0.01). It was also found that the calcium ion absorption was affected by the culture tissue and the status of culture. Higher calcium ion requirement was in primary culture than that in subculture. The calcium ion absorption and secretion was higher in tissue gobbet than that in single cell. A high significant correlation was observed between calcium ion absorption (Y) and pH (X): Y = 52.34 - 5601.23 X (r = 0.9661, p < 0.05). Pepper was characterized by a strong asynchrony of pollen development within a single anther. With the change of culture period, the proportion of dead pollen increased drastically from 2 day after culture. Microspores that were cultured at the late-uninucleate stage followed one of two developmental pathways. In the more common route, the first sporophytic division was asymmetric and produced what appeared to be typical bicellular pollen. Embryogenic pollen was formed by repeated divisions of the vegetative nuclei. An exine with its specific pattern had already been formed, when microspores were released from tetrads. During subsequent pollen development, microspores increased in size and continued to strengthen the exine. After 24 h in culture, the microspores had increased in size. Thereafter, embryogenesis was indicated in some microspores by two different morphological changes. One featured an expansion in volume of the cell cluster around the germination aperture, the other showed cell cluster volume expansion over the entire microspore surface. Morphogenesis of microspore-derived embryos has been analyzed, at both light and electron microscopical levels. The changes in cell organization after embryogenesis induction, and the characterization of the time sequence of a set of structural events, had been also explained. These changes mainly affected the plastids, the vacuolar compartment, the cell wall and the nucleus. Its sex determination type and sex chromosome has not been recognized. In the present study, we report a novel sex-specific DNA marker in loach Misgurnus anguillicaudatus. The animals used in our experiments were collected from a wild population in wetland on the ancient Yellow river, Yanjin Country, Henan Province. 220 random primers were used for random amplified polymorphic DNA-polymerase chain reaction (RAPD-PCR) with the genomic DNA from two sex individuals as template. When using the S2111 primer, a novel 1040 bp sex-specific PCR product was amplified from known male fish. This fragment of DNA was cloned and sequenced. Two primers (MAmf1-P1 and -P2) were designed from the cloned sex-specific sequence to amplify the male-specific fragment using PCR for sexing, with the house-keeping gene beta-actin as positive control. Sex-specific bands in the gel were represented in the males but none were found in the females. The PCR products in the gel were then transferred onto nylon membranes and hybridized with a DIG-labeled probe of the cloned male-specific DNA fragment. Clear hybridization signals were found only in the male loach. The same result was obtained from dot blotting hybridization. The fragment is a repetitive DNA, and presents its closely related species Paramisgurnus dabryanus but not linked to sex.
